        햄스터 볼주머니를 이용한 구강점막 자극평가 조건설정

        Objectives: The purpose of this study is to evaluate the optimum conditions for oral mucosal irritation testing using the buccal pouch of hamsters. Methods: Test materials were applied to the buccal pouch of seven-week old male Syrian hamsters (SLC, Japan) four times at one-hour intervals and macroscopic changes were examined at 24 hours after final treatment. After sacrifice, the buccal pouches were removed and prepared for histopathological evaluation. In order to set the exposure time, we performed exposure tests of 5, 12, 18 and 23 minutes using sodium lauryl sulfate (SLS) 1% and set the treatment volume from the test results at 2, 3, or 4 ml treatment using SLS 1%, Triton X-100 1% and ethanol. After setting the experimental conditions, seven groups of materials [sodium lauryl sulfate (SLS) (1%), Triton X-100 (1%), hydrogen peroxide (3%), ethanol (100%), chlorhexidine (0.2%, 2%), phosphate buffer saline (PBS)] were assessed. Results: Experimental conditions of material exposure time were fixed as 18 minutes from the exposure tests of 5, 12, 18 or 23 min using sodium lauryl sulfate (SLS) 1%. Treated volume was set as 4 ml per each pouch from the test results of 2, 3, or 4 ml treatments using SLS 1%, Triton X-100 1% and ethanol. The results in terms of irritation degree were in the order of sodium lauryl sulfate (SLS) (1%) > Triton X-100 (1%) ${\fallingdotseq}$ hydrogen peroxide (3%) > ethanol (100%) ${\fallingdotseq}$ chlorhexidine (0.2%, 2%) > phosphate buffer saline (PBS). Conclusion: From this study, suitable conditions for hamster mucosal irritation testing were suggested and this method was verified through materials commonly used on oral mucosal membranes.

        발아배지 성분에 따른 발아현미 추출물의 생리활성연구

        This study was supposed to determine the bioactive effects of germinated brown rice extract with high contents of γ-aminobutyric acid(GABA). The optimized germination medium containing black garlic, black beans, blueberry fruit, inonotus obliquus mushroom, black ginseng, was confirmed by measuring the GABA content of germinated brown rice according to the change of germination medium condition. To estimate the antioxidant activity, electron-donating ability, ABTS<sup>+</sup> radical scavenging activity and superoxide dismutase(SOD)-like activity were conducted. DOPA oxidase inhibitory efficacy was evaluated for whitening effect, and BCOP assay was used for safety evaluation. Germinated brown rice in normal medium condition increased GABA content by 2.6 times, and other medium conditions with active ingredients increased GABA contents by 4 or more times, compared to the pre-germinated brown rice. Germinated brown rice in all of the medium conditions with active ingredients showed good antioxidation activity, and DOPA oxidation inhibitory effect, especially in medium with blueberry fruit and black ginseng. The safety was also confirmed in BCOP assay. The results of this study expected to be the basic data for the research of germinated brown rice, and provided a practicability as a functional cosmetic material.

        Spectrophotometric Direct Peptide Reactivity Assay (Spectro-DPRA)를 위한 펩타이드의 보관조건에 따른 품질 변성 연구

        The Spectrophotometric direct peptide reactivity assay (Spectro-DPRA) is now in process for the development and validation of evaluation on skin-sensitizing chemicals. The cysteine peptides (Ac-RFAACAACOOH) and lysine peptides (Ac-RFAAKAA-COOH) is used for Spectro-DPRA. The objective of this study is to confirm the stability of cysteine and lysine peptides which are used for Spectro-DPRA through the storage conditions. We analyzed the stability of peptide using HPLC according to the conditions, such as storage temperature and storage period. The peptide powders of cysteine and lysine were stable on storage -80℃, for 6 and 12 month. And 10 mM stock peptides of cysteine and lysine were also stable on storage -70℃, for 1, 3 and 6 month. But 10 mM stock peptides of cysteine were denatured and not stable on storage -20℃, for 1 month. Taken together, peptides powder for Spectro-DPRA should be stored under -80℃, less than 12 month. And, 10 mM stock peptides should be also stored under -70℃, less than 6 month.

