인체 간암세포에서 β-lapachone 처리에 의한 Tight Junction 관련 유전자의 변화

${\beta}-lapachone$은 남미지역에서 자생하는 Tabebuia avellanedae라는 나무의 수피에서 동정된 quinone계 물질로서 다양한 인체암세포에서 항암효과가 있는 것으로 알려져있다. 본 연구에서는 ${\beta}-lapachone$의 암 전이 억제에 대한 연구의 일환으로 HepG2 및 Hep3B 인체 간암 세포의 전이관련 유전자의 발현에 미치는 ${\beta}-lapachone$의 영향을 조사하였다. MTT assay 및 세포형태변화 관찰 결과에서 ${\beta}-lapachone$ 처리에 따라 HepG2와 Hep3B 세포들은 ${\beta}-lapachone$ 농도 의존적으로 세포의 증식이 억제되었으며 그 형태적 변형도 동반하였다. ${\beta}-lapachone$처리에 의한 암 전이 지표가 되는 IGF-lR, Tjs (ZO-1, claudin-3,-4) 및 Tj 조절인자(${\beta}-catenin$)의 발현을 RT-PCR과 Western blot analysis를 통하여 확인한 결과 ${\beta}-lapachone$ 처리가 IGF-1R의 발현 억제와 Tj 유전자 발현의 증가를 유도함으로써 ${\beta}-lapachone$이 Tj를 강화하여 암세포의 전이 억제작용을 하는 것으로 관찰 되었다. 이상의 결과는 인체 간암세포에서 ${\beta}-lapachone$의 항전이 작용의 이해에 중요한 기초 자료가 될 것으로 생각한다. A hallmark of cancers is 'leaky' tight junctions (Tjs). TJs mediated paracellular permeability is elevated and TJs maintained cell polarity is frequently lost. Concomitantly, TJs-associated proteins including members of the claudin family of proteins are dysregulated. Recent findings indicate that these TJs changes can contribute to cancer progression. In this study, we examined the effects of ${\beta}-lapachone$, a quinone compound obtained from the bark of the lapacho tree (Tabebuia avellanedae), on the Tjs-associated regulators in human hepatocarcinoma cell lines, HepG2 and Hep3B. ${\beta}-lapachone$ treatment downregulated the levels of insulin-like growth factor 1 receptor (IGF-lR) proteins in both HepG2 and Hep3B cells. But the levels of claudin-3 and -4 proteins were increased in ${\beta}-lapachone$-treated HepG2 and Hep3B cells. And also the zonnula occludens-l (la-I) and p-catenin protein levels by ${\beta}-lapachone$ were increased in a time-dependent manner. However, claudin-3 and -4 mRNA levels were uninhibited by ${\beta}-lapachone$ in HepG2 and Hep3B. The present results suggest that the upregulation of claudin-3 and -4 protein levels by ${\beta}-lapachone$ occurs by a post-transcriptional mechanism and points to a novel mechanism by ${\beta}-lapachone$.

본 논문에서는 문자 검출 및 인식 기술을 활용하여 비정형의 폐 기능 검사지 이미지로부터 연구를 위한 관련 정보들을 추출하여 정형화하는 방법을 제안한다. 또한 문자 인식 오차율을 줄이기 위한 후처리 방법 또한 개발하고자 한다. 제안하는 정형화 방법은 폐 기능 검사지 이미지에 대해 문자 검출 모델을 사용해 검사지 내에 존재하는 모든 문자를 검출하고, 검출된 문자 이미지를 문자 인식 모델에 통과시켜 문자열을 얻어낸다. 얻어낸 문자열에 대해 문자열 매칭을 이용한 유효성 검토를 진행하고 정형화를 마무리한다. 제안하는 정형화 시스템의 오차율은 약 1% 이내, 검사지 당 처리속도는 2초 이내로 전문인력의 수작업을 통한 정형화 방법보다 더 효율적이고 안정적인 방식이라는 것을 확인할 수 있다. In this paper, we propose a method of extracting and recognizing related information for research from images of the unstructured pulmonary function test papers using character detection and recognition techniques. Also, we develop a post-processing method to reduce the character recognition error rate. The proposed structuring method uses a character detection model for the pulmonary function test paper images to detect all characters in the test paper and passes the detected character image through the character recognition model to obtain a string. The obtained string is reviewed for validity using string matching and structuring is completed. We confirm that our proposed structuring system is a more efficient and stable method than the structuring method through manual work of professionals because our system’s error rate is within about 1% and the processing speed per pulmonary function test paper is within 2 seconds.

