        불소도포가 상아질 접착에 미치는 영향

        AbstractThe purpose of this study was to investigate the effect of stannous fluoride on the dentin bonding with three kinds of commercially available dentin bonding systems containing diffe- rent adhesive monomers. Dentin specimens with exposed labial dentin prepared from freshly extracted bovine mandibular anterior teeth were divided into experimental and control groups. The specimens of experimental groups were bonded with dentin bonding systems and composite resins including All bond 2 & Bisfil, Scotchbond Multi-Purpose & Z100, and Denthesive II & Charisma after 2% stannous fluorided application for 5 minutes and washing for 1 minute. The specimens of control groups were bonded with the same dentin bonding systems and composite resins as used in the experimental groups. After bonded specimens were stored in 37? distilled water for 24 hours, the tensile bond strength and cohesive failure rate were measured, and then the pretreated dentin surfaces and the fractured dentin surfaces were examined under scanning electron microscope.The results were as follows :Mean bond strength of stannous fluoride applied groups of All bond 2, Scotchbond MP, and Denthesive II were 2.5MPa, l.lMPa, and l.lMPa respectively, and those of control groups were 7.5MPa, 8.1MPa, and 4.6MPa. Bond strength values of stannous fluoride applied groups were significantly lower than those of the control groups(pOn SEM observation of the fractured dentin-r'esin interface, while most of the specimens of stannous fluoride applied groups showed adhesive failure mode, those of All bond 2 and Scotchbond MP control groups showed mainly adhesive-cohesive mixed failure mode, and mainly adhesive failure mode in Denthesive II control group.

        From the water fraction of the MeOH extract, three compounds, 1,3,4,5-terrahydroxycyclohexanecarboxylic acid 3-(3,4-dihydroxycinnamate) (chlorogenic acid), quercetin 3-O-beta-D-galactopyranoside (hyperoside), and 1(R)-hydroxy-3,4-seco-lup-4(23),20(30)-dien-3,11alpha-olactone-1apha-L-rhamnopyranosyl(1->4)-beta-D-glucopyranosyl(1->6)-beta-L-glucopyranosyl ester (chiisanoside) were isolated and their strutures determinated by 1H-NMR, 13C-NMR, IR, and FAB-Nass. Chlorogenic acid and Chiisanoside had been quantitated by HPLC from eight Acanthopanax species per 10g A. koreanum 19.82, 4.17mg, A. nambunensis 65.00, 1.86mg, A chiisanense 67.19, 4.72mg, A. albeofructus 69.58, 4.96mg, A. chungbunensis 31.96, 6.73mg, A. pedunclus 13.95, 1.52mg, A. senticosus 27.19, 8.17mg, A. sessiliflorum 7.49, 5.88mg.

        Anti-arthritic Effect of Glucosamine and Oriental Herbal Composition

        This study was conducted to investigate the effect of an anti-inflammatory and analgesic action of the glucosamine HCl (Glucosamine) and SH-1 (Glucosamine + Oriental herbal composition combined group). Male sprague-Dawley rats (200~250 g) and ICR mice (20~30 g) were randomized and these experimental groups were divided into 4 groups. Two control group were given as negative control (saline) and positive control (Ibuprofen, 100 ㎎/㎏) and two groups given as oral administration of Glucosamine (320 m/㎏) and SH-1. Carrageenan induced paw edema test, hot plate method, croton oil induced granuloma, capillary permeability test and acetic acid writhing syndrome were also shown to be comparable in the SH-1 group to anti-inflammatory drug group such as positive control group (Ibuprofen). Although further studies should be performed to confirm the effects of SH-1, present results suggest that the combined administration of SH-1 have potential action in anti-inflammatory and analgesic action. It could be applicable for the improvement of arthritic symptoms as a new diet-supplement.

      • Combination of Multiple Spectral Libraries Improves the Current Search Methods Used to Identify Missing Proteins in the Chromosome-Centric Human Proteome Project

        <P>Approximately 2.9 billion long base-pair human reference genome sequences are known to encode some 20 000 representative proteins. However, 3000 proteins, that is, ∼15% of all proteins, have no or very weak proteomic evidence and are still missing. Missing proteins may be present in rare samples in very low abundance or be only temporarily expressed, causing problems in their detection and protein profiling. In particular, some technical limitations cause missing proteins to remain unassigned. For example, current mass spectrometry techniques have high limits and error rates for the detection of complex biological samples. An insufficient proteome coverage in a reference sequence database and spectral library also raises major issues. Thus, the development of a better strategy that results in greater sensitivity and accuracy in the search for missing proteins is necessary. To this end, we used a new strategy, which combines a reference spectral library search and a simulated spectral library search, to identify missing proteins. We built the human iRefSPL, which contains the original human reference spectral library and additional peptide sequence-spectrum match entries from other species. We also constructed the human simSPL, which contains the simulated spectra of 173 907 human tryptic peptides determined by MassAnalyzer (version 2.3.1). To prove the enhanced analytical performance of the combination of the human iRefSPL and simSPL methods for the identification of missing proteins, we attempted to reanalyze the placental tissue data set (PXD000754). The data from each experiment were analyzed using PeptideProphet, and the results were combined using iProphet. For the quality control, we applied the class-specific false-discovery rate filtering method. All of the results were filtered at a false-discovery rate of <1% at the peptide and protein levels. The quality-controlled results were then cross-checked with the neXtProt DB (2014-09-19 release). The two spectral libraries, iRefSPL and simSPL, were designed to ensure no overlap of the proteome coverage. They were shown to be complementary to spectral library searching and significantly increased the number of matches. From this trial, 12 new missing proteins were identified that passed the following criterion: at least 2 peptides of 7 or more amino acids in length or one of 9 or more amino acids in length with one or more unique sequences.         Efficient prefractionation of low-abundance proteins in human plasma and construction of a two-dimensional map

