      • 한방화장품의 효능 극대화를 위한 다중리포좀 제형연구

        현재 세계 유수의 화장품 회사들이 액정 기술을 접목한 제품들을 속속 출시하고 있다. 하지만 액정으로 만들어진 화장품을 출시하고 있는 기업들은 그렇게 많지 않으며 , 액정 중 생체 인지질로 만들어 액정속의 유효성분을 제조공정 중에 파괴되지 않도록 하는 기술을 보유한 기업들은 소수인 것으로 보여진다. 본 연구는 해외 유명 화장품 및 국내 화장품에 적용된 액정기술을 살펴봄으로써, 향후 우리나라 화장품 산업에 적용하여야 할 액정기술을 연구개발 하는데 도움을 주고자한다. 특히 우리 고유의 천연물 또는 한약제재 유래 제품들에 이목이 집중되고 있는 상황속에 한방 추출물의 효능을 극대화하기 위한 화장품제형으로 다중리포좀 을 연구하고 있다. 다중리포좀 제형은 피부에 유효한 유효성분들을 피부 속 깊숙이 전달함으로서 각종 피부트러블을 실질적으로 개선시킬 수 있는 제형으로 해외 유명 화장품과 비교해도 손색이 없는 우수한 화장품임을 알리고자 하며 , 이 연구를 토대로 더욱 우수한 화장품을 개발하는데 도움을 주고자 한다.

      • 가토뇌 Microtubule의 효소적 Carboxylmethylation이 Microtubule 기능 및 성상에 미치는 영향

        Microtubule purfied from rabbit brain by 2 cycles of polymerization and depolymerization in the presence of 2 mM GTP and 20% glycerol was carboxyl methylated by a purified protein methylase Ⅱ of rabbit brain, and changes in the properties of microtubule by carboxylmethylation were inverstigated. Tubulin and microtubule associated proteins were endogenously and exogenously carboxelmethvlated by protein methylase Ⅱ in a rate of 28.6 and 71.7 pmoles of methyl group incorporated into 1 mg protein for 15 minutes, respectively. Microtubule was found to be a better substrate than histone for this enzyme. Microtubule carboxylmethylation by protein methylase Ⅱ was activated 8%, 16% , 6%, 15% and 34% by 2 mM GDP, 2 mM Mg^2+, 2 mM Ca^2+, 40 μM colchicine and 40 μM podophyllotoxin, respectively. The binding of colchicine and Ca^2+ to microtubule were slightly enhanced by carboxylmethlated of microtubule. Polymerization kinetics of carhoxvlmethylated and noncarhuxylmethylated microtubule were similar. The inhibitory action of colrhicine and Ca^2+ on the microtubule polymerization was potentiated by carboxylnethylation of microtubule. These results were discussed with regard to the possible role of protein carboxylmethylation by protein methylase Ⅱ in the regulation of microtubule function.

