        Heteromerization of μ-opioid receptor and cholecystokinin B receptor through the third transmembrane domain of the μ-opioid receptor contributes to the anti-opioid effects of cholecystokinin octapeptide

        Activation of the cholecystokinin type B receptor (CCKBR) by cholecystokinin octapeptide (CCK-8) inhibits opioid analgesia. Chronic opiate treatment leads to an increase in the CCK-8 concentration and thus enhances the antagonism of CCK-8 against opioid analgesia; the underlying molecular mechanisms remain of great interest. In the present study, we validated the colocalization of the μ-opioid receptor (MOR) and CCKBR in pain signal transmissionrelated spinal cord dorsal horn and dorsal root ganglion neurons of rats. Co-immunoprecipitation (Co-IP) and fluorescence lifetime-imaging-microscopy-based fluorescence resonance energy transfer (FLIM-FRET) assays showed that MOR heteromerized with CCKBR directly in transfected HEK293 cells. Combined with MOR mutant construction, the third transmembrane domain of MOR (TM3MOR) was demonstrated to participate in heteromerization with CCKBR. Receptor ligand binding, ERK phosphorylation and cAMP assays showed that MOR heteromerization with CCKBR weakened the activity of MOR. A cell-penetrating interfering peptide consisting of TM3MOR and TAT (a transactivator of HIV-1) sequences from the N terminal to the C terminal disrupted the MOR–CCKBR interaction and restored the activity of MOR in transfected HEK293 cells. Furthermore, intrathecal application of the TM3MOR-TAT peptide alleviated CCK-8-injection-induced antagonism to morphine analgesia in rats. These results suggest a new molecular mechanism for CCK-8 antagonism to opioid analgesia in terms of G-protein-coupled receptor (GPCR) interaction through direct heteromerization. Our study may provide a potential strategy for pain management with opioid analgesics.

        Study on the Micro-vibration Suppression of a MnCu Spring Isolation Platform with Low Stiffness and High Damping

        Reaction wheels are the primary micro-vibration sources for microsatellites. To reduce the adverse effects of micro-vibrations, a six-degree-of-freedom (6-DOF) vibration isolation scheme with low stiffness and high damping is proposed. Through six-component force measurements, the harmonic characteristics of the micro-vibrations generated by a reaction wheel are determined. Based on an assumption of small deformation linear elasticity, a dynamic model of a convergent isolation platform is established. With high damping characteristics, helical springs made of MnCu (manganese copper) alloy are selected as the isolation element of the platform. After a modal analysis and stiffness design, the key structural parameters of the isolator are determined. According to the coupling of degrees of freedom, the transmissibility of the isolator is simulated and compared to titanium and aluminum alloys. Combined with on-ground and on-board tests, the effectiveness of the isolation platform is verified. The research shows that the 6-DOF isolation platform based on the MnCu spring realizes multidirectional low-frequency and low-magnitude micro-vibration suppression. The minimum initial isolation frequency of the designed isolator is 16.8 Hz, and the best suppression effect reaches 17.5 dB. The proposed isolator’s application successfully controls high-order harmonics and sensitive load response behaviors.

      • Sleep Duration and Cancer Risk: a Systematic Review and Meta-analysis of Prospective Studies

        To assess the risk of cancers associated with sleep duration using meta-analysis of published cohort studies, we performed a comprehensive search using PubMed, Embase and Web of Science through October 2013. We combined hazard ratios (HRs) from individual studies using meta-analysis approaches. A random effect dose-response analysis was used to evaluate the relationship between sleep duration and cancer risk. Subgroup analyses and sensitivity analyses were also performed. Publication bias was evaluated using Funnel plots and Begg's test. A total of 13 cohorts from 12 studies were included in this meta-analysis, which included 723, 337 participants with 15, 156 reported cancer outcomes during a follow-up period ranging from 7.5 to 22 years. The pooled adjusted HRs were 1.06 (95% CI: 0.92, 1.23; P for heterogeneity =0.003) for short sleep duration, 0.91 (95% CI: 0.78, 1.07; P for heterogeneity <0.0001) for long sleep duration. In subgroup analyses stratified by cancer type, long duration of sleep showed an inverse relation with hormone-related cancer (HR=0.79; 95% CI: 0.65, 0.97; P for heterogeneity =0.009) and a greater risk of colorectal cancer (HR=1.29; 95% CI: 1.09, 1.52; P for heterogeneity =0.346). Further meta-analysis on dose-response relationships showed that the relative risks of cancer were 1.00 (95% CI: 0.99, 1.01; P for linear trend=0.9151) for one hour of sleep increment per day, and 1.00 (95% CI: 0.98, 1.01; P for linear trend=0.7749) for one hour of sleep increment per night. No significant dose-response relationship between sleep duration and cancer was found on non-linearity testing (P=0.5053). Our meta-analysis suggests a positive association between long sleep duration and colorectal cancer, and an inverse association with incidence of hormone related cancers like those in the breast. Studies with larger sample size, longer follow-up times, more cancer types and detailed measure of sleep duration are warranted to confirm these results.

        Vibration Energy Transfer Characteristics of Panels with Multiple Coupling Forms in Satellites

        Micro-vibrations on-board a satellite have degrading effects on the performance of certain payloads like observation cameras. There are numerous sources and extremely complicated transfer paths for micro-vibration in satellites. The vibration energy transfer characteristics of panels with multiple coupling forms in satellites were analyzed, which can guide the vibration isolation measures of satellites. First, the satellite was divided into honeycomb panel subsystems with multiple coupling forms. In the next phase, based on the energy finite element analysis (EFEA), the energy finite element equation of the honeycomb subsystem and the connection matrix at the coupling boundary was established to determine the energy distribution and energy flow characteristics of the multiform coupled honeycomb panels under different excitation frequencies. Finally, the finite difference approximation based on the response obtained from the measurement point array was employed to obtain the energy density. By comparing the results of the experiment and EFEA simulation under different frequency excitations, the average error of the energy density was less than 4 dB, which proved the accuracy of EFEA in the transfer characteristics analysis of the vibration energy of the satellite. The experiment method of vibration transfer based on the finite difference approximation can directly obtain the energy density of the structure.
