        Metastasis associated genomic aberrations in stage II rectal cancer

        Genomic aberrations of rectal carcinoma, especially DNA copy number changes associated with metastasis were largely unclear. We aim to identify the metastasis associated biomarkers in stage II rectal cancer. Formalin-fixed, paraffin-embedded primary tumor tissues of stage II rectal carcinoma were analyzed by array-based comparative genomic hybridization, and genomic aberrations were identified by Genomic Workbench and SAM software. Copy number changes and mRNA expressions were validated by Real-time PCR in an independent rectal cancer samples. The results showed that the most frequent gains in stage II rectal cancer were at 1q21.2-q23.1, 3p21.31, 11q12.2-q23.3, 12q24.11-q24.31, 12q13.11-q14.1 and losses in 18q11.2-q23, 17q21.33-q22, 13q31.1-q31.3, 21q21.1-q21.3, 8p23.3-p23.1 and 4q22.1-q23. Twenty-two amplifications and five homozygous deletions were also identified. We further found that S100A1 (1q21.3-q23.1), MCM7 (7q22.1) and JUND (19p13.11) were amplified and overexpressed in stage II rectal cancer. Interestingly, the genomic aberrations affected 14 signaling pathways including VEGF signaling pathway and fatty acid metabolism. Most importantly, loss of 13q31.1-q34 and gain of 1q44 were associated with distant metastasis. Our results indicated that these metastasis associated genomic changes may be useful to reveal the pathogenesis of rectal cancer metastasis and identify candidate biomarkers.

        A Cross-national Analysis of Global Corporate Web Sites: A Holistic Approach

        The purpose of this paper is to examine the content of multinational corporations’ (MNCs’) web sites from a holistic viewpoint. Specifically, seven types of content, including communication, general information, marketing information, employment information, financial information, service, and social issues, were analyzed to compare eleven cultural clusters: Anglo, Arab, Nordic, Germanic, Latin American, Near Eastern, Latin European, Eastern European, African, Far Eastern, and Confucian. Overall, some content types, including communication, general, marketing and social issues, showed significant differences across cultural clusters, while there were no differences in employment information, financial information and service across cultural clusters. We also calculated standardization scores to determine the degree to which MNCs standardized content. Marketing information had the highest standardization scores, followed by financial information, service, employment information, general informaion, social issues and communication, while communication and social issues had relatively low standardization scores. The results of this study indicate some similarities and dissimilarities across the clusters regarding standardization levels. Some clusters, including Arab, Latin America, East Europe, Latin Europe, Nordic, Germanic and African, showed greater similarities than did Confucian, Far East and Near East. In particular, some cultural clusters, such as Confucian, Far East and Near East, showed unique patterns of standardization.

        Spectral Factorization for Multiple Input Delayed Discrete-Time Systems with Applications to Control

        This paper deals with the linear quadratic regulation problems for the linear discrete-time systems with input delays. The design of the optimal control law is transformed into solving one Diophantine equation and one spectral factorization with delays. A new and simple approach for the spectral factorization is proposed based on reorganized innovation analysis. The calculation of spectral factor comes down to solving Riccati equations with the same dimension as the original systems.

        Information Filtering for Time-delay Systems

        This paper presents the information filter, which is developed in filtered forms, for time-delay systemswhich include continuous-time case and discrete-time case. By introducing delay-free observation, the optimalfiltering problems for time-delay systems can be converted into multiple steps prediction problems for delay-freesystems. Then, the recursive forms for optimal filter are given via innovation approach in Hilbert space. Furthermore,the information filter in Riccati recursions can be derived by using the matrix inversion Lemma to optimalfiltering formulas. Finally, the distributed Kalman filter for time-delay systems is designed based on informationfilter developed.

        A 2D/2D BiPO4/g-C3N4-B Z-type heterojunction for enhanced photocatalytic degradation of dye pollutants

        A 2D/2D BiPO4/g-C3N4-B nano-sheet heterojunction photocatalyst was synthesized via a simple coprecipitation method at room temperature using glacial acetic acid as solvent, which showed excellent activity toward the degradation of rhodamine B (RhB). The heterojunction showed much higher efficiency of separation and transfer of photogenerated carriers compared to that of its constituents. Moreover, the spectral response range of BiPO4 was effectively broadened after the combination of g-C3N4-B and BiPO4. Consequently, a 97.3% degradation of RhB within 25 min by BiPO4/g-C3N4-B heterojunction photocatalyst under visible light irradiation was observed. The difference in work functions of BiPO4 and g-C3N4-B was evident from UPS characterization, which led to the bending of the energy band and the establishment of an internal electric field at the interface of the heterojunction. Therefore, the synthesized direct Z-type BiPO4/g-C3N4-B heterojunction enhanced the oxidation-reduction ability by promoting the effective separation of photogenerated carriers.

