        Super-resolution fluorescent materials: an insight into design and bioimaging applications

        <P>Living organisms are generally composed of complex cellular processes which persist only within their native environments. To enhance our understanding of the biological processes lying within complex milieus, various techniques have been developed. Specifically, the emergence of super-resolution microscopy has generated a renaissance in cell biology by redefining the existing dogma towards nanoscale cell dynamics, single synaptic vesicles, and other complex bioprocesses by overcoming the diffraction-imposed resolution barrier that is associated with conventional microscopy techniques. Besides the typical technical reliance on the optical framework and computational algorithm, super-resolution imaging microscopy resorts largely to fluorescent materials with special photophysical properties, including fluorescent proteins, organic fluorophores and nanomaterials. In this tutorial review article, with the emphasis on cell biology, we summarize the recent developments in fluorescent materials being utilized in various super-resolution techniques with successful integration into bio-imaging applications. Fluorescent proteins (FP) applied in super-resolution microscopy will not be covered herein as it has already been well summarized; additionally, we demonstrate the breadth of opportunities offered from a future perspective.</P>

        Risk factors for cancer-specific survival in elderly gastric cancer patients after curative gastrectomy

        BACKGROUND/OBJECTIVES Previous studies have reported that protein supplementation contributes to the attenuation of inflammation. Serious trauma such as burn injury usually results in the excessive release of inflammatory factors and organs dysfunction. However, a few reports continued to focus on the function of protein ingestion in regulating burn-induced inflammation and organ dysfunction. MATERIALS/METHODS This study established the rat model of 30% total body surface area burn injury, and evaluated the function of blended protein (mixture of whey and soybean proteins). Blood routine examination, inflammatory factors, blood biochemistry, and immunohistochemical assays were employed to analyze the samples from different treatment groups. RESULTS Our results indicated a decrease in the numbers of white blood cells, monocytes, and neutrophils in the burn injury group administered with the blended protein nutritional support (Burn+BP), as compared to the burn injury group administered normal saline supplementation (Burn+S). Expressions of the pro-inflammatory factors (tumor necrosis factor-α and interleukin-6 [IL-6]) and chemokines (macrophage chemoattractant protein-1, regulated upon activation normal T cell expressed and secreted factor, and C-C motif chemokine 11) were dramatically decreased, whereas anti-inflammatory factors (IL-4, IL-10, and IL-13) were significantly increased in the Burn+BP group. Kidney function related markers blood urea nitrogen and serum creatinine, and the liver function related markers alanine transaminase, aspartate aminotransferase, alkaline phosphatase, and lactate dehydrogenase were remarkably reduced, whereas albumin levels were elevated in the Burn+BP group as compared to levels obtained in the Burn+S group. Furthermore, inflammatory cells infiltration of the kidney and liver was also attenuated after burn injury administered with blended protein supplementation. CONCLUSIONS In summary, nutritional support with blended proteins dramatically attenuates the burn-induced inflammatory reaction and protects organ functions. We believe this is a new insight into a potential therapeutic strategy for nutritional support of burn patients.

        A New Constraint Handling Method for Economic Dispatch

        For practical consideration, economic dispatch (ED) problems in power system have non-smooth cost functions with equality and inequality constraints that makes the problems complex constrained nonlinear optimization problems. This paper proposes a new constraint handling method for equality and inequality constraints which is employed to solve ED problems, where the incremental rate is employed to enhance the modification process. In order to prove the applicability of the proposed method, the study cases are tested based on the classical particle swarm optimization (PSO) and differential evolution (DE) algorithm. The proposed method is evaluated for ED problems using six different test systems: 6-, 15-, 20-, 38-, 110- and 140-generators system. Simulation results show that it can always find the satisfactory solutions while satisfying the constraints.

      • Combination of LLT Model and TV Model for Image Denoising

        Developing a variational model that is capable of restoring both smooth (no edges) and non-smooth (with edges) images is still a valid challenge at the image processing. In this paper, we present two methods for image denoising problems based on the use of the LLT model (see [14]) and TV model (see [20]). The idea of our methods is, add the texture which is separated from the cartoon and noisy, back to the original noisy image or the texture plus noisy part, and the sum then processed. In order to obtain the texture, we first separate texture plus noise from cartoon by LLT model, and then use TV model to remove some noisy from texture. Numerical experiments show our method is able to maintain some important information such as small details in the image, and at the same time to get a better visualization.

        Design, Modeling, Control and Experiment for a 2-DOF Compliant Micro-Motion Stage

        A new XY micro-motion stage is proposed with the double four-bar prismatic joints to transfer linear motions and mechanicaldisplacement amplifier. The compliance models of the amplifier, the prismatic joints, and the whole stage are established based onthe flexibility matrix method. The simulation is made by using finite element analysis ANSYS software. The output cross-talk is about2% and the parasitic motions of the input points in the other limbs are less than 1.8%, which shows a good decoupling property. Themechanical prototype is fabricated, the experimental results show that the input/output of the stage has a very good linearity, the ratioof output displacement to input displacement is 5.06 and the working range is 156 μm × 156 μm. To tackle for the serious hysteresisnonlinear problems of the stage, model reference adaptive PID controller is designed, the final resolution can reach ±0.2 μm.

