      • 이담자균 효모의 성분화 과정중 인지질의 작용과 배지조성의 제한이 성분화에 미치는 영향

        The action of phospholipid on the rhodotorucine A(Rh.A) acceptance by heterobasidiomyceteous yeast Rhodospori-dium toruloides mating type a cells and the effect of medium composition during sexual differentiation were investigated. Activation of trigger peptidase(TPase) was very sensitive to the originated phospholipid from R. toruloides and was more sensitive to phospholipid liposome made up of phospholipid. Phospholipid present on the membrane of mating type a cells consists of phospatidylglycerol(PG), phosphatidylethanolamine(PE), phospatidylcholine(PC), phospatidylinositol(PI), and phosphatidylserine(PS) of 12.9, suprisingly 45.4, 11.0, and 13.9%, respectively. As the result of using C-1 and N-1 mediums which limited C and N sources capable of inhibiting the synthesis of phospholipid. it resulted inhibiting sexual differentiation and production of Rh.A from mating type A cells.

      • 이담자 효모균의 성분화과정에서 막단백질 중 Ca^2+ -ATPase와 trigger peptidase(TPase)의 상호관계

        We have studied the relation between Ca^2+-ATPase and trigger peptidase(TPase) which are membrane protein well known as their significant role for signal transduction of mating pheromone in heterobasidiomycetous yeast, Rhodosporidium toruloides. We found out that there were Ca^2+-ATPase and TPase together in isolated calmodulin binding protein(CBP), using clamodulin affinity column chromatography after solubilization of mating type a cell membrane protein, and that the dependence of enzyme activity of both the enzymes of Ca^2+, phospholipid and nonionic detergent are similar. However, Ca^2+-ATPase had quite absolute dependence on calmodulin and, on the other hand, TPase didn't have any dependence. Judging from the fact that there are both enzymes in CBP which the dependence of calmodulin are quite different, we found out that both enzymes were made to their compound and existed in mating type a cell membrane.

        [γ-^32P]ATP was used to test phosphorylation of membrane proteins of mating type a cells of heterobasidiomycetous yeast Rhodosporidium toruloides separated by non-denaturing electrophoresis. The phosphoprotein was observed in the membrane proteins. The phosphorylation was inhibited by the pheromone rhodotorucine A (Rh. A) secreted by mating type A of the yeast. Rh. A didn't inhibit the phosphorylation in the presence of a trigger peptidase (TPase) inhibitor, antipain. Partially digested Rh. A by trypsin maintained the phosphorylation inhibitory activity. These results show that TPase activity plays an important role in the transduction of pheromone signal in the yeast.

        The optimal medium composition for the production of carotenoid pigment from Haloarcular sp. EH-1 as a dietary for fishes were 1.0% sucrose, 1.0% yeast extract, 25% sodium chloride, 0.3% sodium citrate, 0.2% potassium chloride, 2.0% magnesium sulphate, 0.002% ferric sulphate(pH7.0). The incubation temperate, aeration rate and agitation speed were 40℃, 100ml medium/500ml vol. shaking flask, and 180 rpm/min. The carotenoid pigment production was highest after 5 days of incubation with the light.

      • Bacillus sp. YJ-63이 생산하는 항곰팡이 항생물질의 구조분석

        토양으로부터 분리한 Bacillus sp. YJ-63으로부터 항진균성 항생물질을 분리하여 그 구조를 분석한 결과는 다음과 같다. 항생물질은 7개의 α-amino acid와 1개의 β-amino acid가 결합한 환상구조를 가지는 iturin E 계에 속하며 특히, β-amino acid의 methylene 수가 기존의 iturin E 보다 더 많은 것으로 나타났다. Structural analysis was performed by the ^1H-NMR, ^13C-NMR, amino acid composition analysis and FAB-mass. The instrumental analysis represented that the potential antifungal antibiotic belonged to the iturin E group antibiotic, consisting of 7 α-amino acid residues and a collection of β-amino acid with aliphatic side chain. Compared to the iturin E group, notably, the potent antifungal antibiotic from bacillus sp. YJ-63 carried longer β-amino acid side chain. In conclusion, these findings identified a potential antibiotic, which contained a stable cyclopeptide structure with long β-amino acid side chain.

