실험적으로 항원에 의하여 일차 자극된 CD4⁺ T 세포의 이차 면역 반응의 분석

Memory T lymphocytes of the immune system provide long-term protection in response to bacterial or viral infections/immunization. Ag concentration has also been postulated to be important in determining whether T cell differentiation favors effector versus memory cell development. In the present study we hypothesized that naive Ag-specific CD4⁺ T cells briefly stimulated with different Ag doses at the primary exposure could affect establishment of memory cell pool after secondary immunization. Methods: To assess this hypothesis, the response kinetics of DO11.10 TCR CD4⁺ T cells primed with different Ag doses in vitro was measured after adoptive transfer to naive BALB/c mice. Results: Maximum expansion was shown in cells primarily stimulated with high doses of ovalbumin peptide (OVA_323-339), whereas cells in vitro stimulated with low dose were expanded slightly after in vivo secondary exposure. However, the cells primed with low OVA_323-339 peptide dose showed least contraction and established higher number of memory cells than other treated groups. When the cell division was analyzed after adoptive transfer, the high dose Ag-stimulated donor cells have undergone seven rounds of cell division at 3 days post-adoptive transfer. However, there was very few division in naive and low dose of peptide-treated group. Conclusion: These results suggest that primary stimulation with a low dose of Ag leads to better memory CD4⁺ T cell generation after secondary immunization. Therefore, these facts imply that optimally primed CD4⁺ T cells is necessary to support effective memory pool following administration of booster dose in prime-boost vaccination.

Paraffin을 이용한 Thyroid Phantom제작에 따른 유용성에 관한 연구

  방사성의약품을 이용한 핵의학적 영상기록에 있어 장기가 함유하고 있는 정보량을 최대한 묘출시켜 양질의 진단정보를 제공해야 한다는 것은 매우 중요하다. 그것을 위한 기술적인 문제를 비롯하여 장비성능의 유지관리에 많은 노력을 기울이고 있으며, 다양한 종류로 상업화되어 이용되고 있는 우수한 phantom들이 사용되고 있다.<BR>  본 연구에서 인체의 연부조직과 매우 흡사한 물리적 조건을 가지고 있으며 구입하기 쉬운 Paraffin을 이용하여 기존에 사용되고 있는 acryl thyroid phantom과 동일하게 paraffin phantom을 제작하였으며, 판매되고 있는 acryl thyroid phantom에서 측정할 수 있는 것 보다 더 높은 분해능을 측정할 수 있도록 작은 3 mm와 6 mm 직경의 cold area를 삽입한 paraffin thyroid phantom을 수작업으로 제작하였다. 방사성의약품 <SUP>99m</SUP>TcO4를 이용하고 pinhole collimator를 이용하여 상업화되어 사용되고 있는 acryl thyroid phantom과 본 연구를 위하여 제작한 paraffin thyroid phantom의 특성을 비교 분석한 결과 paraffin을 이용한 phantom도 상업화되어 사용 중인 acryl thyroid phantom과 동일한 물리적 특성을 유지하고, 오히려 분해능이 높은 부분을 측정할 수 있고 연부조직의 특성을 연구할 수 있는 등가물질로서 구입하기 쉽고 제작이 수월한 장점을 비롯하여 경제성이 있음을 증명하는 계기가 되었다.   Phantoms are very necessary for quality assurance of radio nuclides imaging systems to maintain standards and to ensure reproducibility of test. General quality assurance and instrument quality control are essential in every hospital. The human tissue equivalent materials are aluminum, arcryl, water and epoxy .. etc. It is very important to select optimum equivalant materials for a phantoms in QC ..<BR>  Especially, paraffin is very similar with human soft tissue in X or Gamma-ray physical characteristics and easy to buy wiith economically.<BR>  We made a paraffin thyroid phantom and compare with thyroid arcryl phantom, also used commercially in practice. Two small size cold spots (3 and 6 mm diameter) and a hot spot (3 mm diameter) embeded in paraffin phantom. And imaged with <SUP>99m</SUP>TcO₄ by camera for analysis about spatial resolution and noise at the hot and cold spots.<BR>  We got some results as below:<BR>  1. No difference in counting rate and noise between both arcryl and paraffin thyroid phantoms.<BR>  2. The best spatial resolution can be seen 6 cm distance between pinhole collimator and thyroid phantoms(arcryl and paraffin).<BR>  3. More optimal spatial resolution could acquired in paraffin thyroid phantom.<BR>  Paraffin is very similar with human soft tissue in atomic number, density and relative absorbtion fuction, and can be shaped easily what we wanted.<BR>  So we can recommendation paraffin as quality assurance phantom because its usefulness, economical benefit and purchasability.