본 연구의 목적은 건강 검진 무증상 수진자 관상동맥 전산화단층촬영(Coronary artery CT angiography; CCTA) 검사 시 심전도창 획득 범위(ECG Window Acquisition Range)를 적절하게 설정함으로 수진자의 방사선 피폭 선량 감소를 할 수 있는지 알아보고자 연구를 수행하였다. 2022년 5월부터 2023년 1월까지 9개월간 본원에서 건강증진센터를 방문하여 CCTA 검사를 시행한 심장 박동(Heart rate) 70 이하인 무증상 성인 1000명을 대상으로 하였다. 상대적 지연 스캔 방법과 절대적 지연 스캔 방법으로 ECG Window Acquisition Range를 변경하여 설정하여 총 네 가지의 방법으로 방사선량 차이를 체계적으로 비교평가하였다. CCTA 검사에서 주로 얻는 상대적 지연 스캔 방법이 아닌 절대적 지연 스캔 방법에 따라 검사했을 경우, 방사선 피폭 선량이 10% 이상 감소하였고, 같은 상대적 지연 스캔에서도 ECG Window Acquisition Range를 기존보다 반으로 줄였을 때 또한 방사선피폭 선량이 20% 이상 감소하였다. CCTA 검사 시 ECG Window Acquisition Range 구간을 5% 나 60 ms로 구간을 설정하여도 얻고자하는 영상 획득에 변함없이 방사선피폭 선량 감소에 도움을 줄 수 있었다. This study was to reduce the radiation exposure dose of recipients by setting the ECG window acquisition range appropriately during Coronary artery CT Angiography (CCTA) examination. Group 1 (control group) opened ECG window acquisition range to 65% to 75% using the relative delay scan (RDS), which is divided by percentage (%) based on the R-R peak interval of recipient's Average HR, and tested by setting pitch to auto. Group 2 fixed the pitch according to the recipient's HR under the same conditions as Group 1. Group 3 used absolute delay scan (ADS), which is divided by milliseconds (ms) based on the R-R peak interval of recipient's Average HR and fixed the ECG window acquisition range by 60 ms intervals. Group 4 narrowed the scan range from 68% to 73% under the same conditions as Group 1. The values of the mean, minimum, maximum, the volume CT dose index (CTDIvol), and dose length product (DLP) were compared. The exposure dose was reduced by more than 10% when acquired with ADS, not RDS mainly obtained from CCTA. Also, compared to the existing RDS, when the scan range was reduced by half, it was reduced by more than 20%. In conclusion, In CCTA testing, setting the ECG window acquisition range to 5% or 60 ms can reduce radiation exposure without compromising the best phase image acquisition and image quality.

Hazardous chemicals, such as polycyclic aromatic hydrocarbons (PAHs), volatile organic compounds (VOCs) and heavy metals, are known as being harmful to human health were included in oils released by the Herbei Spirit Oil Spill accident in December 2007. To investigate changes of eating habits by the exposure to harmful substances, we conducted 5 focus group interviews for residents at Taean coast areas, who had experienced the oil spill accident. Participants included 46 women (mean age: 57.2 ± 10.9) who were mainly responsible for preparing family meals. Focus group discussions were audio-taped, transcribed and categorized by themes. Participants expressed more frequent illness symptoms such as dizziness, vomiting, visual loss, and skin diseases after the accident. They mentioned that their worries about economic hardship and worsened health status since the accident induced mental problems, such as depression about their children. Regarding eating habits, participants reported less intakes of fishes and meats and relatively more intakes of vegetables and kimchi due to the lack of household incomes after the accident. Although the participants had been used to collecting or catching fish or shellfish for their consumption previously, they mainly purchased these foods from local markets after the accident. Changes of eating habits induced by the accident included drinking boiled water and having steamed or fried seafood rather than raw seafood. Changes of food intakes occurred less frequently in older adults due to their long-term fixed eating habits, although they felt uncomfortable for having raw fish. The findings of this study clearly present that the exposure of hazardous substances by the oil spill accident had a significant impact on changes in eating habits besides economic, physical, and mental problems among the residents in Taean. Continuous health and nutrition monitoring and support are needed. (Korean J Community Nutr 16(4) : 466~472, 2011)