        과학교육에 있어서 올바른 개념의 정립은 매우 중요하다. 그런데 고등학교 「물리Ⅱ」 교과서 `에너지와 열' 단원에 나와있는 자유도에 대한 개념 설명이 교과서마다 달라서 이의 정확한 설명이 필요한 실정이다. 그래서 본 연구에서는 역학에서 사용되는 자유도와 열역학에서 사용되는 자유도에 대한 정의를 비교하고, 이를 고등학교 「물리Ⅱ」 교과서 `에너지와 열' 단원에 나와있는 단원자 및 이원자 기체분자에 적용하여 각각의 자유도를 계산하였다. 이를 통하여 자유도에 대한 올바른 개념 형성에 도움이 될 것이라 기대된다. It is very important to establish a correct concept in science education. The explanation on the degree of freedom in the section of thermodynamics of textbooks of high school level is different each other. It is needed to explain about the term in a correct and unified way. We compared definitions for degree of freedom between unit of mechanics and thermodynamics, and apply it to monatomic and diatomic ideal gas molecules used in physics textbook of high school. We hope the result play an important role to construct the concept for degree of freedom in textbooks of high school level.

      • 이담자효모 Rhodosporidium toruloides의 성pheromone(Rh.A)에 의한 성접합형 특이적 인산화 저해 반응

        Two phosphorylated proteins having molecular weights of 57kD and 72kD were detected from the solubilized membrane protein fraction of mating type a cells of Rhodosporidium toruloides which belongs to heterobasidiomycetous yeast. The phosphorylation of the protein was inhibited by a sexual pheromone, Rhodotorucine A (Rh. A), which is secreted from mating type a cells. On the other hand, counterpart mating type A cells and M-39 strain which is a sterile mutant derived from a cells, had also the same two phosphorylated proteins. However, the phosphorylation of the protein from A cells and M-39 strain were not inhibited by the Rh. A. It suggests that inhibition of the phosphorylation reaction by the Rh. A in mating type a cells is a mating-type-specific reaction that relate to transduction mechanism of sexual pheromone signaling.

        이담자 효모균 Rhodosporidium toruloides 접합형 a 세포의 막단백질 가용화 분획을 비변성 전기영동에 의하여 인산화 한 결과 인산화 되는 단백질을 발견하였다. 이 단백질의 인산화는 Rhodotorucine A (Rh. A)에 의하여 저해되었다. 본 인산화 단백질에는 trigger-peptidase (TPase)가 존재하고 있는 것을 확인 하였으며, TPase inhibitor(antipain)를 반응액에 첨가할 경우 Rh. A에 의한 인산화 저해작용은 나타나지 않았다. 그리고, Rh. A를 trypsin으로 분해한 산물을 반응액에 첨가했을 경우 인산화 저해현상이 있는 것으로 보아, 인산화의 저해반응에는 TPase가 중요한 작용을 하는 것이 확인되었다.       • 김치와 김치재료의 콜레스테롤 저하 및 항암효과

        The purpose of the paper is to explore the current knowledge on the nutritional evaluation, cholesterol-lowering effect and antitumor activity of kimchi and its ingredients(Korea cabbage, garlic, red pepper powder, ginger and onion). Kimchi contains high contents of nutrients such as vitamins(ascorbic acid, β-carotene and vitamin B complex), minerals(calcium, potassium, iron and phosphorous), essential amino acids and dietary fiber. Kimch also contains high levels of lactic acid bacteria, allicin, capsaicin, organic acid, phenol compounds, flavonoid and sulfur compounds. The dietary fiber and lactic acid bacteria isolated from kimchi are effective in improving intestinal microflora of human. Insoluble dietary fiber shows anticancer activity, but soluble dietary fiber shows hypocholesterolemic effect. Lactic acid bacteria isolated from kimchi acts as a hypocholesterolemic or anticancer agent. A major ingredient of kimchi is mainly cruciferous and allium family vegetables, which were also reported to prevent cancer and atherosclerosis. It is suggested that kimchi is important not only as one of traditional fermented Korea foods but also as therapeutic agent for carcinogenesis and hypercholesterolemic state.