        Production of Transgenic Goats by Sperm-mediated Exogenous DNA Transfer Method

        In this study, the production of transgenic goats using sperm to integrate exogenous DNA and artificial insemination (AI) was carried out and the technical protocols for sperm-mediated gene transfer (SMGT) in the goat were optimized. The standard sperm parameters and the ability to bind foreign genes were assessed to select suitable sperm donor bucks. A total of 134 oestrous does were divided into 4 groups and inseminated using different methods and sperm numbers. The does of Groups I to III were inseminated with fresh semen ($1-2\times10^{7}$ and $10^{6}$ sperm) or frozen-thawed semen ($10^{6}$ sperm), respectively, through conventional intra-cervical AI, and the does of Group IV with frozen-thawed semen ($10^{6}$ sperm) through intrauterine AI. Total genomic DNAs were extracted from ear biopsies of the offspring. The presence of $pEGFP-N_{1}$ DNA was screened by PCR and then by Southern blotting analysis. A total of 76 live kids were produced and 8 kids were tested transgene positive on the basis of agarose gel electrophoresis of the PCR-amplified fragment. Southern blotting analysis of the samples showed 5 positive kids. A transgenic ratio of 10.53% was detected using PCR and 6.58% using Southern blotting. The positive kid rate assayed by PCR and Southern blotting of frozen-thawed goat semen was 3.61% and 9.27% higher than that of untreated semen. The results show that transgenic goats can be produced efficiently by the method of artificial insemination using sperm cells to integrate the exogenous DNA and intrauterine insemination allowed low numbers of DNA-transfected spermatozoa to be used, with satisfactory fertility.

        Synergic enhancing-effect of DEET and dodecylamine on the skin permeation of testosterone from a matrix-type transdermal delivery system.

        <P>The synergic in vitro skin permeation enhancing-effect of N,N-diethyl-m-toluamide (DEET) and dodecylamine was investigated in order to develop a novel non-scrotal matrix-type transdermal delivery system of testosterone (TS). When DEET was loaded in DuroTak 87-2510 together with 2% TS and 3% dodecylamine, the in vitro rat skin permeation rate of TS synergistically increased as DEET concentration increased up to 0.5%. No further increase in permeation was observed thereafter and a plateau was observed up to 3.8% DEET. Moreover, compared to 0.5% DEET concentration, the addition of 3.8% of DEET in combination with 3% dodecylamine and 6% TS further increased the permeation rate of TS, and the maximum permeation rate of 11.21 microg/cm(2)/h was achieved. The in vitro skin permeation rates of TS from a transdermal delivery system of DuroTak 87-2510 containing 6% TS, 3% dodecyamine, and 3.8% DEET were in the following order: hairless mouse skin > rat skin > human cadaver skin. Assuming that a system with a surface area of 60 cm(2) is applied, the human cadaver skin permeation rate of 5.74 microg/cm(2)/h achieved in this study can be interpreted as being equivalent to delivering approximately 8. 27 mg of TS per day. Considering that the commercially available product (Testoderm TTS) for non-scrotal skin of the same surface area is designed to administer 5 mg of TS per day, the new formulation could maintain therapeutic plasma concentration of TS at a smaller surface area of 40 cm(2).</P>

      • Association between the NQO1 C609T Polymorphism with Hepatocellular Carcinoma Risk in the Chinese Population

        Background: Associations between the NQO1 C609T polymorphism and hepatocellular carcinoma (HCC) risk are a subject of debate. We therefore performed the present meta-analysis to evaluate links with HCC susceptibility. Materials and Methods: Several major databases (PubMed, EBSCO), the Chinese national knowledge infrastructure (CNKI) and the Wanfang database were searched for eligible studies. Crude odds ratios (ORs) with 95% confidence intervals (CIs) were used to measure the strength of associations. Results: A total of 4 studies including 1,325 patients and 1,367 controls were identified. There was a significant association between NQO1 C609T polymorphism and HCC for all genetic models (allelic model: OR=1.45, 95%CI=1.23-1.72, p<0.01; additive model: OR=1.96, 95%CI=1.57-2.43, p<0.01; dominant model: OR=1.62, 95%CI=1.38-1.91, p<0.01; and recessive model: OR=1.53, 95%CI=1.26-1.84, p<0.01). On subgroup analysis, similarly results were identified in Asians. For Asians, the combined ORs and 95% CIs were (allelic model: OR=1.50, 95%CI=1.24-1.82, p<0.01; additive model: OR=2.11, 95%CI=1.48-3.01, p<0.01; dominant model: OR=1.69, 95%CI=1.42-2.02, p<0.01; and recessive model: OR=1.59, 95%CI=1.16-2.19, p<0.01). Conclusions: The current meta-analysis suggested that the NQO1 C609T polymorphism could be a risk factor for developing HCC, particularly in the Chinese population.
