Purpose Capecitabine is an extensively used oral prodrug of 5-fluorouracil in treatment of colon cancer and is known to cause hand-foot syndrome (HFS). As the target enzyme for capecitabine, thymidylate synthase (TYMS) plays a key role for 5-fluorouracil metabolism and has been associated with some side effects caused by capecitabine. The aim of our study is to identify the possible genetic predictors of capecitabine-induced HFS (CAP-HFS) in Chinese colorectal cancer patients.Materials and Methods Whole exons of TYMS were sequenced for 288 extreme phenotype HFS patients, including 144 severe or early-onset (first 2 cycles) moderate HFS extreme cases and 144 extreme controls with no reported HFS. The associations between polymorphisms and CAP-HFS were analyzed using logistic regression under an additive model.Results We identified a novel risk mutation (c.1A>G, chr18:657743), was associated with severe HFS in an extreme case who was affected during the first cycle of treatment. Moreover, we identified three new variants, rs3786362, rs699517, rs2790, and two previously reported variants, 5’VNTR 2R/3R and 3′-untranslated region 6-bp ins-del, which were significantly associated with CAP-HFS (p < 0.05). In silico analysis revealed that the effect of these polymorphisms in the TYMS region on the development of HFS might not be restricted solely to the regulation of TYMS expression, but also the TYMS catalytic activity through the indirect effect on ENOSF1 expression.Conclusion This study identified new polymorphisms in TYMS gene significantly associated with CAP-HFS, which may serve as useful genetic predictors for CAP-HFS and help to elucidate the underlying mechanism of HFS.
Bcl11b Regulates IL-17 Through the TGF-β/Smad Pathway in HDM-Induced Asthma

Purpose: T helper (Th) 17 cells play a critical role in the development of asthma, but the underlying mechanism of how interleukin (IL)-17 is regulated in allergic airway inflammation is poorly understood. In this study, we investigated the impact of Bcl11b on Th17 response in asthma. Methods: Blood samples from patients with mild asthma (MA) and severe asthma (SA) were collected. Expression of Bcl11b, IL-4, IL-5, IL-13, IL-17A and transforming growth factor (TGF)-β1 were determined in CD4+ T cells and plasma by polymerase chain reaction (PCR) and enzyme-linked immunosorbent assay (ELISA). Relative mRNA and protein levels of Bcl11b, IL-17A and genes involved in the TGF/Smad signaling pathway were examined by PCR, ELISA and western blot analysis in house dust mite (HDM)-challenged mice. Ectopic expression of Bcl11b in HDM-stimulated primary mouse splenocytes was achieved by nucleofection of Bcl11b expression plasmid. Results: We found significantly decreased Bcl11b but increased IL-17A and TGF-β1 expression in patients with asthma and a strongly negative correlation between Bcl11b and these 2 cytokines in SA patients. Similar expression patterns of Bcl11b, IL-17A and TGF-β1 were also found in mice with HDM-induced allergic airway inflammation. We demonstrated further that Smad2/3 phosphorylation was increased in HDM-challenged mice and that ectopic expression of Bcl11b in HDM-stimulated primary mouse splenocytes reduced Smad2 phosphorylation and IL-17 expression. Conclusions: Our findings demonstrate a potential effect of Bc111b in controlling IL-17-mediated inflammation in asthma and suggest that Bc111b may be a useful therapeutic target for asthma.
생육시기에 따른 품종별 양마 잎의 항산화 활성과 플라보노이드 함량의 변화