      • 水晶體 α-Crystallin의 Aggregation에 對한 Calcium의 影響에 關한 硏究

        The elution profile of CaCI_2 treated and EGTA plus CaC1_2-treated soluble fraction protein of bovine lens are compared by Sephacryl S-300 chromatography, and then changes of protein patterns of CaCI_2-treated crystallin and EGTA plus CaCI_2-treated α-crvstallin fractions have been investigated by urea/polyacrylamide gel electrophoresis. 1. In the presence of 0.8 mM CaCI_2, 10 mM CaCI_2, or 10 mM CaCI_2 plus 20 MM EGTA, absorbance at 500 nm exhibited 0.039, 0.087, 0.040 respectively. 2. Physiologic concentration (0.8 mM) CaCI_2-treated, 10 mM CaCI_2-treated and 10 mm CaCI_2 plus 20 mM EGTA-treated soluble fractions were analyzed by Sephacryl S-300 chromatography. In the 0. 8 mM CaCI_2-treated proteins four peaks were eluted in the tube No. 17, 27, 31, 35 and identified as α, β_H, β_L, γ-crystallin, but eluted in the tube No. 16, 26, 30, 33 in 10 mM CaCI_2-treated protein, respectively. In presence of 10 mM CaCI_2 plus 10 mM EGTA, protein patterns exhibited the same. 3. Physiological concentration (0. 8 mM) CaC_2-treated α-crystallin was seperated ten subtractions by urea/polyacrylamide gel electrophoresis. In the 10 mM CaCI_2-treated α-crystallin, No. 9 subfraction was disappeared and No. 3B was newly appeared when compared with that of 0.8 mM CaCI_2 treated and 10 mM CaCI_2 plus 20 mM EGTA. 4. Absorbance and electrophoretic pattern of 10mM CaCI_2-treated native a-crystallin exhibited the same result of 0.8 mM CaCI_2-treated. 5. Ten inilimoles mercaptoethanol treated soluble fraction protein exhibited the same elution profile when compared with nontreated soluble fraction proteins. From the above results, it is suggested that the aggregation of a-crystallin of lens by calcium may be resulted to the interaction of (β,γ-crystallin, and not related the formation of disulfide bond.

        슈크로오스디스테아레이트를 사용한 액정구조의 생성과 보습효과에 관한 연구

        이 연구는 슈크로오스디스테아레이트(Sucro-DS)를 사용하여 친수형 O/W 에멀젼을 만들고, 이 에멀젼의 드로플렛(droplet)은 다중층의 구조를 가지는 액정생성에 관한 것이다. Sucro-DS 의 물리화 학적 특성을 알아보았고, 이를 이용한 유화성능에 대하여 연구하였다. 액정을 형성하기 위하여 3wt%의 Sucro-DS, 5wt%의 글리세린, 5wt%의 수쿠알란, 5wt%의 카프릭/카프릴릭트리글리세라이드, 3wt%의 세토스테아릴알코올, 1wt%의 글리세릴모노스테아레이트, 78wt%의 정제수의 혼합계에서 안정한 다중층 의 라멜라구조가 형성됨을 알 수 있었다. 이를 응용하여 불안정한 활성물질을 봉입한 크림을 만드는 방 법에 대하여 기술하였다. 또한, 이 기술을 이용한 크림의 보습효과에 대하여 연구하였다. 인체 임상시험 을 통한 피부개선효과에 대하여 연구한 결과를 보고한다. Sucro-DS 를 사용하여 안정한 액정상을 생성 하는 pH 범위는 5.2~7.5 에서 안정한 액정구조를 유지하고 있었다. 액정의 안정성이 우수한 농도는 3wt%의 베헤닐알코올을 함유할 경우의 경도는 13kg/mm,min 이었다. 동일 함량의 점도는 25,000 mPas/min 이었다. 유화제의 영향에 대하여 실험한 결과, Sucro-DS 의 농도는 5wt%가 적합하였고, 안정 한 액정의 입경분포는 4~6mm 이었다. 이를 현미경분석을 통하여 관찰하였고, 3 개월동안 액정변화의 안 정성은 4℃, 25℃, 45℃에서 안정함을 알 수 있었다. 임상시험으로, 바르기 전의 보습력은 13.4±7% 이 었다. 액정이 형성되지 않은 크림의 보습력은 14.5±5%로 바르기 전보다는 약 8.2% 보습력이 상승하였 다. 반면, 액정크림의 경우 19.2±7%로, 도포 전보다는 43.3%보습력이 상승하였다. 응용분야로 Sucro-DS 유화제를 사용한 액정크림, 로션, 아이크림 등 다양한 제형개발이 가능하고, 화장품산업은 물 론 의약품산업 및 제약산업에서 피부외용제의 유화기술로 폭넓게 응용이 가능할 것으로 기대한다. This study is to make the liquid crystalline structure using sucrose distearate (Sucro-DS) emulsifier to create the hydrophilic type oil-in-water (O/W) emulsion, the droplets of the emulsion having a structure of a multi-lamellar structure. We have studied the physicochemical properties of Sucro-DS using those techniques. And it has been studied in the emulsion performance. In order to form the liquid crystalline structure applying 3 wt% of Sucro-DS, 5 wt% of glycerin, 5 wt% of squalane, 5 wt% of capric/caprylic triglyceride, 3wt% of cetostearyl alcohol, 1wt% of glyceryl mono-stearate, 78 wt% of pure water in mixture having the lamellar structure of stable multi-layer system was found to formed. By applying them, they were described how to create an unstable active material encapsulated cream. Further, the moisturizing cream was studied using this technique. It reported the results to the skin improvement effect by the human clinical trials. The pH range to produce a stable liquid crystal phase using a Sucro-DS was maintained in 5.2~7.5. In order to increase the stability of the liquid crystal, it was when behenyl alcohol containing 3 wt%, the hardness at this time was 13 kg/mm,min. Viscosity of the same amount was 25,000mPas/min. After a test for the effects of the emulsions, the concentration of 6 wt% Sucro-DS is that was appropriate, the particle size of the liquid crystal was 4~6mm. It was observed through a microscope analysis, reliability of the liquid crystal changes for 3 months was found to get stable at each 4°C, 25°C and 45°C. In clinical trial test, before applying a moisturizing effect it was 13.4±7%. Moisturizing cream liquid crystal was not formed in 14.5±5%. Therefore, applying than ever before could see the moisture about 8.2% was improved. On the other hand, it was the moisturizing effect of the liquid cream is 19.2±7%. The results showed that 43.3% improvement than that previously used. Applications fields, Sucro-DS emulsifier used liquid cream, lotion, eye cream and a variety of formulations can be developed, as well as the cosmetics industry is expected to be wide fields in the application of the external preparation for skin emulsion technology in the pharmaceutical industry and pharmaceutical industry.

