        Trophic Effect of Bee Pollen on Small Intestine in Broiler Chickens

        In this study, the effects of bee pollen on the development of digestive organs were evaluated in broiler chick-ens. A total of 144 1-day-old AA broiler chickens were randomly and equally divided into two groups, assigned as the con-trol group and the pollen group, respectively. The control group was fed with a basic diet, while the pollen group was fedwith a basic diet supplemented with 1.5% bee pollen over a period of 6 weeks. At the end of each week, the digestive organswere obtained for comparison from 12 broilers randomly selected from each group. The results demonstrated that comparedto the control group, the small intestine villi from the duodenum, jejunum, and ileum were longer and thicker in the pollengroup. This difference was more significant during early development, especially through the first 2 weeks. Bee pollen in-creased the length of the villi by 37.1% and 29.4% in the duodenum, 28.1% and 33.7% in the jejunum, and 18.6% and 16.2%in the ileum in week 1 and 2, respectively. Furthermore, the small intestinal glands were developed at a higher density in thepollen group, and the depth of the glands was significantly increased by bee pollen in the first 2 weeks. These findings sug-gest that bee pollen could promote the early development of the digestive system and therefore is a potentially beneficial foodsupplement for certain conditions, such as short bowel syndrome.

      • Aberrant DNA Methylation and Epigenetic Inactivation of hMSH2 Decrease Overall Survival of Acute Lymphoblastic Leukemia Patients via Modulating Cell Cycle and Apoptosis

        Objective: Altered regulation of many transcription factors has been shown to play important roles in the development of leukemia. hMSH2 can modulate the activity of some important transcription factors and is known to be a regulator of hematopoietic differentiation. Herein, we investigated epigenetic regulation of hMSH2 and its influence on cell growth and overall survival of acute lymphoblastic leukemia (ALL) patients. Methods: hMSH2 promoter methylation status was assessed by COBRA and pyrosequencing in 60 ALL patients and 30 healthy volunteers. mRNA and protein expression levels of hMSH2, PCNA, CyclinD1, Bcl-2 and Bax were determined by real time PCR and Western blotting, respectively. The influence of hMSH2 on cell proliferation and survival was assessed in transient and stable expression systems. Results: mRNA and protein expression of hMSH2 and Bcl-2 was decreased, and that of PCNA, CyclinD1 and Bax was increased in ALL patients as compared to healthy volunteers (P<0.05). hMSH2 was inactivated in ALL patients through promoter hypermethylation. Furthermore, hMSH2 hypermethylation was found in relapsed ALL patients (85.7% of all cases). The median survival of patients with hMSH2 methylation was shorter than that of patients without hMSH2 methylation (log-rank test, P=0.0035). Over-expression of hMSH2 in cell lines resulted in a significant reduction in growth and induction of apoptosis. Conclusions: This study suggests that aberrant DNA methylation and epigenetic inactivation of hMSH2 play an important role in the development of ALL through altering cell growth and survival.

      • Rapid construction delivery of COVID-19 special hospital: Case study on Wuhan Huoshenshan hospital

        Infectious disease emergency hospitals are usually temporarily built during the pneumonia epidemic with higher requirements regarding diagnosis and treatment efficiency, hygiene and safety, and infection control.This study aims to identify how the Building Information Modeling (BIM) + Industrialized Building System (IBS) approach could rapidly deliver an infectious disease hospital and develop site epidemic spreading algorithms. Coronavirus-19 pneumonia construction site spreading algorithm model mind map and block diagram of the construction site epidemic spreading algorithm model were developed. BIM+IBS approach could maximize the repetition of reinforced components and reduce the number of particular components. Huoshenshan Hospital adopted IBS and BIM in the construction, which reduced the workload of on-site operations and avoided later rectification. BIM+IBS integrated information on building materials, building planning, building participants, and construction machinery, and realized construction visualization control and parametric design. The delivery of Huoshenshan Hospital was during the most critical period of the Coronavirus-19 pneumonia epidemic. The development of a construction site epidemic spreading algorithm provided theoretical and numerical support for prevention. The agent-based analysis on hospital evacuation observed "arched" congestion formed at the evacuation exit, indicating behavioral blindness caused by fear in emergencies.