        안자극 평가의 대체시험법인 인공각막모델(SkinEthic<sup>TM</sup>, HCE)의 국내 확립 연구

        The objective of this study is to investigate the utility of reconstructed human corneal epithelium model(SkinEthic<sup>TM</sup>, HCE) as an alternative method to eye irritation test. We confirmed the pre-check test method for eye irritation test through condition setting of production for freeze-killed epidermis, possible direct MTT reduction with test substances, and colorant properties in test chemicals. And, we conducted eye irritation test using 8 chemicals listed in the proficiency test of the OECD test guideline 492. Summarizing the results above of assessment, it was confirmed that the SkinEthicTM HCE proves that it is so efficient and trustworthy enough to replace the eye irritation test. Taken together, the results indicated that the SkinEthic<sup>TM</sup> HCE proposed by OECD test guideline 492 is expected to become a well-established method in Korea.

        단백질 분해 효소 처리에 따른 당근추출물의 항산화 활성 및 Collagenase 활성 저해 효과

        The purpose of this study is to investigate the antioxidant activity and anti-wrinkle effects of carrot extracts prepared by enzymatic hydrolysis using in vitro model. To hydrolyze for carrot extracts, we used papain and bromelain as the enzyme. The antioxidant activity was evaluated the total contents of polyphenol and flavonoid. Additionally we measured the ABTS radical scavenging ability and superoxide dismutase(SOD)-like ability with carrot extracts prepared by enzymatic hydrolysis. And the collagenase activity inhibition was also performed to evaluate anti-wrinkle effects. We set the concentration of carrot extracts prepared by enzymatic hydrolysis as 10, 20%. The total contents of polyphenol was 16.41 mg/g(10%) and 19.06 mg/g(20%) in papain treatment group, 16.42 mg/g(10%) and 19.08 mg/g(20%) in the bromelain treatment group. In the results of total flavonoid contents, the treatment group of bromelain showed 12.34 mg/g(10%), 14.94 mg/g(20%) and 9.54 mg/g(10%), 12.17 mg/g(20%) in treated papain group. ABTS radical scavenging ability of carrot extracts prepared by enzymatic hydrolysis in high concentration was almost 100% each test group and SOD-like activities were measured and papain treatment group showed higher activity than bromelain treatment group. At all concentration of carrot extracts prepared by enzymatic hydrolysis showed the high collagenase inhibition activity over 60-80%, it means carrot extracts prepared by enzymatic hydrolysis is effective in anti-wrinkles. These results indicated that carrot extracts prepared by enzymatic hydrolysis might have antioxidant activity and anti-wrinkle effects and using enzyme as papain and bromelain can enhance these activities of carrot extract.

        아사이베리 에탄올추출물의 항염증 효과

        The purpose of this study is to investigate the anti-inflammatory effects of acai berry ethanol extract (ABEE) using in vitro and in vivo. The cell toxicity of ABEE was evaluated with or without LPS using the RAW 264.7 cells. And the anti-inflammatory effects of ABEE were also estimated in RAW 264.7 cells by secretion of nitric oxide (NO) and interleukin (IL)-1α, and in lipopolysaccharide (LPS)- induced acute inflammatory mouse ear edema model. The inhibition rate of LPS-induced NO secretion was increased by ABEE dosedependently up to 40.5% at 2,000 μg/mL, and LPS-induced IL-1α secretion was decreased by ABEE in dose dependent manner. TPA (12-o-tetradecanoyl phorbol-13-acetate)-induced ear thickness and weight were decreased by 10% ABEE treatment by 61.4% and 56.0% each. As in the histopathological examination, TPA-induced epidermis thickness, inflammatory cell infiltration and mast cell increment were improved in 10% ABEE treatment group. This results indicated that ABEE might have anti-inflammatory effects in both in vitro and in vivo assays, and can be developed as an inflammatory ingredients in the future.