영재아의 실행기능 향상을 위한 게임놀이프로그램 효과

This study was designed to examine the effect of game play program developed for enhancing the executive function in gifted children The subjects of this study were 24 children, who first grade middle school students. The subjects were assigned into either experimental group(12 children) or Control group(12 children) using matching subject method on the basis of selection evaluation. The experimental groups had attended of 10 group sessions two times a week for 5 weeks the including one week pretest. The post test was conducted within a week after the program. The pre-post test scales used in this study were Stroop Test, Fluency Test, Children's Color Trails Test, Digit SpanTest, and Maze Test of measure executive function. The results of this study were as follow: The Game Play Program had positive effects on executive function domains .In attentional Control, a significant improvement has been observed in selective attention, self control and sustained attention through game play therapy program for gifted children. The results showed positive effect on complex-letter and complex-colo of Stroop Test for measuring inhibition control executive function. In information .In information processing, a significant improvement has been observed in word fluency through Game Play Program. In cognitive flexibility, a significant improvement has been observed in CCTT-difference interrupt through Game Play Program. 본 연구의 목적은 영재아의 실행기능 향상을 위한 게임놀이프로그램을 개발하고 적용하여 그 효과를 검증하는 것이다. 이를 위해 대전시 S 교육청 지역공동 영재아 중학교 1학년 24명 중 12명을 실험집단으로 게임놀이프로그램을 실시하고, 프로그램에 참여하지 않은 12명의 통제집단의 실행기능 검사의 사전, 사후변화를 비교하였다. 그 결과 게임놀이프로그램을 실시한 실험집단이 실시하지 않는 통제집단 보다 주의 통제영역을 재는 Stroop 단어, 색상점수에서 사전보다 사후에 유의하게 높았다. 또한 정보처리영역의 유창성 범주에서도 실험집단이 통제집단보다 사전에 비해 사후에 유의하게 향상되었으며, 인지적 융통성 영역인 주의전환능력을 재는 CCTT 검사에서 실험집단이 통제집단 비해 사전보다 사후에 유의하게 향상되었다. 이러한 결과를 볼 때, 게임놀이프로그램이 영재아의 실행기능을 향상시키는데 효과적이라는 것을 알 수 있다.

오이 및 토마토 생육에 미치는 토양개량제 효과

염류집적 토양에서 광합성균, Soil plus, 목초액, 활성탄 등 토양개량제가 오이와 토마토의 생육과 수량에 미치는 영향을 시험하였다. 그 결과 활성탄이 수량을 증진 시키고 뿌리의 활력도 좋았다. 토마토 배꼽썩음병도 활성탄 처리구가 가장 낮았다. 토마토의 당도는 Soil plus구가 높았으며 질산염과 인산함량은 처리간에 차이가 없었다. To investigate effect of soil conditioners on salt reducing effect, saline soil were treated with photomicrobe, soil plus, pyroligneous liquor and active charcoal and cucumber and tomato were grown. Yield and root development were enhanced in active charcoal treatment overall and tomato blossom end rot frequency was lowest in active charcoal treatment. Total sugar content was highest in soil plus treatment and nitrate and phosphate content were not significantly different within 4 treatments.

PET/CT 장치의 감마선 검출과정

  SPECT camera와 PET scanner 그리고 PET/CT 장치는 감마선을 검출하여 얻어진 자료(data)를 정보화 한다. 특히 PET/CT 장치는 PET영상에서 나타나는 해부학적 불선예함을 CT로 보완하여 선예한 해부학적 구조의 영상 묘출능력이 뛰어나 양질의 진단정보를 제공하고 있게 되었다. 신속 정확한 자료의 획득과 정보화를 위하여 우선적으로 검출기(detector)에서의 효율적인 감마선검출은 매우 예민하고 중요한 역할 중의 하나이다. 섬광체(scintillator)의 개발과 함께 data 처리를 위한 software의 발전은 영상재구성처리시간의 단축은 물론 영상의 질적인 향상을 초래하였다. PET scanner에서는 SPECT camera의 100~200keV와는 달리 511keV의 고에너지 감마선을 검출한다. 섬광체 GSO, LSO는 PET scanner의 섬광체로 비교적 많이 이용되고 있다. 고에너지 감마선에너지를 빛으로 전환하여 여러 단계를 거쳐 정보화하는 과정은 결과적으로 고화질의 영상을 비롯한 정확하고 신속한 진단정보획득에 목적을 두고 있다. 화질의 질적향상은 획득한 정보량에 대한 영상의 재구성 방법과 기술에 따라 영향을 받는다.<BR>  본 연구는 PET/CT 장치에서 특히 PET부분에서 섬광체(scintillator)의 특성과 감마선의 검출과정에서부터 영상 재구성단계까지 문헌적 고찰을 통하여 요약 설명하였다.   The PET/CT scanner is an evolution in image technology. The two modalities are complementary with CT and PET images. The PET scan images are well known as low resolution anatomic landmak, but such problems may help with interpretation detailed anatomic framework such as that provided by CT scan. PET/CT offers some advantages-improved lesion localization and identification, more accurate tumor staging. etc. Conventional PET employs tranmission scan require around 4min./bed position and 30min. for whole body scan. But PET/CT scanner can reduced by 50% in whole body scan. Especially nowadays PET scanner LSO scintillator-based from BGO without septa and operate in 3-D acquisition mode with multidetectors CT.<BR>  PET/CT scanner fusion problems solved through hardware rather than software. Such device provides with the capability to acquire accurately aligned anatomic and functional images from single scan.<BR>  It is very important to effective detection from gamma ray source in PETdetector. And can be offer high quality diagnostic images. So we have study about detection processing of PET detector and high quality imaging process.
