<P>Pulmonary neuroendocrine tumors, especially small cell carcinoma and typical carcinoids, are the most common cause of ectopic Cushing syndrome (CS). Sometimes these adrenocorticotropic hormone (ACTH)-producing tumors are too small to localize, and it may take a long time to identify them. This report describes the case of a 27-year-old-man with CS. This CS was due to the ectopic ACTH produced by the pulmonary atypical carcinoids, which were recognized at the disseminated stage 5 years after the initial presentation of CS. Microscopically, multiple whitish nodules (up to 1.0 cm in diameter) of the wedge-resected lung were composed of small round cells appearing to be typical neuroendocrine but were diagnosed as atypical carcinoid, based on the findings of focal necrosis, high mitotic rates and multiple endolymphatic tumor emboli. Immunohistochemically, tumor cells robustly stained for ACTH. The delay of diagnosis is, in part, attributable to the pulmonary tuberculosis (one of the complications of hypercortisolism), because the pulmonary tuberculosis presented in the form of multiple nodules throughout the lung. In addition, this patient had a Crooke's cell hyperplasia of the pituitary gland, which represents the reactive change of corticotrophs exposed to the excess cortisol rather than neoplastic change leading to CS.</P>
Expression Profile of <i>LGR5</i> and Its Prognostic Significance in Colorectal Cancer Progression

<P>We investigated the expression profile of leucine-rich, repeat-containing, G-protein-coupled receptor 5 (LGR5) during colorectal cancer (CRC) progression and determined the prognostic impact of LGR5 in a large cohort of CRC samples. LGR5 expression was higher in CRCs than in normal mucosa, and was not associated with other cancer stem cell markers. LGR5 positivity was observed in 68% of 788 CRCs and was positively correlated with older age, moderately to well-differentiated cells, and nuclear D-catenin expression. Enhanced LGR5 expression remained persistent during the adenoma-carcinoma transition, but markedly declined in the budding cancer cells at the invasive fronts, which was not due to altered wingless-type mouse mammary tumor virus integration site family (Wnt) or epithelial-mesenchymal transition signaling. LGR5 showed negative correlations with microsatellite instability and CpG island methylator phenotype, and was not associated with KRAS or BRAF mutation. Notably, LGR5 positivity was an independent prognostic marker for better clinical outcomes in CRC patients. LGR5 overexpression attenuated tumor growth by decreasing ERK phosphorylation along with decreased colony formation and migration abilities in DLD1 cells. Likewise, knockdown of LGR5 expression resulted in a decline in the colony-forming and migration capacities in LoVo cells. Taken together, our data suggest a suppressive role of LGR5 in CRC progression.</P>

다양한 식용식물에 함유되어 있는 것으로 알려진 phenolic acids의 일종인 p-coumaric acid의 항암활성을 규명하고자, 인체 급성백혈병 T 세포주인 Jurkat T 세포에 대한 p-coumaric acid의 에폽토시스 유도기전을 조사하였다. Jurkat T 세포를 p-coumaric acid (50-150 μM)로 처리한 결과, 세포독성, 에폽토시스-관련 DNA fragmentation, 및 pro-apoptotic multidomain Bcl-2 family member인 Bak의 활성화, Δψm loss, caspase-9, -3, -7, 및 -8의활성화, 그리고 PARP 분해 등의 여러 에폽토시스-관련 생화학적 현상들이 농도의존적으로 나타났다. 그러나 이러한 에폽토시스-관련 생화학적 현상들은 Jurkat T 세포에 anti-apoptotic Bcl-2 단백질을 과발현할 경우에는 나타나지 않았다. 또한 p-coumaric acid처리에 의해 유도되는 Jurkat T 세포의 에폽토시스에는 necrosis가 수반되지 않는 것으로 확인되었다. Jurkat T 세포를 pan-caspase inhibitor인 z-VAD-fmk를 전처리할 경우, p-coumaric acid 처리에 의해 유도되는 apoptotic sub-G₁ peak는 차단되어 나타나지 않았으나 Δψm loss는 여전히 나타났는데, 이는 p-coumaric acid처리에 의한 에폽토시스의 유도에 caspase cascade 활성화가 필수적이며Δψm loss의 downstream 현상임을 나타낸다. 한편, FADD 및caspase-8을 함께 발현하는 Jurkat T 세포주 A3, FADD-결손 Jurkat T 세포주 I2.1, 그리고 caspase-8-결손 Jurkat T 세포주 I9.2의 p-coumaric acid의 세포독성에 대한 감수성은 서로 유사하게 나타났는데, 이는 p-coumaric acid처리에 의한 에폽토시스의 유도가 Fas와 FasL간의 상호작용에 의해 개시되지 않음을 시사한다. p-Coumaric acid의 세포독성은 Jurkat T 세포에 비해 인체 정상 말초혈액 T 세포에서 훨씬 낮게 나타났다. 이러한 결과들은 p-coumaric acid 처리에 의해 유도되는 Jurkat T 세포의 에폽토시스가 Bak 활성화, Δψm loss, caspase-9, -3, -7, 및 -8로 이루어진 caspase cascade의 활성화, 그리고 PARP 분해에 의해 유도되며, 또한 anti-apoptotic 단백질인 Bcl-2의 과발현에 의해서 음성적으로 조절됨을 나타낸다. In this study, we investigated the anti-inflammation effect of Persicaria thunbergii (P. thunbergii) on RAW 264.7 murine macrophage cells. The anti-inflammatory activity of P. thunbergii was determined by measuring expression of the LPS-induced inflammatory proteins, inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2) and nuclear factor-κB (NF-κB), and the production of nitric oxide (NO) and prostaglandin E₂ (PGE₂). Methanol extract of P. thunbergii decreased the expression of iNOS, COX-2 and NF-κB, and increased the expression of HO-1 in LPS-stimulated RAW264.7 cells. Methanol extract was fractioned by n-butanol, hexane and ethyl acetate (EtOAc) and each fraction was tested for inhibitory effects on inflammation. Among the sequential solvent fractions, the EtOAc soluble fraction was investigated by the expression of prostaglandin E2 (PGE2), and showed decreasing form to the dose-dependent manner. EtOAc extract showed the most effective inhibitory activity of the expression of iNOS, COX-2 and NF-κB, and the production of NO. The study showed that P. thunbergii has anti-inflammatory activity through the decrease of NO and inhibition of iNOS, COX-2, PGE₂ and NF-κB expression, and by the increase of HO-1 enzyme. This study needs for more investigation to find out the most effective single compound with anti-inflammatory activity.
직렬 배치된 두 부유체에 작용하는 조류력 및 풍력 특성에 관한 수치해석 연구

