<P>The skin sensitizer 2,4-dinitrofluorobenzene (DNFB) provokes delayed hypersensitivity responses as a result of topical application to the skin. Here, we demonstrate that DNFB modifies proteins in RAW 264.7 cells and skin tissues in NC/Nga mice; we also show the functional involvement of DNFB-induced modification of cellular proteins in the DNFB-induced macrophage inflammatory protein (MIP)-2 gene expression in RAW 264.7 cells. In addition, we demonstrate that DNFB strongly induces reactive oxygen species (ROS) production. Our RT-PCR analysis and reporter gene assays reveal that the DNFB-induced intracellular ROS production is necessary for MIP-2 gene expression by DNFB. We observed that the vitamin C and chemical oxidant scavenger N-acetyl-cysteine have an inhibitory effect on the generation of ROS, the activation of MAP kinase pathways, and the MIP-2 gene expression in DNFB-treated RAW 264.7 cells. These results provide insight into the mechanisms involved in DNFB-induced contact hypersensitivity.</P>
Finite element analysis for temperature distributions in a cold forging

In this research, the finite element method is utilized to predict the temperature distributions in a cold-forging process for a cambolt. The cambolt is mainly used as a part of a suspension system of a vehicle. The cambolt has an off-centered lobe that manipulates the vertical position of the knuckle and wheel to a slight degree. The cambolt requires certain mechanical properties, such as strength and endurance limits. Moreover, temperature is also an important factor to realize mass production and improve efficiency. However, direct measurement of temperature in a forging process is infeasible with existing technology; therefore, there is a critical need for a new technique. Accordingly, in this study, a thermo-coupled finite element method is developed for predicting the temperature distribution. The rate of energy conversion to heat for the workpiece material is determined, and the temperature distribution is analyzed throughout the forging process for a cambolt. The temperatures associated with different punch speeds are also studied, as well as the relationships between load,temperature, and punch speed. Experimental verification of the technique is presented.

CpG-DNA has potent immunostimulatory effects to induce expression of cytokines and chemokines in macrophages, dendritic cells, and B cells. KG-1 is a human myeloid leukemia cell line used as an in vitro model system for dendritic cell differentiation. Here, we investigated effect of CpG-DNA on IFN-${\gamma}$ expression in KG-1 cells. Based on the mRNA and protein expression, stimulation of KG-1 cells with the phosphorothioated form of CpG-DNA induced expression of IFN-${\gamma}$. The IFN-${\gamma}$ promoter includes one NF-${\kappa}B$ binding site and two AP-1 binding sites. Induction of IFN-${\gamma}$ was markedly reduced in the presence of NF-${\kappa}B$, p38, and MEK1 inhibitors. Furthermore, AP-1 was found to interact with both of the two binding sites in the IFN-${\gamma}$ promoter. Therefore, we conclude that CpG-DNA induced activation of NF-${\kappa}B$ and AP-1 leads to induction of IFN-${\gamma}$, which may further contribute to immunostimulatory activity of CpG-DNA.
Expression of UNC93A Induced by CpG-DNA-liposome Complex in Mice

UNC93B1 is involved in the delivery of nucleotide-sensing Toll-like receptors (TLR) including TLR9 to endolysosomes. However, possible functions of UNC93A, another member of the UNC-93 superfamily, have not been studied in the context of TLR signaling. Here, we injected naked CpG-ODNs or liposome-encapsulated CpG-ODN (Lipoplex(O)) into the BALB/c mouse peritoneal cavity and investigated expression of mouse UNC93B1 and UNC93A genes in the peritoneal cells. UNC93A mRNA expression was increased by Lipoplex(O) in a time-dependent manner, whereas the expression level of UNC93B1 was not changed. To evaluate whether the expression of UNC93A involves TLR9, TLR9 knock out (TLR-/-) mice were injected with Lipoplex(O) or LipoplexGC(O), and the peritoneal cells were analyzed. The expression of UNC93A was not induced by Lipoplex(O) in TLR9-/- mice. These results suggest that UNC93A is closely associated with TLR9 signaling induced by Lipoplex(O) in the peritoneal cells in vivo. Primary cells including peritoneal cells stimulated with CpG-ODNs, Lipoplex(O), and several cytokines in vitro were prepared. However, UNC93A expression was not induced by any stimulation. To identify cellular localization of UNC93A, human embryonic kidney 293 cells stably expressing UNC93A were established and analyzed by confocal microscopy. The human and mouse UNC93A proteins were detected in the cytoplasm. Further investigation of the UNC93A function related with the CpG-DNA-mediated immune response may provide information to support efficient application of CpG-DNA immunotherapeutics.

