<P>Anethole is known to possess anti-inflammatory and anti-tumor activities and to be a main constituent of fennel, anise, and camphor. In the present study, we evaluated anti-metastatic and apoptotic effects of anethole on highly-metastatic HT-1080 human fibrosarcoma tumor cells. Despite weak cytotoxicity against HT-1080 cells, anethole inhibited the adhesion to Matrigel and invasion of HT-1080 cells in a dose-dependent manner. Anethole was also able to down-regulate the expression of matrix metalloproteinase (MMP)-2 and -9 and up-regulate the gene expression of tissue inhibitor of metalloproteinase (TIMP)-1. The similar inhibitory effect of anethole on MMP-2 and -9 activities was confirmed by zymography assay. Furthermore, anethole significantly decreased mRNA expression of urokinase plasminogen activator (uPA), but not uPA receptor (uPAR). In addition, anethole suppressed the phosphorylation of AKT, extracellular signal-regulated kinase (ERK), p38 and nuclear transcription factor kappa B (NF-κB) in HT-1080 cells. Taken together, our findings indicate that anethole is a potent anti-metastatic drug that functions through inhibiting MMP-2/9 and AKT/mitogen-activated protein kinase (MAPK)/NF-κB signal transducers.</P>
Role of Ganglioside GD1a on the Differentiation of Human Mesenchymal Stem Cell into Osteoblast

We isolated human mesenchymal stem cells (hMSC) from dental pulp. These cells are multipotent and serve as precursors for various mesoderm-type cells such as osteoblasts, chondroblasts and adipocytes, as well as several other types of cells. This study investigated the possible role of gangliosides in osteoblast differentiation of hMSCs. Gangliosides, which exist in glycosphingolipid-enriched domains on the cell membrane The role of gangliosides that play in osteoblastogenesis is not yet clearly understood. Therefore, in this study, We investigated the relationship between gangliosides and osteoblast differentiation of human mesenchymal stem cells (hMSCs). The results of high-performance thin-layer chromatography (HPTLC) showed that ganglioside GD1a expression was increased during the differentiation of hMSCs into osteoblasts. Furthermore, we showed that involvement of alkaline phosphate (ALP) with GD1a by immunostaining. Taken together, these results suggest that gangliosides GD1a may play a role in the osteoblast differentiation process of hMSCs.
Secretion of adenylate kinase 1 is required for extracellular ATPsynthesis in C2C12 myotubes

Extracelular ATP (exATP) has been known to be a crit-ical ligand regulating skeletal muscle diferentiation and contractibility. ExATP synthesis was greatly in-creased with the high level of adenylate kinase 1 (AK1) and ATP synthase βexATP synthesis was abolished by the knock-down of AK1 but not by that of ATP synthase β in C2C12 my-otubes, suggesting that AK1 is required for exATP syn-thesis in myotubes. However, membrane-bound AK1β was not involved in exATP synthesis because its ex-pression level was decreased during myogenesis in spite of its localization in the lipid rafts that contain vari-ous kinds of receptors and mediate cell signal trans--ingly, cytoplasmic AK1 was secreted from C2C12 my-otubes but not from C2C12 myoblasts. Taken together all these data, we can conclude that AK1 secretion is required for the exATP generation in myotubes.

Human dental pulp-derived stem cells (hDPSCs) have been considered alternative sources of adult stem cells because of their potential to differentiate into osteoblasts, chondroblasts and adipocytes. The osteoblast is a mononucleate cell that is responsible for bone formation. Gangliosides are key lipid molecules that are required for the regulation of cellular processes such as proliferation and differentiation. The role of gangliosides that play in osteoblastogenesis is not yet clearly understood. Therefore, in this study, we demonstrated the clearly role of gangliosides in the osteoblast differentiation of hDPSCs by shRNA. The results of high-performance thin-layer chromatography (HP-TLC) showed that ganglioside GD1a expression was increased during the differentiation of hMSCs into osteoblasts. We showed a localization of ganglioside GD1a with ALP by immunofluorescence staining. Biosynthesis of ganglioside GD1a synthesize with the formation of sialic acid from GM1a by ST2 beta-galactoside alpha-2,3-sialyltransferase 2 (ST3Gal2). We used lentivirus-mediated shRNA expression for knockdown of ST3Gal2 gene expression. Down-regulation of ganglioside GD1a was decreased GD1a expression and ALP activity. Furthermore, these data indicate that the knockdown of ST3Gal2 by shRNA is inhibition of ERK1/2-phosphorylation. Taken together, these results suggest that ganglioside GD1a may play a role in the osteoblast differentiation process of hDPSCs.

The purpose of this study is to understand what we have learnt thus far on the sustainable consumption of natural resources by household in Korea. We propose a two-wing model designating households as the main agent of sustainable living where both the production of human resources and the consumption of natural resources take place. In the current study, we focused on the sustainability of natural resource consumption. Undertaking an empirical study, we selected 10 major journals that represent each area of human ecological research and then, identified 120 relevant studies regarding sustainability published since 2000. First, we classified 381 keywords into three categories: production, consumption, and social environment, and the identified studies were then classified in one of the seven categories created by combinations of the abovementioned three categories: production (P), consumption (C), social environment (S), productionconsumption (PC), production-social environment (PS), consumption-social environment (CS), and productionconsumption- social environment (PCS). The results showed that the sustainability of natural resources has been mainly investigated in the fields of clothing & textiles and food & nutrition. Further, consumer science research has been conducted in the area of consumption-social environment and consumption-production. The current study indicates that the field of human ecology has created and accumulated knowledge to support sustainable living. In the future, we need to conduct more integrated research across disciplines and methods and promote research interest in the underdeveloped area of sustainability research in human ecology.

Cell surface gangliosides, which exist in glycosphingolipids (GSLs)-enriched domains, play important regulatory roles in cell proliferation and differentiation .Recently, gangliosides are known as important role in growth of neuronal progenitor cells. In this study, the relationship between gangliosides expression and neuronal cell development was investigated using an in vitro model of neuronal differentiation from human mesenchymal stem cells (hMSCs) that are able to differentiate into a range of specific cell types in vitro and in vivo. First, to verify the isolated population contained pure hMSCs, we performed positive and negative characterization by fluorescence- activated cell sorting (FACs) analysis, thus we identified pure hMSCs. Next, hMSCs were differentiated into neuronal cell withα-MEM + 20%FBS + 1mM β-mercaptoethanol (BME) for 24h and transferred to serum free α-MEM + 2mM BME for 5 hour. Neuronal induced cells maintenance with α-MEM + 10%FBS + 2% DMSO +200uM β- hydroxyanisole (BHA) + 10ng/mL bFGF + 10ng/mL EGF + 25ng/mL NGF for 2weeks. High-performance thin-layer chromatography (HPTLC) showed that gangliosides GM3, GD3 and GD1a expressed in differentiated into neuronal cells for 2weeks, especially GD3 increased comparison to control cells. Immunofluorescence staining also agreed with the results of HPTLC assay. These differentially expressed gangliosides suggest that gangliosides may have specific functions in stem cells and during neuronal differentiation. Therefore, these result also suggest that regulation of gangliosides expression have used as the marker for differentiated neuronal cell from hMSCs.