In this study, the characteristics of the current and wind forces acting on two floating bodies were numerically investigated using a commercial CFD software, STAR-CCM+. In the numerical analyses, LNGC was located right behind FSRU under uniform current or wind conditions. Steady calculations were carried out using a Reynolds averaged Navier?Stokes (RANS) solver and the realized k-epsilon model. First, the current coefficients of FSRU based only the CFD were compared with the model test data. Through this comparison, the present numerical models and mesh systems were indirectly verified. Next, computations for FSRU and LNGC in a uniform current were performed using different relative positions. It was found that the current coefficients were great affected by the longitudinal positions. Finally, the wind forces acting on FSRU and LNGC in tandem configurations were studied. The focus was on the shielding effects due to the aerodynamic interactions between FSRU and LNGC.

FPSO model tests of mooring line failure were carried out in the ocean basin at KRISO. The characteristics of the motions and mooring lines were investigated. The FPSO ship was moored using an internal turret and catenary mooring. The test model was 1/60 scale. The mooring lines were designed to satisfy the characteristics of the original mooring lines using the truncation method. The experiments were conducted under various environments: a safe wave, current and wind condition; single broken mooring line condition; and transient mode condition. The moment of the break was determined based on experimental test results. The results showed that the FPSO behavior and mooring line tensions were acceptable under the failure condition.

The excessive yaw motion of turret moored vessel is investigated. The floating body is moored by internal turret and catenary mooring. The experiments are conducted in regular waves. Turret moored vessel can oscillate side to side or move to one direction, large yaw motion. The results shows that the excessive yaw occurs for particular condition, ratio of wave lengths and wave period. The time series, comparison of moments and time domain analysis are performed. The analysis shows that force and ratio of wave lengths and vessel length influence the excessive yaw motion. The static analysis and time domain analysis results indicate that wave drift moment is the main parameter of excessive yaw motion.

To improve the mechanical properties of hydroxyapatite (HA)/waterborne polyurethane (WBPU) composites, the hydroxyl group of HA was modified by urethane reactions: the hydroxyl groups of HA were reacted with aliphatic or cyclic diisocyanate, and then the modified HAs were extended by adding polyol and/or ε-caprolactone. Composites were prepared by the prepolymer process method: the modified HA was directly pured into the urethane reaction of isocyanate and polyol. The properties of modified HA/WBPU composites were investigated by thermogravimetric analysis, tensile strength, and water resistance. The results showed that the reactivity of aliphatic diisocyanate to the hydroxy group of HA was faster than that of cyclic one. Comparing to those of pure HA/WBPU composite films, the thermal stability, water resistance, and mechanical properties of the modified composite films increased with a degree of modification of HA.