        Important Role of Immunological Responses to Environmental Exposure in the Development of Allergic Asthma

        Allergic asthma is a public health problem that affects human health and socioeconomic development. Studies have found that the prevalence of asthma has significantly increased in recent years, which has become particularly pronounced in developed countries. With rapid urbanization in China in the last 3 decades, the prevalence of asthma has increased significantly in urban areas. As changes in genetic backgrounds of human populations are limited, environmental exposure may be a major factor that is responsible for the increased prevalence of asthma. This review focuses on environmental components of farms and rural areas that may have protective effects in reducing the development of asthma. Farm and rural related microorganism- and pathogen-associated molecular patterns are considered to be important environmental factors that modulate host's innate and adaptive immune system to induce protection effects later in life. Environmental microbial-related immunotherapy will also be discussed as the future research direction for the prevention of allergic asthma.

      • <i>OsASR5</i> enhances drought tolerance through a stomatal closure pathway associated with ABA and H <sub>2</sub> O <sub>2</sub> signalling in rice

        <P><B>Summary</B></P><P>Drought is one of the major abiotic stresses that directly implicate plant growth and crop productivity. Although many genes in response to drought stress have been identified, genetic improvement to drought resistance especially in food crops is showing relatively slow progress worldwide. Here, we reported the isolation of <I>abscisic acid</I>,<I> stress</I> and <I>ripening</I> (<I>ASR</I>) genes from upland rice variety, IRAT109 (<I>Oryza sativa</I> L. ssp. <I>japonica</I>), and demonstrated that overexpression of <I>OsASR5</I> enhanced osmotic tolerance in <I>Escherichia coli</I> and drought tolerance in <I>Arabidopsis</I> and rice by regulating leaf water status under drought stress conditions. Moreover, overexpression of <I>OsASR5</I> in rice increased endogenous ABA level and showed hypersensitive to exogenous ABA treatment at both germination and postgermination stages. The production of H<SUB>2</SUB>O<SUB>2</SUB>, a second messenger for the induction of stomatal closure in response to ABA, was activated in overexpression plants under drought stress conditions, consequently, increased stomatal closure and decreased stomatal conductance. In contrast, the loss‐of‐function mutant, <I>osasr5</I>, showed sensitivity to drought stress with lower relative water content under drought stress conditions. Further studies demonstrated that OsASR5 functioned as chaperone‐like protein and interacted with stress‐related HSP40 and 2OG‐Fe (II) oxygenase domain containing proteins in yeast and plants. Taken together, we suggest that <I>OsASR5</I> plays multiple roles in response to drought stress by regulating ABA biosynthesis, promoting stomatal closure, as well as acting as chaperone‐like protein that possibly prevents drought stress‐related proteins from inactivation.</P>

        Genetic dissection of leaf-related traits using 156 chromosomal segment substitution lines

        A two-line super-hybrid rice (Oryza sativa L.) variety [Liangyoupei9 (LYP9)] demonstrated superiority over its both parents, viz. elite inbred lines 93-11 and Pei-ai64S (PA64S), as well as other conventional hybrids, and had long been exploited in China. However, the genetic basis of its leaf-related traits, supposed to be an important component for yield potential, remains elusive. Here, initially a set of chromosome segment substitution lines (CSSLs) was constructed, in which the genome of Pei-ai64S has been introgressed into the background of 93-11. This set was developed by marker aided selection, based on 123 polymorphic SSR markers. The introgressed chromosomal segments presented in the 156 CSSLs covered 96.46% of Pei-ai64S genome. Afterwards, the CSSLs were deployed to assess the genetic basis of leaf size (length and width) and chlorophyll content of top three leaves across five different environments. The CSSLs showed transgressive segregation for all of the traits, and significant correlations were detected among most of the traits. A total of 27 quantitative trait loci (QTL) were identified on ten chromosomes, and three QTL cluster affecting related traits were found on chromosome 3, 6, and 8, respectively. Remarkably, two key QTLs, qALW3-1 and qALW3-2, both controlling the antepenultimate leaf width, were identified in all five environments, and their effect were further validated by CSSLs harboring the two QTL alleles. Our results indicate that developing CSSLs is a powerful tool for genetic dissection of quantitative traits. Meanwhile, the QTLs controlling leaf-related traits uncovered here provide useful information for marker-assisted selection in improving the performance of leaf morphology and photosynthetic ability.

        Analysis of high speed bearing based on virtual rods model

        The traditional high-speed ball bearing analysis is based on the quasi-static model proposed by Jones-Harris (J-H). This study gives another bearing analysis model based on the relative position of the ball center and the centers of curvature of the raceway grooves, described by the virtual rods. Specifically, the load and deformation of the virtual rods are determined according to the principle of force transmissibility, the translation theorem of a force, and elastic-contact deformation between steel ball bearings and raceway grooves. And the iterative variables such as the axial, radial and angular deformations are gradually introduced into the calculation model to solve the force and kinematic parameters under pure axial load, bidirectional and tri-directional combined loads. The correctness of the virtual rods model is verified by the contact angle for axial and bidirectional load. The influence of gyroscopic moment and raceway grooves control parameter on the actual contact angle under tridirectional loading conditions are calculated and analyzed respectively. The proposed model uses fewer equations and lower-dimension parameters than those used by the traditional highspeed ball bearing analysis method, improves the solution rate of the equations.