        <P>Human plasma is the most clinically valuable specimen, containing not only a dynamic concentration range of protein components, but also several groups of high-abundance proteins that seriously interfere with the detection of low-abundance potential biomarker proteins. To establish a high-throughput method for efficient depletion of high-abundance proteins and subsequent fractionation, prior to molecular analysis of proteins, we explored how coupled immunoaffinity columns, commercially available as multiple affinity removal columns (MARC) and free flow electrophoresis (FFE), could apply to the HUPO plasma proteome project. Here we report identification of proteins and construction of a human plasma 2-DE map devoid of six major abundance proteins (albumin, transferrin, IgG, IgA, haptoglobin, and antitrypsin) using MARC. The proteins were identified by PMF, matching with various internal 2-DE maps, resulting in a total of 144 nonredundant proteins that were identified from 398 spots. Tissue plasminogen activator, usually present at 10–60 ng/mL plasma, was also identified, indicative of a potentially low-abundance biomarker. Comparison of representative 2-D gel images of three ethnic groups (Caucasian, Asian-American, African-American) plasma exhibited minor differences in certain proteins between races and sample pretreatment. To establish a throughput fractionation of plasma samples by FFE, either MARC flow-through fractions or untreated samples of Korean serum were subjected to FFE. After separation of samples on FFE, an aliquot of each fraction was analyzed by 1-D gel, in which MARC separation was a prerequisite for FFE work. Thus, a working scheme of MARC → FFE → 1-D PAGE → 2-D-nanoLC-MS/MS may be considered as a widely applicable standard platform technology for fractionation of complex samples like plasma.</P>

        심장 이식 수술의 마취 1예 보고

        Cardiac transplantation has become an acceptable therapy for patients with end stage heart disease over the last decade. We experienced a case of heart transplantation to the 50 year old female patient. We used sterile technique all the time to reduce the incidence of infection. Anesthesia was induced and maintained with fentanyl 100 ㎍/kg. After cardiopulmonary bypass, blood products, inotropics and vasodilators were given to maintain myocardial contractility and cardiac output. The patient recovered uneventfully and discharged on 29th postoperative day.

        <P>A mobile phone with a glucometer integrated into the battery pack (the 'Diabetes Phone') was launched in Korea in 2003. We compared its effect on management of type 2 diabetes to the Internet-based glucose monitoring system (IBGMS), which had been studied previously. We conducted a randomized trial involving 69 patients for three months. Participants were assigned to an Internet group or a phone group. The phone group communicated with medical staff through the mobile phone only. Their glucose-monitoring data were automatically transferred to individual, web-based charts and they received medical recommendations by short message service. The Internet group used the IBGMS. There were no significant differences between the groups at baseline. After three months' intervention, HbA(1c) levels of both groups had decreased significantly, from 7.6% to 6.9% for the Internet group and from 8.3% to 7.1% for the phone group (P < 0.01). Levels of patient satisfaction and adherence to medical advice were similar. Mobile, bidirectional communication between doctors and patients using the diabetes phone was as effective for glucose control as the previously-studied Internet-based monitoring system and it was good for patient satisfaction and adherence.</P>

        Antithrombotic activity of Vitis labrusca extract on rat platelet aggregation

        <P>Vitis labrusca is a grapevine that has antioxidant, neuroprotective, hepatoprotective, and anticarcinogenic activity. However, the antithrombotic effect of Vitis labrusca leaves on platelets is yet to be ascertained. We investigated the inhibitory effect of V. labrusca leaf extract (VLE) on platelet aggregation in vitro and ex vivo. The thromboxane B-2 (TXB2) and serotonin concentrations were measured by ELISA. The flavonoids content was measured by ultraperformance liquid chromatography (UPLC). The antithrombotic activity of VLE was evaluated using various agonists in vitro. VLE strongly inhibited adenosine diphosphate (ADP)-induced platelet aggregation. In rats, VLE treatment (100mg/kg) reduced ADP-stimulated platelet aggregation, without affecting tail bleeding and coagulation time. Moreover, VLE significantly suppressed TXB2 and serotonin secretion. UPLC analysis indicated that VLE contains quercetin, isorhamnetin, and rutin. Our results indicate that VLE possesses antiplatelet activity via the suppression of TXB2 and serotonin, without affecting bleeding. Further, we identified the flavonoids present in VLE.
