돼지 수정란의 체외생산은 난자의 체외성숙과 체외수정에 관한 기술의 부족으로 아직까지 만족스럽지 못한 수준이다. 특히 돼지 수정란의 체외생산에는 복잡한 세포질의 성숙과정과 높은 다정자침입율 및 전핵형성의 억제등의 문제점이 있다. 본 연구에서는 돼지 난자의 체외성숙 체계를 개선하기 위하여 transforming growth factorβ(TGFβ)의 첨가가 난자 및 난구세포에 미치는 효과에 대하여 검토하였다. 체외성숙용 배지에 TGFβ를 1~10ng/ml의 농도로 첨가하여 미성숙 난자를 배양한 결과 성숙율이 높아졌다. TGFβ의 효과는 난구세포가 제거된 난자의 성숙에도 효과적이었다. TGFβ의 첨가시기에 따른 차이는 없었으나, 난구세포를 제거한 난자의 경우에는 성숙배양 전반기(59%) 또는 후반기(57%) 24시간 동안에만 TGFβ를 첨가하는 것이 48시간 동안 계속하여 첨가(27%)하는 경우 또는 비첨가(38%)에 비하여 유의적으로 높은 성숙율을 나타냈다(p<0.05). 이와 같은 결과는 난구세포가 돼지 난자의 체외성숙에 필수적이지만 TGFβ는 난구세포가 제거된 난자의 체외성숙에 어느정도 유익한 효과를 발휘하는 것으로 추측된다.

The purpose of this study was to conduct diagnosis of bovine paratuberculosis in Kangwon area. Blood samples were collected from 2,261 dairy cows of 162 herds, and the ELISA and immunoblotting using recombinant 34KDa protein of M. paratuberculosis were conducted. The feces collected from dairy cows were cultured on HEY medium with mycobactin-J and PCR was conducted with washing solution of medium 4 weeks after culture. The ELISA had sensitivity of 83.3% and specificity of 96.7%. And the immunoblotting had sensitivity of 83.3% and specificity of 100%. Of the 2,261 dairy cows, 371 cows(16.4%) were positive in ELISA and 75 cows(3.3%) were positive in immunoblotting. And of the 162 herds, 109 herds(67.3%) had an apparent paratuberculosis prevalence by ELISA and 40 herds(24.7%) by immunoblotting. The geographic distribution of herds with paratuberculosis was not uniform. Of the 241 feces samples including 110 feces from ELISA positive cow, 9 feces were positive in culture and PCR. PCR was able to detect the growth of M. paratuberculosis as early as 4 weeks of culture.

This study was performed to investigate the rabies surveillance in Gangwon-do from 1997 to 2001. Of 185 laboratory submissions for rabies diagnosis, 65(35.1%) cases were determined to be infected with the rabies virus. Annual incidence of animal rabies was 10 cases in 1997, 20 cases in 1998, 16 cases in 1999, 3 cases in 2000, and 16 cases in 2001, respectively. The total number of the reported cases were increased 382% compared with that of the previous period from 1993 to 1996(17 cases). The relative contributions of infected animals in this period were as follows: cattles(24 cases), dogs(26 cases), and raccoons(15 cases). Rabies outbreak in raccoon were steadily expanded. Epidemiologic surveillance showed the rabies outbreaks spreaded to the southern area in Gangwon-do. All rabies cases were showed positive results to rabies tests such as clinical signs, histopathological findings, indirect flourescent antibody test, and mouse inoculation test. Further studies such as genetic and phylogenetic analysis of the isolates should be performed in order to figure out accurate rabies outbreak.

This experiment was carried out to detect the residues of sulfonamides in raw milk. Raw milks which does not contain sulfonamides was collected from one of the farm and fortified with 5 sulfonamides(sulfamerazine, sulfamethazine, sulfamonomethoxine, sulfadimethoxine, sulfaqinoxaline). The sulfonamides in the fortified sample were extracted and detected by High Performance Liquid Chromatography. UV/vis detector was used in this experiment. The results obtained were summarized as follows: 1. Chloroform was good as a extracting solution. 2. 15.5% methanol in PDP as a mobile phase solution was best detective condition for SMR, SMT, SMM. But for SDM and SQN the best condition was 23% methanol. 3. The detectable limits of SMR, SMT, SMM were 2ppb. but SDM and SQN were 20ppb because of delayed retention time and relatively low recovery rate. 4. The peaks of SMR, SMT, SMM and SDM were erected at baseline and the apexes were sharp but SQN was round shape.
아카바네 생바이러스 백신 접종에 의한 한우 백혈구 아군의 변화

To identify immune response of leukocytes in peripheral blood of cattle vaccinated with an attenuated live Akabane virus vaccine, leukocytes were reacted with monoclo-nal antibodies which are specific to bovine lymphocyte surface antigens and assayed by the flow cytometry. Serum neutralizing(SN) test was used to measure antibody titers after vaccination. SN antibody was appeared to 7 days post-vaccination(PV) and 2-8 antibody titers were observed in 14 days PV. Proportion of CD8-MHCclassⅡ+ expressing cells were rapidly increased at 3 days PV. CD8+MHCclassⅡ- cells were increased at 7 days PV. CD4+CD8-, WC+CD4-, CD4+CD8+, WC1-CD4+, WC1-CD8+, and CD4-CD8+ cells were highly increased at 3, 3, 7, 7, 14, 14 days PV, respectively.

This study was conducted to investigate the current status of rabies in wild animals and to elucidate the wild animal transmitting rabies to domestic animals in the Republic of Korea. Through epidemiological survey of rabies near the Demilitarized Zone(DMZ) in Kyonggi and Kangwon Provinces, we found that raccoon dogs( Nyctereutes procyonoides) transmitted rabies to domestic animals in the infected area.To do this we tested the sera or/and brain tissue collected from 8 kinds of wild animals( Nyctereutes procyonoides, Meles meles, Lepus coreanus, Apodemus agrarius, Eothenomys regulus, Felis catus, Mutela sibirica coreana, Martes flavigula aterrima). Only raccoon dogs showed positive reaction in fluorescent antibody test. Naturally acquired antibodies against rabies virus could not be detected in other animals as well as raccoon dogs captured in the rabies infected and non-infected area in Korea. Antibodies against rabies virus could not be detected even raccoon dogs diagnosed with rabies. Therefore, it is considered that raccoon dogs would die before forming antibodies against rabies virus, if infected.