The drug repurposing strategy has been applied to the development of emergency COVID-19 therapeutic medicines. Current drug repurposing approaches have been directed against RNA polymerases and viral proteases. Recently, we found that the inhibition of the interaction between the SARS-CoV-2 structural nucleocapsid (N) and spike (S) proteins decreased viral replication. In this study, drug repurposing candidates were screened by in silico molecular docking simulation with the SARS-CoV-2 structural N protein. In the ChEMBL database, 1994 FDA-approved drugs were selected for the in silico virtual screening against the N terminal domain (NTD) of the SARS-CoV-2 N protein. The tyrosine 109 residue in the NTD of the N protein was used as the center of the ligand binding grid for the docking simulation. In plaque forming assays performed with SARS-CoV-2 infected Vero E6 cells, atovaquone, abiraterone acetate, and digoxin exhibited a tendency to reduce the size of the viral plagues without affecting the plaque numbers. Abiraterone acetate significantly decreased the accumulation of viral particles in the cell culture supernatants in a concentration-dependent manner. In addition, abiraterone acetate significantly decreased the production of N protein and S protein in the SARS-CoV-2-infected Vero E6 cells. In conclusion, abiraterone acetate has therapeutic potential to inhibit the viral replication of SARS-CoV-2.
MUC1-C influences cell survival in lung adenocarcinoma Calu-3 cells after SARS-CoV-2 infection

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) induces coronavirus disease 2019 (COVID-19) and may increase the risk of adverse outcomes in lung cancer patients. In this study, we investigated the expression and function of mucin 1 (MUC1) after SARS-CoV-2 infection in the lung epithelial cancer cell line Calu-3. MUC1 is a major constituent of the mucus layer in the respiratory tract and contributes to pathogen defense. SARS-CoV-2 infection induced MUC1 C-terminal subunit (MUC1-C) expression in a STAT3 activation-dependent manner. Inhibition of MUC1-C signaling increased apoptosis-related protein levels and reduced proliferation-related protein levels; however, SARS-CoV-2 replication was not affected. Together, these results suggest that increased MUC1-C expression in response to SARS-CoV-2 infection may trigger the growth of lung cancer cells, and COVID-19 may be a risk factor for lung cancer patients. Purpose: The increasing use of electric personal mobility devices (ePMDs) has been accompanied by an increasing incidence of associated accidents. This study aimed to investigate the characteristics of ePMD-related injuries and their associated factors. Methods: This cross-sectional study was conducted using data from the Emergency Department-based Injury In-depth Surveillance database from 2014 to 2018. All patients who were injured while operating an ePMD were eligible. The primary outcome was the rate of severe injury, defined as an excess mortality ratio-adjusted Injury Severity Score of ≥25. We calculated the adjusted odds ratios (AORs) of outcomes associated with ePMD-related injuries. Results: Of 1,391,980 injured patients, 684 (0.05%) were eligible for inclusion in this study. Their median age was 28 years old, and most injuries were sustained by men (68.0%). The rate of ePMD-related injuries increased from 3.1 injuries per 100,000 population in 2014 to 100.3 per 100,000 population in 2018. A majority of the injuries occurred on the street (32.7%). The most commonly injured area was the head and face (49.6%), and the most common diagnosis was superficial injuries or contusions (32.9%). Being aged 55 years or older (AOR, 3.88; 95% confidence interval, 1.33-11.36) and operating an ePMD while intoxicated (AOR, 2.78; 95% confidence interval, 1.52-5.08) were associated with severe injuries. Conclusions: The number of emergency room visits due to ePMD-related injuries is increasing. Old age and drunk driving are both associated with serious injuries. Active traffic enforcement and safety regulations regarding ePMDs should be implemented to prevent severe injuries caused by ePMD-related accidents.

인터넷상에서의 정보검색은 검색엔진을 이용하여 이루어지는데, 방대한 사이트들을 검색하여야 하므로 검색효율이나 검색된 정보의 유용성에 문제가 있게 된다. 만약 이러한 정보들을 미리 자동적으로 검색, 분류해서 저장한다면 위의 두 가지 문제들을 해결할 수 있을 것이다. 자동적으로 이런 일을 처리하도록 고안된 것이 웹 로봇 에이전트라고 하며 현재 국내에도 여러 개의 웹 로봇 에이전트를 이용한 검색엔진이 사용되고 있다. 본 논문에서는 검색엔진을 구현하기 위해 하이퍼텍스트 전송규약에 대한 연구와 웹 로봇 에이전트에 대한 연구를 하여 올바른 로봇 에이전트를 구현하여, 구현된 검색엔진을 통한 효율적인 정보검색을 실현하는데 목적이 있다.