        아토피성 피부염 동물모델에서 Black currant seed oil의 비장세포 cytokine 및 조직병리학적 변화에 미치는 영향

        This study investigated the cytokine excretion of spleen cells and histopathological changes in atopic-like dermatitis model, and evaluated the anti-atopic effects of black currant seed oil. BALB/c mice (five-week old) were divided into four groups: N (normal group, nontreated), C (control group, induced atopic dermatitis), PC (positive control group, Tacrolimus ointment treatment for induced atopic dermatitis), and E (experiment group, Black currant seed oil treatment for induced atopic dermatitis, E). During the experiment, no significant changes were observed in feed intake, water, intake, body weight. The INF-γ levels of spleen cells in PC and E showed a significant increase compared to those of C, and. IL-4 levels in PC and E decreased significantly than those of C (p<0.05). In histological observation using H&E staining, the tissue thickness of the epidermis and the degree of inflammatory cell infiltration were improved in PC and E compared to C. The numbers of mast cells by toluidine blue staining were significantly decreased in PC and E than C (p<0.05). The results of this study indicated that black currant seed oil can improve the atopic dermatitis by regulating the Th1/Th2 cytokine ratio and histopathological mechanism.

        이동 경로 데이터에 기반한 이동 객체의 시공간 위치 예측 기법

        사용자에게 적응된 서비스를 제공하기 위하여 환경으로부터 얻어지는 다양한 형태의 데이터를 이용한 다양한 방법이 연구되고 있다. 그 중 과거 이동 경로 자료는 사용자의 현재 이동 위치를 예측하고 이와 관련된 서비스를 제공하는데 유용하게 사용될 수 있다. 본 논문에서는 사용자의 과거 이동 경로의 분석을 통하여 이동중인 사용자의 시공간 위치예측 기술을 제안한다. 환경으로부터 발생한 사용자의 이동 경로를 수집하고, 수집된 데이터에서 이동 경로 선택(Path Selection) 방법을 이용한다. 이동 경로 선택 방법은 이동 중에 발생한 경로의 거리, 시간, 방향의 요소와 동적정합법을 사용하여 유사성(Similarity)을 측정하며 유사성이 가장 높은 경로를 선택한다. 선택된 경로는 시간에 따른 공간 정보 및 위치에 따른 시간 예측 서비스를 위하여 사용가능 하며, 실험을 통하여 유사성이 높은 이동 경로를 선택하는 모습을 확인하였다. User adaptive services have been important features in many applications. To provide such services, various techniques with various kinds of data are being used. In this paper, we propose a method to analyze user's past moving paths and predict the goal position and the path to the goal by observing the user's current moving path. We develop a spatiotemporal similarity measure between paths. We choose a past path which is the most similar to the current path using the similarity. Based on the chosen path, user's spatiotemporal position is estimated. Through experiments we confirm this method is useful and effective.

      • 온톨로지와 게임 커뮤니티의 질의/응답 게시글을 이용한 대화형 NPC의 구현

        최근 컴퓨터 게임에 등장하는 NPC(Non-player Character)에 각종 인공지능 기법을 적용하는 연구들이 이루어지고 있다. 하지만 대부분의 연구가 플레이어를 상대하는 적대적 입장의 NPC들의 움직임 조절에 초점을 맞추고 있고 아직까지 게임 상에서 등장하는 모든 NPC는 항상 같은 말과 비슷한 행동을 되풀이하는 모습만을 보여주고 있다. 이는 플레이어가 게임을 비현실적으로 느끼게 만들고 결과적으로 게임의 재미를 저하시키는 요소로 작용한다. 플레이어에게 보다 현실적인 게임 환경을 제공하기 위해서는 NPC가 단순히 게임의 배경을 구성하는 오브젝트가 아니라 다양한 대화를 통해 플레이어에게 많은 영향을 주게하여 게임의 기여도를 높여주어야 한다. 본 논문에서는 이를 위해 게임 속에서 주어지는 퀘스트를 구성하는 NPC, 몬스터, 보상 등의 속성 정보를 온톨로지화 하고, 인터넷에 존재하는 게임 커뮤니티에서 퀘스트 질의/응답 게시판의 글을 추출하여, 플레이어의 관련 질의에 응답하는 NPC를 구현하고자 한다. 이를 위해 온톨로지 정보를 이용한 검색 알고리즘을 구현하였고, 시뮬레이션을 통해 NPC가 커뮤니티 게시글 정보를 이용하여 유저에게 고정되지 않은 다양한 메시지를 전달하면서 동시에 유저의 게임 진행을 도와주는 모습을 확인하였다.