        어린이 전시 공간에 나타난 스캐폴딩 특성에 관한 연구

        (Background and Purpose) As the times have evolved, the way of education has also changed, focusing on creative education that can cultivate creative thinking in injecting education that delivers unilateral knowledge and acquires information. As the educational environment centered on contents that can cultivate creative thinking becomes important, children's exhibition spaces also need a scaffolding change from a simple play experience space to a space where they can develop creative thinking, improve problem-solving skills, and increase potential development skills. In this study, the environment creation is important to enhance the educational learning effect of the children, and it is a children's experience space centering on Vygotsky's theory of cognitive development that emphasizes social interaction through scaffolding (scaffolding) that can enhance children's learning ability. The purpose of this study is to derive the scaffolding characteristics and to show the effect on children's exhibition and experience space. Furthermore, I would like to suggest the direction of the exhibition space for children and to help research space development that raises creative thinking of children. (Method) Through prior studies of children's space, we want to identify the characteristics of exhibition space and to consider the concept of close development area and scapolding based on Vigotsky's development theory. Based on the theoretical considerations, the characteristics of scapolding appearing in the children's exhibition space were derived. In addition, the Seoul Sangsangnara, Goyang Children's Museum, Gyeonggi Children's Museum, the National Children's Museum, and the Folk Children's Museum were selected as examples to analyze the characteristics of Scapolding, which increases children's creative thinking and potential development levels.(Results) The characteristics appearing in the children's exhibition space show the same spatial characteristics as the characteristics of the scaffolding for cognitive development, and it can be concluded that this is applied to the children's exhibition and experience space in various ways. Spatial characteristics of scaffolding are classified into social series, structural motivation, and instrumental scaffolding, and it can be seen that it is a platform for children's cognitive development and creative thinking through the interaction. (Conclusions) The characteristics of various scaffolding appear in the children's exhibition space, and it proves that it is a platform for raising children's cognitive and potential development levels through interactions between experiences. It is expected that the space plan that applies the characteristics of scaffolding will serve as an important platform for children's cognitive development and creativity development in the future children's exhibition and experience space.
