The geothermal sites near neutral and alkalescent thermal springs in Tengchong Rehai National Park were examined through cultivation-dependent approach to determine the diversity of thermophilic fungi in these environments. Here, we collected soils samples in this area, plated on agar media conducive for fungal growth, obtained pure cultures, and then employed the method of internal transcribed spacer (ITS)sequencing combined with morphological analysis for identification of thermophilic fungi to the species level. In total, 102 strains were isolated and identified as Rhizomucor miehei, Chaetomium sp., Talaromyces thermophilus, Talaromyces byssochlamydoides, Thermoascus aurantiacus Miehe var. levisporus, Thermomyces lanuginosus, Scytalidium thermophilum, Malbranchea flava, Myceliophthora sp. 1, Myceliophthora sp. 2,Myceliophthora sp. 3, and Coprinopsis sp. Two species, T. lanuginosus and S. thermophilum were the dominant species, representing 34.78% and 28.26% of the sample, respectively. Our results indicated a greater diversity of thermophilic fungi in neutral and alkaline geothermal sites than acidic sites around hot springs reported in previous studies. Most of our strains thrived at alkaline growth conditions.
Action Recognition Based on Multi-scale Oriented Neighborhood Features

The spatio-temporal (ST) position information between local features plays an important role in action recognition task. To use the information, neighborhood-based features are built for describing local ST information around ST interest points. However, traditional methods of constructing neighborhood, such as sub-ST volumetric method and nearest-neighbor-based neighborhood method, ignore the orientation information of neighborhood. To make the neighborhood-based features more discriminative, we construct a novel, oriented neighborhood by imposing weights on the distance components. Specifically, in our scheme, firstly, local features are produced, and encoded by locality-constrained linear coding (LLC). Then, oriented neighborhoods are constructed by imposing weights on the distance components between features, and obtain single-scale oriented neighborhood features (SONFs). Next, multi-scale oriented neighborhood features (MONFs) are formed by concatenating SONFs. As a result, action video sequences are represented as a collection of MONFs. Finally, locality-constrained group sparse representation (LGSR) is used as classifier upon MONFs. Experimental results on the KTH and UCF Sports datasets show that our method achieves better performance than the competing local ST feature-based human action recognition methods.

Purpose: This study aims to analyze the clinical-pathological characteristics of multifocal and multicentric breast cancer (MMBC) in Chinese women. Methods: Sixty-seven cases with MMBC were randomly collected and reviewed at seven hospitals in representative districts of China during 1999 to 2008. Results: The incidence of MMBC in breast cancer in China was 1.75%. Compared to those with unifocal breast cancer, women with MMBC were more likely to have larger tumor size, lymph node metastasis (59.70% vs. 45.62%) and stage III to IV (46.26% vs. 21.10%). The peak age at onset of MMBC was 40 to 49 years old and has been gradually increasing during 1999 to 2008. Most of the MMBC women were treated with surgery and adjuvant therapy. Conclusion: In China, the incidence of MMBC in breast cancer is significantly lower than that in Western countries. Compared to unifocal breast cancer, MMBC is biologically more aggressive. Most MMBC women underwent mastectomy, instead of breast conservation surgery.

Therapeutic effects of zoledronic acid (ZOL) on giant cell tumour of bone (GCT) have been proven. Apoptosis induction was considered to be one of the mechanisms of ZOL tumour inhibition. In this study, we presented the possibility of an osteogenic differentiation stimulation mechanism of ZOL and further investigated dosage and time effects. We treated stromal cells of GCT (GCTSC) with ZOL for 48 hours at different concentrations ($0{\mu}M$, $0.01{\mu}M$, $0.1{\mu}M$, $1{\mu}M$, 5${\mu}M$, $30{\mu}M$) and assessed apoptotic and osteogenic differentiation markers with immunohistochemical techniques and real-time quantitative RT-PCR. Our results suggested that ZOL enhanced mRNA expression of Cbfa-1, osterix and osteocalcin genes with a maximum effect at $1{\mu}M$ in GCTSC. Time course experiments indicated a time dependent osteogenic differentiation effect. In conclusion, ZOL may be considered as an adjuvant in the treatment of GCT not only by inducing apoptosis but also by stimulating osteogenic differentiation of remaining tumor stromal cells after surgery.

