OMC-2010 구성약재가 마우스의 비장세포 cytokine 생성에 미치는 영향

Objective: We recently reported that OMC-2010 has an immuno-modulatory effects via inhibiting tumor necrosis factor (TNF)-α and interleukin (IL)-5. However, we did not find out which constituents play an important role in immuno-modulatory effect of OMC-2010, Thus, this study was performed to estimate the effects of constituents of OMC-2010 on cytokine production in mouse spleen cells, then ultimately reach to find out effective constituents regulating splenic cytokine production. Methods: Mouse spleen cells were pre-treated with water and ethanol extract of constituents of OMC-2010 such as Rehmannia glutinosa (RG), Pinellia ternata (PT), Citrus unshiu Markovich (CUM), Glycyrrhiza uralensis (GU), Platycodon grandiflorum (PG), Schisandra chinensis (SC). After 1 h, the cells were stimulated with lipopolysaccharide (LPS, 1 ㎍/ml) for 48 h. Then the cells were harvested for real-time reverse transcription polymerase chain reaction to detect cytokine productions. Results: The water extract of RG extract significantly inhibited the LPS-induced inTNF-α and IL-5 mRNA expressions, but the water extract of PT, CUM, GU, PG, and SC did not. The ethanol extract of RG, PT, and SC significantly inhibited the LPS-induced TNF-α, and IL-5 mRNA expressions, but the ethanol extract of CUM, GU, and PG did not. Conclusions: Theses results could suggest that the water extract of RG and the ethanol extract of RG, PT, and SC inhibited the expression of TNF-α and IL-5, which means that the possible candidate of OMC-2010 water extract's action might be RG, and ethanol extract's action might be RG, PR, and SC.

Nypa fruticans Wurmb. (NF) has been used as a folk remedy to treat inflammatory diseases in Asia. In this study, we aimed to investigate the protective effect of NF water extract on cerulein-induced acute pancreatitis (AP). To measure the protective effects of NF on AP, AP was induced via intraperitoneal injection of cerulein (50 μg/kg) hourly for 6 h in mice. NF water extract (100, 250, or 500 mg/kg) or saline was administrated to intraperitoneal injection 1 h before the first injection of cerulein. The mice were sacrificed at 6 h after the final injection of the cerulein. Pancreas, and blood sample were taken for further analysis. Administration of NF water extract inhibited the pancreatic injury, the elevation of pancreatic weight/ body weight ratio, and the elevation of serum digestive enzymes such as amylase and lipase during cerulein-induced AP in mice. Also, pancreas MPO activity, which represents neutrophil infiltration, was inhibited by administration of NF water extract. Taken together, administration of NF water extract reduces the severity of cerulein-induced AP, which suggests a clinical basis that NF could be a potential agent to prevent AP.
TNF-α로 유도된 HaCaT 각질형성세포의 염증반응에서 해죽순의 항염증 효과

Objectives : Nypa fruticans Wurmb. (NF) have been used as a traditional medicine to treat inflammatory diseases in East-South Asia. However, it is largely undiscovered whether NF water extract could exhibit anti-inflammatory activities against tumor necrosis factor-α (TNF-α)-induced inflammatory responses on human keratinocytes, HaCaT cells. Therefore, this study was aimed to investigate the anti-inflammatory activity of NF water extract on TNF-α-induced inflammatory responses in HaCaT cells. Methods : To investigate the anti-inflammatory activites of NF water extract in HaCaT cells, the inflammatory model of HaCaT cells was established under a suitable concentration (10 ng/ml) of human TNF-α (hTNF-α). HaCaT keratinocyte cells were pre-treated with NF water extract for 1 h, and then stimulated with hTNF-α. Then, the cells were harvested to measure the inflammatory mediators such as inducible nitric oxide synthase (iNOS), cyclooxygenase (COX)-2 and prostaglandin E<sub>2</sub> (PGE<sub>2</sub>), and pro-inflammatory cytokine including TNF-α and interleukin (IL)-6. In addition, we examined the inhibitory mechanisms of NF, mitogen activated protein kinases (MAPKs) and inhibitory kappa B alpha (Iκ-Bα) Results : The treatment of NF inhibited the hTNF-α-induced elevation of iNOS, COX-2, and PGE<sub>2</sub> in HaCaT cells. In addition, NF treatment inhibited the hTNF-α-induced elevation of TNF-α and IL-6. Furthermore, NF treatment inhibited the activation of MAPKs but not degradation of Iκ-Bα. Conclusions : Taken together, our result suggest that treatment of NF could inhibit the hTNF-α-induced inflammatory responses via deactivation of MAPKs in HaCaT cells. This study could suggest that NF could be a beneficial agent to prevent skin damage or inflammation.