        Anti-oxidative-initiated cognitive impairment amelioration in Alzheimer's disease model rats through preventive transcutaneous electrical acupoint stimulation

        Background: Alzheimer's disease (AD) is a chronic and irreversible neurodegenerative disease. Oxidative stress emerges at the early AD stage. As a non-invasive therapy with few adverse reactions, transcutaneous electrical acupoint stimulation (TEAS) combines acupuncture points of traditional Chinese medicine (TCM) and electrical stimulation. This study aimed to investigate the amelioration effects of preventive TEAS treatment (P-TEAS) on cognitive impairment and oxidative stress in AD model rats. Methods: The AD model was established via subcutaneous injections of D-galactose (D-gal, 120 mg/kg/d) into the back of neck for 9 weeks in Sprague Dawley (SD) rats to simulate the oxidative stress in the early AD stage. On the first day of the 10th week, Aβ1–42 (1 μg/μl) was injected into the CA1 regions of the bilateral hippocampus. P-TEAS was synchronized from the first day of subcutaneous D-gal injections for 9 weeks. Results: Empirical measurements showed that P-TEAS can improve the spatial memory ability of AD model rats in the Morris water maze. Superoxide dismutase (SOD) was upregulated in the P-TEAS group. Through the detection of the anti-oxidative stress signaling pathway, namely, Kelch-like ECH-associated protein 1 (Keap1)/ NFE2-related factor 2 (Nrf2), it was found that P-TEAS could promote Nrf2 entering into the nucleus and upregulating the production of protective factors heme oxygenase 1 (HO-1) and NADPH quinone oxidoreductase 1 (NQO1). It was also found that P-TEAS could downregulate the expressions of BCL2-associated X-protein (Bax), caspase 3, and caspase 9 to inhibit neuronal apoptosis. Conclusions: P-TEAS has similar efficacy to electroacupuncture in preventing AD occurrence and development. P-TEAS is a new non-invasive intervention therapy for the prevention of AD.

        Transcriptome analysis to characterize the genes related to gonad growth and fatty acid metabolism in the sea urchin Strongylocentrotus intermedius

        Background Sea urchin gonads of both sexes, commonly termed “roe”, are highly valued seafood delicacies, and Strongylocentrotus intermedius is considered one of the tastiest sea urchins. In order to produce high-quality gonads for consumption and clarify the mechanism of gonad growth and development of the sea urchin, more genetic information, especially at the transcriptome level, is needed. Objective A more thorough understanding of sea urchin gonad growth and development in both sexes could enable regulation of these processes at several stages with the aim of suppressing gametogenesis in order to produce high-quality gonads for consumption. Methods The adult sea urchins S. intermedius were cultured for 3 months, and were sampled for the gonadal transcriptome analysis which has been performed on the RNAs of three male and female adults of S. intermedius in each gonad development stage. Results Illumina sequencing raw sequence data was deposited in the NCBI Sequence Read Archive (SRA) database (PRJNA532998). It generated 560,196,356 raw reads and 548,956,944 clean reads were acquired, which were assembled into 107,850 transcripts with 44,124 genes. Comparative analysis showed the differentially expressed genes (DEGs) from 114 to 2566. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analyses were used to determine the functional significance of these DEGs. We have selected 9 genes related to growth and 12 genes related to fatty acid biosynthesis and metabolism in sea urchin gonads. Conclusion These data for sea urchins were intended to provide markers for gonad growth and development that can be accumulated for use in aquaculture applications.

        New Safrole Oxide Derivatives: Synthesis and in vitro Antiproliferative Activities on A549 Human Lung Cancer Cells

        A number of novel small molecules, safrole oxide derivatives 4a-c, 6a-c, 9a-h, were synthesized by the reaction of safrole oxide with anilines 3 and 5, or its alkyl allyl ether derivative 7 with alkyl bromide 8 in moderate yields. The antiproliferative effects of all the target molecules on A549 cell growth were investigated and it was found that the 14 novel compounds could suppress A549 lung cancer cell growth. Among them, compound 6b was the most effective compound in inhibiting the proliferation of A549 cells.

      • Detection of a new genetic locus for the high amylose content in rice mutant.

        Goami 2 is now well known as its extinguishing endosperm characteristics - it is far from the wild type, Ilpum, a premium taste Korean japonica cultivar. The endosperm of Goami 2 is high in fat, protein, and indigestible carbohydrate contents. One of the most extraordinary endosperm characteristic of Goami 2 is high level of amylose content, even though the hulled rice (brown rice) is totally opaque. There have been many studies to address the unique physico-chemical properties and possible usages as a healthy and functional food ingredient, especially, the high-amylose rice had a positive effect on lowering the blood glucose response in obesity and type 2 diabetes. Genetic analysis by using 44 SSR markers, crude linkage map (3~5 anchor markers per chromosome) was then constructed based on the genotypes detected among 112 F2 progenies derived from Goami 2 / Milyang 23 showed that major chromosomal regions on Chromosome 2 responsible for the variation of amylose contents. M2-53 on Chromosome 2 explains the highest variation and this region has not been reported as a putative QTL for amylose contents yet. More closely markers for application to breeding program can be developed using MutMap or Re-sequence methods.