<P><B>Abstract</B></P> <P>In this study we conducted serial kinetic studies integrated with computational simulations to judge the inhibitory effect of pyrogallol on α-glucosidase, due to the association between this enzyme and the treatment of type 2 diabetes. As a result, we found that pyrogallol bound to the active site of α-glucosidase, interacting with several key residues, such as ASP68, MET69, TYR71, PHE157, PHE158, PHE177, GLN181, HIS348, ASP349, ASP406, VAL407, ASP408, ARG439, and ARG443, which was predicted by performing a protein-ligand docking simulation. Subsequently, we evaluated the inhibitory effect of pyrogallol on α-glucosidase, and found that it induced a mixed type of inhibition in a reversible and quick-binding manner. The relevant kinetic parameters were evaluated to be: <I>IC</I> <SUB>50</SUB> =0.72±0.051mM; <I>K</I> <SUB>i</SUB> =0.37±0.018mM. A tertiary conformational change was synchronized with pyrogallol inhibition and modulation of the shape of the active site was correspondingly observed. Our study provides insight into the functional inhibitory role of pyrogallol, which results from its triple-hydroxyl groups interacting with the active site of α-glucosidase. We suggest that compounds similar to pyrogallol (phenolic hydroxyl compounds) which target the key residues of the active site of α-glucosidase could be potential agents for α-glucosidase inhibition.</P>
Multiscale Spatial Position Coding under Locality Constraint for Action Recognition

– In the paper, to handle the problem of traditional bag-of-features model ignoring the spatial relationship of local features in human action recognition, we proposed a Multiscale Spatial Position Coding under Locality Constraint method. Specifically, to describe this spatial relationship, we proposed a mixed feature combining motion feature and multi-spatial-scale configuration. To utilize temporal information between features, sub spatial-temporal-volumes are built. Next, the pooled features of sub-STVs are obtained via max-pooling method. In classification stage, the Locality-Constrained Group Sparse Representation is adopted to utilize the intrinsic group information of the sub-STV features. The experimental results on the KTH, Weizmann, and UCF sports datasets show that our action recognition system outperforms the classical local ST feature-based recognition systems published recently.
Growth and Differentiation Effects of Homer3 on a Leukemia Cell Line

The Homer protein family, also known as the family of cytoplasmic scaffolding proteins, which include three subtypes (Homer1, Homer2, Homer3). Homer3 can regulate transcription and play a very important role in the differentiation and development for some tissues (e.g. muscle and nervous systems). The current studies showed that Homer3 abnormal expression changes in acute myeloid leukemia (AML). Forced expression of Homer3 in transfected K562 cells inhibited proliferation, influenced the cell cycle profile, affected apoptosis induced by $As_2O_3$ through inhibition of Bcl2 expression, and also promoted cell differentiation induced by 12-O-tetra decanoylphorbol-acetate (TPA). These results showed that Homer3 is a novel gene which plays a certain role in the occurrence and development of AML.

To identify novel therapeutic agents to treatcancer, we synthesized a series of diaryl ether derivatives. Structure–activity relationship studies revealed that thepresence of a chlorine or hydroxyl at the para-position onthe phenyl ring (5h or 5k) significantly enhanced antitumoractivity. Compound 5h had stronger growth inhibitory activityin HepG2, A549, and HT-29 cells than compound 5k,with IC50 values of 2.57, 5.48, and 30.04 lM, respectively. Compound 5h also inhibited the growth of other cells lines,including Hep3B, PLC/PRF5, SMMC-7721, HeLa, andA375, with IC50 values of 2.76, 4.26, 29.66, 18.86, and10.21 lM, respectively. The antitumor activity of compound5h was confirmed by a colony forming assay. Further,our results indicated that the antitumor activity ofcompound 5h may be mediated by enhancing expression ofp21 and cl-caspase3, and leading to apoptosis of cancercells.

The multi-nozzle electrospinning is under extensive investigations because it is an easy way to enhance theproductivity and also feasible to produce special structure fibers such as core-shell fibers and to fabricate composite fibers ofthose polymers that cannot form blend solution in common solvent. Control over the multi-nozzle electrospinning fibersdeposition has attracted increasing attentions. The most common method was to use the auxiliary electrode. However, theconcentrated effect of the works of control multi-nozzle electrospinning deposit was inconspicuous. To enhance thecontrolling of multi-nozzle electrospinning deposition, a set-up based oppositely charged electrospinning was designed. Inthis set-up the air flow was used to transport neutralized nanofibers. This electrospinning method was named oppositelycharged and air auxiliary electrospinning (OCAAES). The capacity of OCAAES in deposition area and pattern controllingwere investigated. By the OCAAES, concentrated and several patterned nanofibers deposition were fabricated. Resultsshowed that nanofiber deposition area and pattern of multi-nozzle electrospinning could be controlled actively, and nanofiberdeposition could be fabricated in a quick thickening rate.
