Optimization of vitamin D₂ production in enoki mushroom (Flammulina velutipes) using response surface methodology (RSM), and the effect of adding Enoki mushroom on the general quality characteristics of Doenjang during the fermentation process were studied. For the conversion of ergosterol in enoki mushroom powder to vitamin D₂ via ultraviolet-B (UV-B), the three independent variables of exposure time (40∼120 min), ambient temperature (10∼30℃) and irradiation intensity (0.9 ∼2.1 W/m²) were investigated. The ridge analysis predicted that a maximum yield of 393.73 μg/g was obtained when ambient temperature, exposure time, and irradiation intensity were 23.54℃, 82.33 min and 1.11 W/m2, respectively. During the fermentation process, in comparison with the control sample, bacterial and yeast counts, pH value, protease activity, amino-type nitrogen content, vitamin D₂ content and 5’-nucleotides contents increased in Doenjang after adding enoki mushroom. Beyond this, the addition of enoki mushroom reduced total acidity and had no effect on reducing sugar content, total amylase or α -amylase activity in Doenjang.
젖산 처리에 따른 천마의 불쾌취 감소 및 Gastrodin 함량 변화

전통적인 포제 방법을 이용하여 천마의 불쾌취 및 쓴맛, 비린맛 등을 제거하고, 천마의 주된 유효성분인 gastrodin과 p-hydroxybenzyl alcohol을 증가시키기 위한 연구를 시도하였다. 포제법의 일환으로 생천마와 건천마를 쌀 뜨물과 젖산발효액에 침지하였으며 유의적인 결과를 얻었다. 이러한 결과가 침지액 중의 유기산 등에 의한 효과에 기 인한 것으로 판단되어 대표적 유기산 10종을 선별하여 0.5%로 제조 한 후, 24시간동안 침지시켰다. 그 결과 젖산에 침지된 천마에서 가장 좋은 관능평가를 얻을 수 있었으며, 농도별로 젖산액 처리하였을 경우 1.0%에서 가장 좋은 관 능평가를 얻었다. 또한 1.0% 젖산에 침지한 후 4시간 증자한 후 천마의 유효성분인 gastrodin, p-hydroxybenzyl alcohol 의 함량이 약 20%가 증가되었으며, 기타 유효성분의 대부분 유의적 변화를 나타내지 않았다. 또한 젖산 침지에 따른 천마의 휘발성 향기성분을 측정한 결과 천마 불쾌취의 주성분이라고 판단되는 4-methylphenol과 butanal 등 총 11종을 분리하였다. 이러한 결과로 부터 천마의 관능 및 유효성분의 향상을 위한 전처리 방법으로 1.0% 젖산액에 생천마를 24시간 침지한 후 98℃에서 4시간 증자 후 건조하는 방법이 가장 우수한 것으로 나타났다. This study was carried out to decrease intrinsic objectionable odor, bitter and fishy taste and change the biobeneficial components (gastrodin and p-hydroxybenzyl alcohol) of Gastrodia elate Blume. First of all, Gastrodia elata Blume. was dipped in the rice-washed water and rice-bran solution fermented with Lactobacillus brevis and Lactobacillus plantarum. The objectionable odor, bitter and fishy taste were decreased considerably with the treatments. Gastrodia elata Blume. was also dipped in organic acids (10 different varieties). The result showed that the most effective treatment to reduce objectionable odor, bitter and fishy taste of Gastrodia elata Blume. was treated with 1.0% lactic acid. Eleven different kinds of volatile components such as acetaldehyde, formic acid ethyl ester, acetic acid ethyl ester, 4-methylphenol and 3-hydroxy-2-butanone were separated from Gastrodia elata Blume when treated with or without 1.0% lactic acid. 4-Methylphenol, which is an objectionable odor in Gastrodia elata Blume, decreased with the treatment of lactic acid. While the contents of gastrodin and p-hydroxybenzyl alcohol in Gastrodia elata Blume. increased by 20% when dipped in 1.0% lactic acid solution for 24 hours and steamed at 98℃ for 4 hours.
상추첨가 고지방식이가 흰쥐의 혈청 중 효소활성 변화에 미치는 영향

This study was performed to investigate effects of functional food stuff(Lactuca sativa)-added high fat diets on lipid compounds in the serum of rats. Five-week-old male Sprague-Dawley rats (n=10/group) were fed control diet with high fat(HF), combined with 2.0% powder of lettuce leaf (HF+2.0% RLP), 2.0% powder of lettuce stem (HF+2.0% RSP), 2.0% fermented powder of lettuce stem (HF+2.0% FSP), or 4.0% fermented powder of lettuce stem (HF+4.0% FSP), respectively, for 10 weeks. Then the change of body and viscera weights, ASTase, ALTase, ALPase, ACPase, LDHase, amylase, and ChEase with enzyme activities in serum were analyzed. The results are as follow ; The food efficiency ratio(FER) was at the order of HF+4% FSP < HF+2% RLP < HF+2% FSP < control (HF) < HF+2% RSP. ASTase (GOT) and ALTase(GPT) activities in serum were significantly enhanced in lettuce power treatment groups compared with control(HF) group (p<0.05). The ALPase in serum was significantly increased in HF+4% FSP group compared with other experiment groups (p<0.05). The LDHase activities in serum were shown in order of HF+4% FSP < control (HF) < HF+2% RSP < HF+2% RLP < HF+2%FSP, respectively. The serum glucose content was decreased significantly in the lettuce(leaves, stem, and stem fermentation) parts powder added-diets groups than 162.4±11.93 mg/dl of control group(HF) (p<0.05). These results showed that intake of lettuce can significanly reduce harmful factors, as well as enhance beneficial factor in lipid profile. It was suggestible that intake of 2% fermented stem powder of the lettuce is appropriate level for function rise.
결정형 규산분진에 폭로된 섬유모세포의 자가증식 : Evaluation by H₂O₂and PDGF-AA 와 TGFβ

Objectives : The aim of this study is to find out the activity of autoproliferation of rat fibroblast exposed to crystalline silica and the role of mediators secreted from rat fibroblast. Methods : The effect of α-quartz on production of growth factor (platelet-derived growth factor-AA and transforming growth factorβ) from rat fibroblasts were evaluated by ELISA and immunocytochemical analysis. Gene expression of these growth factors in rat fibrobast exposed to crystalline silica was evaluated by RT-PCR. Furthermore, fibroblast proliferation by culture supernatant of rat fibroblast was assayed by the neutral red test. Results : The amounts of H2O2 and growth factors synthesized in rat fibroblasts were significantly increased by the stimulation of crystalline silica (α-quartz) , which showed the dose-dependent manner to the concentration of α-quartz with the maximum response at the dosage of 100 ㎍/㎠. The result of RT-PCR demonstrated that α-quartz induced gene expression of PDGF-AA and TGFβ in rat fibroblast. We also found that supernatant of α-quartz-cocultured rat fibroblast induced a significant proliferation of fibroblast. Conclusion : Crystalline silica directly induce functional change In fibroblast such as increased release of reactive oxygen species and growth factors. The products of these functional change promote fibroblast proliferation via autocrine loop.