Objective : Nypa fruticans Wurmb. Fruit (NF) has been used as a conventional medicine to treat inflammatory peridontal diseases in Myanmar and Eastern Asia. However, the anti-inflammatory effect of NF aqueous extract on lipopolysaccharide (LPS)-induced inflammatory responses was not well-investigated. Therefore, this study was aimed to investigate the anti-inflammatory effect of NF on LPS-induced inflammatory responses on RAW 264.7 cells. Methods : To induce inflammation on the macrophage cell line, RAW 264.7 cells were treated with 500 ng/mL of LPS. Water extracts of NF was treated 1 h prior to treatment of LPS. Cell viability was measured by MTT assay. Production of nitrite was measured with Griess assay and pro-inflammatory cytokines such as interleukine (IL)-1 β and IL-6, and tumor necrosis factor (TNF)-α was measured by enzyme-linked immunosorbent assay (ELISA) and real-time polymerase chain reaction (PCR). In addition, we examined the inhibitory mechanisms of NF by western blot and immunocytochemistry. Result : Water Extract from NF itself did not have any cytotoxic effect at the concentration of 200 ㎍/ml in RAW 264.7 cells. Treatment of NF inhibited the production of nitrite, and pro-inflammatory cytokines inlcuding IL-1β, IL-6 and TNF-α in a dose dependant. In addition, NF treatment inhibited the LPS-induced activation and translocation of nuclear factor (NF)-κB. Conclusion : In summary, our result suggest that treatment of NF could reduce the LPS-induced inflammatory responses via deactivation of NF-κB. This study could suggest that NF could be a beneficial drug or agent to prevent inflammation.

Objective : This study was performed to estimate the effects of OMC-2010 extract on cytokine production in mouse spleen cells. OMC-2010 water extract significantly inhibited the LPS-induced TNF-alpha, and IL-5 mRNA expressions, but not shown such changes in IL-1beta, IL-6, IL-4, IL-13. Conclusions : Theses results could suggest that both ethanol and water OMC-2010 extract could inhibit the TNF-alpha and IL-5 mRNA expression.

Objectives : Dioscorea batas Decaisne (DB) has been known to be good for the digestive system on Eastern Asia. However, the protective effect of DB on acute pancreatitis (AP) has not been studied. In this study, we tried to investigate the protective effect of DB water extract on caerulein-induced AP. Methods : To measure the protective effect of DB on AP, Mice were injected with cholecystokinin analogue caerulein (50 μg/kg) hourly for 6 times. DB water extract (200 or 400 mg/kg) or saline (control group) was administered orally 1 h before the first injection of caerulein. The mice were sacrificed at 6 h after the last injection of caerulein. The pancreas tissues and serum samples were immediately taken for further analysis. Results : Administration of DB water extract showed the inhibitory effect on the increase of pancreas weight/body weight ratio, pancreatic histological damage. And the rise of serum lipase level was significantly reduced in DB water extract treatment group during AP in mice. However administration of DB water extract did not show significant reduction in serum amylase level. Also, mRNA levels of pro-inflammatory cytokines Interleukin (IL)-6 and Tumor necrosis factor (TNF)-α but not IL-1β were inhibited by administration of DB water extract. Conclusions : Taken together, we found that administration of DB water extract ameliorates the severity of caeruleininduced AP, which suggests the potential to be an effective treatment on AP.
