<P><B>Abstract</B></P> <P>Defect of dendritic cell (DC) maturation in tumor microenvironments is an important immunological problem limiting the success of cancer immunotherapy. Here, we investigated the effects of polysaccharide (cordlan) isolated from <I>Cordyceps militaris</I> on DC maturation. Phenotypic maturation of DCs by cordlan was demonstrated by the elevated expressions of CD40, CD80, CD86, MHC-I, and MHC-II molecules, and functional maturation by increased expression of IL-12, IL-1β, TNF-α, and IFN-αβ, enhanced allogenic T cell stimulation, and decreased endocytosis. Of note was that cordlan induced maturation of <I>tlr4</I> <SUP>+/+</SUP> DCs from C3H/HeN mice, but not <I>tlr4</I> <SUP>−/−</SUP> DCs from C3H/HeJ mice, suggesting the promising membrane receptor of cordlan. In addition, cordlan increased phosphorylation of ERK, p38, and JNK, and nuclear translocation of NF-κB p50/p65, which were main signaling molecules down-stream from TLR4. These results indicate that cordlan induces DC maturation through TLR4 signaling pathways.</P>

N/A Background: Systemic PUVA therapy is a widely used method for treatment of vitiligo and psoriasis. However, there have been few studies concerning complications of systemic PUVA therapy. Objective: We examined the dermatological and ophthalmologic manifestations in-patients receiving PUVA therapy. Method: We studied 42 patients who received systemic PUVA therapy for more than six months in our department. Results the results are summarized as follows l. Among the 42 patients, the number of male and female patients were 18 (42.9%) and 24 (57.1%) respectively. The mean age at the start of PUVA was 42 years and mean cumulative UVA dace was 996 J/cm². 2. Among the 42 patients, hyperpigmentation found in 9 (21.4%), PUVA lento in 8 (19.0%), prorates in 5 (11.9%) and burn in 3 (7.1%). 3. The ophthalmologic manifestations were conjunctivitis (13 cases, 31.0%), pteygium (7 cases, 16.7%), pingueculum (6 cases, 14.3%) and cataract (3 cases, 7.1%). Conclusion. Long term exposure to PUVA causes chronic clinical side effects of PUVA. Therefore careful follow-up of patients who receive long term PUVA therapy is necessary. N/A Background: The elastic fibers are a major fibrillar component of the extracellular matrix of several organs, and their presence provides elastic properties to these tissues. A variety of cytokines, growth factors, and hormones have been shown to modulate elastin gene expression. So far most interest increased the effects of external environment on elastin metabolism in the skin. It has become generally accepted that cigarette smoking contributes to accelerated coronary and peripheral vascular disease, pulmonary fibrosis and periodontal disease. Nicotine is a major component of the particulate phase of tobacco smoke. Objective : Only little is known about the molecular and cellular mechanism underlying the effect of nicotine on the skin fibroblasts. Our study was performed to determine the effects of nicotine on elastin gene expression. Method: In this study, the effects of nicotine were examined by Northern blot hybridization, chloramphenicol acetyltransferase (CAT) assay, and laser scanning microscopy in cultured human fibroblasts. Results : In Northern blot hybridization, steady-state levels of elastin mRNA were decreased 0.9-fold at 1 ㎍/mL of nicotine, 0.7-fold at 10 ㎍/mL and 0.5-fold at 100 ㎍/mL, compared to untreated control. Nicotine caused a marked alteration in the elastin mRNA expression in a dose-related fashion. In CAT assay, the relative elastin CAT activity was 1.0 in the untreated control, 0.9 at a concenturation of 1 ㎍/mL, 0.3 at 10 ㎍/mL, and 0.2 at 100 ㎍/mL. Nicotine caused a marked decrease on elastin promoter activity. In laser scanning microscopy, the immunosignal for elastin in nicotine-treated fibroblasts shows in less intense than in untreated control. Conclusion: These results indicate that nicotine may be a powerful down-regulator of elastin production, suggesting transcriptional depression of gene expression in cultured skin fibroblasts.

Recessive dystrophica epidermolysis bullosa(RDEB) is rare, chronic non - inflammatory bullous disease of hereditary trait, which produces blister formation in early childhood. A localized, less severe form, often called RDEB-mitis, occurs at birth and usually involves the acral areas, with atrophic scarring of the joint surfaces and nail dystrophy but little mucosal involvement. Severe RDEB is a multilating disease, known also as the Hallopeau-Siemens (HS) varient. We report three cases which developed at birth and infancy, respectively. Bullae, atrophic scars were noted on the knees and ankles, and milia on the both hands and feet. Histologically, sections from the old blister lesion showed characteristic separation of the epidermis from the dermis forming bulla. Identification of the mutations COL7A1 gene was revealed by direct sequencing of each exon. We present three cases of RDEB-mitis considering the abscence of family history with clinical, histological and molecular analysis findings. (Korean J Dermatol 2001;39(10): 1138-1143)

Paecilomyces cicadae Miquel Samson is the anamorph of Cordyceps cicadae Shing and is used in functional foods for the prevention and treatment of various diseases. In the present study, we examined the effects of P. cicadae polysaccharide (PCP) on dendritic cell (DC) maturation. Phenotypic maturation of DCs by PCP was confirmed by the elevated expressions of CD80, CD86, major histocompatibility complex (MHC)-I, and MHC-II molecules and functional maturation by increased expression of interleukin-12, interleukin-1β, and tumor necrosis factor-α, enhanced allogenic T cell stimulation, and decreased endocytosis. PCP induced the maturation of DCs from C3H/HeN and C57BL/6 mice but not from Toll-like receptor (tlr) 4–/– knockout mice and TLR4-mutated C3H/HeJ mice, which suggests that TLR4 is the membrane receptor for PCP. PCP increased the degradation of inhibitor of nuclear factor-κB (NF-κB) α/β, which enhanced the nuclear translocation of NF-κB p50/p65 and induced the phosphorylation of extracellular signal-regulated kinase, c-Jun N-terminal kinase, and p38 mitogen-activated protein kinases, which are signaling molecules downstream of TLR4. These results indicate that PCP induces DC maturation through TLR4 signaling.

To know the correlation of punch-biopsy proven cervical intraepithelial neoplasia (CIN) 3 with the results of cone and hysterectomy. Methods We reviewed the medical records from 1999 to 2008, retrospectively, at a single institution. We checked age, cone and hysterectomy biopsy results, preoperative cervical smear results as well as human papillomavirus results. Results There were 104 patients whose punch biopsy results were CIN 3. 59 (56.7%) patients underwent cone and 45 did hysterectomy after punch biopsies. Cone found 6.8% of cervical cancer. Among patients undergoing hysterectomy without cone, 8.9% had cervical cancers. There were a lot of inflammations, atypical squamous cells of unknown significance, low-grade squamous intraepithelial lesion done as a Pap results just before cone. The positive rate of HPV was 80.0%. There were 5.1% of underestimated and 6.8% of over-estimated results according to the cone results, and 6.7% of under-estimated and 8.9% of overestimated according to the hysterectomy results. Conclusion Through conization, 93.2% of the patients whose punch biopsy results were CIN 3 had CIN and did not need hysterectomy, and 6.8% had cervical cancers and might need further treatment. Through hysterectomy, 91.2% had CIN and might undergo overtreatment.
