Emerging roles of polyunsaturated fatty acid synthesis pathway in colorectal cancer

The development of colorectal cancer typically involves the accumulated influences of geneticalterations, medical issues, lifestyle, and diet. Dietary fatty acids appear to affect thetumorigenesis and progression of colorectal cancer. Despite conflicting results, the currentconsensus on the effects of very long-chain polyunsaturated fatty acids on colorectal cancer isthat low levels of eicosapentaenoic acid and docosahexaenoic acid, and high levels ofarachidonic acid are associated with an increased risk of colorectal cancer. Altered levels ofarachidonic acid in membrane phospholipids can change the levels of prostaglandin E2, whichaffect the biological activities of cancer cells in multiple stages. Arachidonic acid and other verylong-chain polyunsaturated fatty acids can affect tumorigenesis in prostaglandin E2-independent manners as well, including stabilization of β-catenine, ferroptosis, ROS generation,regulation of transcription factors, and de novo lipogenesis. Recent studies have revealed anassociation between the activities of enzymes synthesizing very long-chain polyunsaturatedfatty acids and tumorigenesis and cancer progression, although the mechanisms are stillunknown. In this study, PUFA effects on tumorigenesis, the endogenous very long-chainpolyunsaturated fatty acid synthesis pathway, metabolites of arachidonic acid and their effectson tumorigenesis and progression of CRC, and current knowledge that supports the associationof the enzymes involved in the polyunsaturated fatty acid synthesis pathway with colorectalcancer tumorigenesis and progression are reviewed.
Simple Purification of Adeno-Associated Virus-DJ for Liver-Specific Gene Expression

Recombinant gene expression using adeno-associated viruses (AAVs) has become a valuable tool in animal studies, as they mediatesafe expression of transduced genes for several months. The liver is a major organ of metabolism, and liver-specific expressionof a gene can be an invaluable tool for metabolic studies. AAV-DJ is a recombinant AAV generated by the gene shuffling of various AAV serotypes and shares characteristics of AAV2 and AAV8. AAV-DJ contains a heparin-binding domain in its capsid, which suggests that a heparin column could be used for the purification of the AAV. Given that AAV-DJ has been only recently available, relatively little is known about the optimal preparation/purification and application of AAV-DJ. Here, we present a simplelarge-scale preparation method that can generate 3×1013 viral particles for in vivo experiments and demonstrate liver-specific gene expression via systemic injection in mice.
The SCAP/SREBP Pathway: A Mediator of Hepatic Steatosis

Nonalcoholic fatty liver disease (NAFLD) is strongly associated with insulin resistance, obesity, and dyslipidemia. NAFLD encompassesa wide range of states from the simple accumulation of triglycerides in the hepatocytes to serious states accompanied by inflammationand fibrosis in the liver. De novo lipogenesis has been shown to be a significant factor in the development of hepatic steatosisin insulin-resistant states. Sterol regulatory element binding protein-1c (SREBP-1c) is the main transcription factor that mediatesthe activation of lipogenesis, and SREBP cleavage activating protein (SCAP) is required for the activation of SREBPs. Here, recentanimal studies that suggest SCAP as a therapeutic target for hepatic steatosis and hypertriglyceridemia are discussed.
