The investigation of the interaction between metal ions and DNA has always attracted much attention in the fields of bioinorganic chemistry, supramolecular coordination hemistry, and DNA nanotechnology. Its mode of action can be simply divided into two aspects. On the one hand, it is non-specific electrostatic adsorption, mainly including Na+, K+, Mg2+, Ca2+ and other physiologically regulating ions; on the other hand, it is specific covalent binding, such as Pt2+, Hg2+, Ag+ and other heavy metal ions. This article focuses on the mechanism of action between Ag+ and DNA mismatch pair C-C, and summarizes its main characterization methods and various applications. It aims to provide a certain reference for the field of biological devices. With the development of cryoelectron microscopy and liquidcell TEM, the structure of C-Ag+-C is expected to be further characterized, which will be more widely used.
An Efficient PEG/CaCl2-Mediated Transformation Approach for the Medicinal Fungus Wolfiporia cocos

Sclerotia of Wolfiporia cocos are of medicinal and culinary value. The genes and molecular mechanisms involved in W. cocos sclerotial formation are poorly investigated because of the lack of a suitable and reproducible transformation system for W. cocos. In this study, a PEG/ CaCl2-mediated genetic transformation system for W. cocos was developed. The promoter Pgpd from Ganoderma lucidum effectively drove expression of the hygromycin B phosphotransferase gene in W. cocos, and approximately 30 transformants were obtained per 10 μg DNA when the protoplast suspension density was 106 protoplasts/ml. However, no transformants were obtained under the regulation of the PtrpC promoter from Aspergillus nidulans.
A Theoretical Study on the Mechanism of Decarboxylations for Hydroxymandelate Synthase

Hydroxymandelate synthase is one type of alpha-keto acid-dependent oxygenases and plays an important role in the biosynthesis of hydroxyphenyl-pyruvate. The ferrous and ferryl intermediates in the reaction were considered to be very important in the DNA repair process. The mechanism of hydroxymandelate synthase was investigated by a theoretical method based on B3LYP/LACVP*. The data indicated that each decarboxylation went through a process of direct dioxygen insertion. The first oxidative decarboxylation experienced only one transition state, while there were two potential pathways for the second decarboxylation process. The calculation data showed that the stepwise one was favored to the concerted one due to the lower energy barrier. The process to form the iron-superoxide was the rate-limiting step. After two consecutive decarboxylations, a proton transfer reaction was needed to obtain the target products. Two possible transition states were found out to achieve this step. The one with a six-membered ring structure was preferred because of the lower activation energy. In addition, several ferryl species with high spins were captured in the whole process, which could be supplied as requisite substrates for DNA repair reaction.
Identification of Plasma Biomarkers in Drug-Naïve Schizophrenia Using Targeted Metabolomics

Objective Schizophrenia (SCZ) is a severe psychiatric disorder with unknown etiology and lacking specific biomarkers. Herein, we aimed to explore plasma biomarkers relevant to SCZ using targeted metabolomics. Methods Sixty drug-naïve SCZ patients and 36 healthy controls were recruited. Psychotic symptoms were assessed using the Positive and Negative Syndrome Scale. We analyzed the levels of 271 metabolites in plasma samples from all subjects using targeted metabolomics, and identified metabolites that differed significantly between the two groups. Then we evaluated the diagnostic power of the metabolites based on receiver operating characteristic curves, and explored metabolites associated with the psychotic symptoms in SCZ patients. Results Twenty-six metabolites showed significant differences between SCZ patients and healthy controls. Among them, 12 metabolites were phosphatidylcholines and cortisol, ceramide (d18:1/22:0), acetylcarnitine, and γ-aminobutyric acid, which could significantly distinguish SCZ from healthy controls with the area under the curve (AUC) above 0.7. Further, a panel consisting of the above 4 metabolites had an excellent performance with an AUC of 0.867. In SCZ patients, phosphatidylcholines were positively related with positive symptoms, and cholic acid was positively associated with negative symptoms. Conclusion Our study provides insights into the metabolite alterations associated with SCZ and potential biomarkers for its diagnosis and symptom severity assessment.

A physiological disorder, called tipburn, commonly occurs in lettuce (Lactuca sativa L.), particularly in greenhouse cultivation and in hot seasons, and reduces its marketability. Calcium (Ca) deficiency is the main cause of tipburn injury. In the present study, we investigated the effect of exogenous spermidine (Spd) on the occurrence of tipburn in hydroponically cultivated head lettuce (var. capitata L.) and elucidated the role of Ca ions (Ca<SUP>2+</SUP>) in its prevention with respect to absorption and transport. Ca deficiency in the nutrient solution resulted in reduced uptake of Ca<SUP>2+</SUP> in lettuce roots, thereby resulting in Ca deficiency in leaves, leading to leaf senescence and tipburn. An application of exogenous Spd promoted Ca<SUP>2+</SUP> uptake in lettuce roots under Ca deficiency and promoted Ca<SUP>2+</SUP> transportation to leaves, especially to inner leaves, thus delaying leaf senescence and reducing the incidence of tipburn. The results indicated that application of exogenous Spd is an effective method to decrease tipburn in lettuce caused by Ca deficiency.

This study aimed to validate the high yield and soluble expression of proteins carrying the transactivator of transcription (Tat) peptide tag, and further explored the potential mechanism by which the Tat tag increases expression. Escherichia coli superoxide dismutase (SOD) proteins, including SodA, SodB and SodC, were selected for analysis. As expected, the yields and the solubility of Tat-tagged proteins were higher than those of Tat-free proteins, and similar results were observed for the total SOD enzyme activity. Bacterial cells that overexpressed Tat-tagged proteins exhibited increased anti-paraquat activity compared with those expressing Tat-free proteins that manifested as SodA4SodC4SodB. When compared with an MG1655 wild-type strain, the growth of a ΔSodA mutant strain was found to be inhibited after paraquat treatment; the growth of ΔSodB and ΔSodC mutant strains was also slightly inhibited. The mRNA transcript level of genes encoding Tat-tagged proteins was higher than that of genes encoding Tat-free proteins. Furthermore, the α-helix and turn of Tat-tagged proteins were higher than those of Tat-free proteins, but the β-sheet and random coil content was lower. These results indicated that the incorporation of the Tat core peptide as a significant basic membrane transduction peptide in fusion proteins could increase mRNA transcripts and promote the high yield and soluble expression of heterologous proteins in E. coli.
New degradation feature extraction method of planetary gearbox based on alpha stable distribution

The planetary transmission system has been widely used in industry because of its various advantages. And the study on degradation feature extraction method of planetary gearbox is of major significance for mechanical system prognostics and health management (PHM). In this paper, the alpha stable distribution characteristics of planetary gearbox vibration signals in performance degradation process are verified. By observing the change of alpha stable distribution for planetary gearbox degradation experiment data, a new degradation feature extraction method based on alpha stable distribution is proposed, which is called the height of probability distribution (HPD). Through comparative analysis, it is determined that HPD has better linearity and less fluctuation compared with conventional degradation features in planetary gearbox accelerated degradation stage. Moreover, in the accelerated degradation stage, the degradation trend prediction result based on HPD is closer to the actual data than conventional degradation features no matter using Wiener-based or LSSVM-based prediction method. These conclusions indicate that the newly proposed HPD works well and gives accurate estimates for condition monitoring and degradation trend prediction of planetary gearbox.