Background : This study analyzes changes in DPPH radical scavenging activity, total polyphenol contents, and total flavonoid contents of six different kinds of Kenaf (Hibiscus cannabinus L.) leaves which are identified as herbaceous plants, and are living in the subtropics. The data of changes in polyphenol, flavonoid contents and DPPH radical scavenging activities were analyzed, based on two different lengths of growth period.. Methods and Results : The six different kinds of cultivars ; Jangdae in Korea, and El-1, R, G-1, Ef-1, Ef-2 in the State of Israel were collected on the 43th and 62nd day from when they have planted. The collected cultivars were dried at 70℃, and 80% of methanol concentration was applied to each ground specimen. Extraction was made by immersion and filtering process at room temperature. Samples were classified into two groups, one for cultivars that were collected on the 43th day, and the other for the collection of the 62nd day. We measured the rate of DPPH radical scavenging activity, total polyphenol contents, and total flavonoid contents for every Samples. For the group of the 43th day collection, Jangdae showed the highest rate of radical scavenging activity as well as the highest polyphenol contents, and El-1had the highest flavonoid contents. For the group ofthe 62nd day collection, R demonstrated not only the highest contents of polyphenol and flavonoid, but alsothe highest rate of antioxidant activity. When we compared each cultivar from another regardless its date of collection, R resulted biggest increase in all of functional components. Conclusion : Based on the test results, useful components and antioxidant activity of kenaf leaves increase in proportion to longer growing period. Particularly, the study indicates that R can be utilized as a cosmetic ingredient crop, as the rate of increase in polyphenol and flavonoid contents were highest among the six cultivars.
Molecular characterization of HEXOKINASE1 in plant innate immunity

Hexokinase1 (HXK1) is an Arabidopsis glucose sensor that has a variety of roles during plant growth and devlopment, including during germination, fowering, and senescence. HXK1 also acts as a positive regulator of plant immune responses. Previous research suggested that HXK1 might infuence plant immune responses via responses to glu‑ cose. Plant immune responses are governed by two main pathways: PAMP-triggered immunity (PTI) and efectortriggered immunity (ETI). PTI involves the recognition of Pathogen-Associated Molecular Patterns (PAMPs) and leads to increased callose formation and accumulation of pathogenesis response (PR) proteins. ETI acts in response to efectors secreted by Gram-negative bacteria. During ETI, the membrane-localized protein RPM1-interacting protein 4 (RIN4) becomes phosphorylated in reponse to interactions with efectors and mediates the downstream response. In this study, the efects of glucose on plant immune responses against infection with Pseudomonas syringae pv. tomato DC3000 and other P. syringae strains were investigated in the presence and absence of HXK1. Infltration of leaves with glucose prior to infection led to decreases in bacterial populations and reductions in disease symptoms in wild-type Arabidopsis plants, indicating that glucose plays a role in plant immunity. Both PTI and ETI responses were afected. However, these efects were not observed in a hxk1 mutant, indicating that the efects of glucose on plant immune responses were mediated by HXK1-related pathways.

Frog or toad is an animal adoration of ancient ancestors in ethnic minorities in South China, in Li tribe tradition, frog is not only a symbol of avoiding evil and the expression of maternal love, but also the decoration of missing the ancestors. This article has discussed the origin of frog pattern, summarized the frog pattern`s forms in Li brocade. At last also discussed the common frog pattern in Li tribe.

Juglans mandshurica is a fast growing hard species, which is a tree in family of Juglandaceae and has a wide distribution in China, Korea and eastern Russia. Plant materials from J. mandshurica have extensively been used in folk medicines to prevent or cure gastric, esophageal, lung and cardiac cancer. As one chain of our searching for anticomplementary agents from natural sources, two epimeric ellagitannins, [2,3-O-4,4′,5,5′,6,6′,-hexahydroxydiphenoyl (HHDP))-(α,β)-D-glucose] (I) and pedunculagin (II) were purified from 70% acetone extracts of the stem barks of J. mandshurica by Thin Layer Chromatography and Sephadex LH-20 column chromatography approaches. The chemical structures of the isolated compounds were characterized by MS, NMR, and a careful comparation with published literatures. The epimeric ellagitannins I and II exhibited inhibitory properties against a classical pathway of complementary system with 50 % inhibitory concentrations (IC<sub>50</sub>) values of 65.3 and 47.7 μM, respectively, comparing with riliroside (IC<sub>50</sub>=104 μM) and rosmarinic acid (IC<sub>50</sub>=182 μM), which were used as positive controls. Thus, the work indicated both the two secondary metabolites possess excellent inhibitory activity and might be developed as potential anticomplementary chemicals.
