      • Pituitary Adenoma Biomarkers Identified Using Proteomic Fingerprint Technology

        Objective: To determine whether pituitary adenomas can be diagnosed by identifying protein biomarkers in the serum. Methods: We compared serum proteins from 65 pituitary adenoma patients and 90 healthy donors using proteomic fingerprint technology combining magnetic beads with matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MS). Results: A total of 42 M/Z peaks were identified as related to pituitary adenoma (P<0.01). A diagnostic model established based on three biomarkers (3382.0, 4601.9, 9191.2) showed that the sensitivity of diagnosing pituitary adenoma was 90.0% and the specificity was 88.3%. The model was further tested by blind analysis showing that the sensitivity was 88.0% and the specificity was 83.3%. Conclusions: These results suggest that proteomic fingerprint technology can be used to identify pituitary adenoma biomarkers and the model based on three biomarkers (3382.0, 4601.9, 9191.2) provides a powerful and reliable method for diagnosing pituitary adenoma.

      • Association of GSTP1 and RRM1 Polymorphisms with the Response and Toxicity of Gemcitabine-cisplatin Combination Chemotherapy in Chinese Patients with Non-small Cell Lung Cancer

        Background: Previous studies showed that genetic polymorphisms of glutathione S-transferase P1 (GSTP1) were involved in glutathione metabolism and genetic polymorphisms of ribonucleotide reductase (RRM1) were correlated with DNA synthesis. Here we explored the effects of these polymorphisms on the chemosensitivity and clinical outcome in Chinese non-small cell lung cancer (NSCLC) patients treated with gemcitabine-cisplatin regimens. Materials and Methods: DNA sequencing was used to evaluate genetic polymorphisms of GSTP1 Ile105Val and RRM1 C37A-T524C in 47 NSCLC patients treated with gemcitabine-cisplatin regimens. Clinical response was evaluated according to RECIST criteria after 2 cycles of chemotherapy and toxicity was assessed by 1979 WHO criteria (acute and subacute toxicity graduation criteria in chemotherapeutic agents). Results: There was no statistical significance between sensitive and non-sensitive groups regarding the genotype frequency distribution of GSTP1 Ile105Val polymorphism (p>0.05). But for RRM1 C37A-T524C genotype, sensitive group had higher proportion of high effective genotype than non-sensitive group (p=0.009). And according to the joint detection of GSTP1 Ile105Val and RRM1 C37A-T524C polymorphisms, the proportion of type A (A/A + high effective genotype) was significantly higher in sensitive group than in non-sensitive group (p=0.009). Toxicity showed no correlation with the genotypes between two groups (p>0.05). Conclusions: Compared with single detection of genetic polymorphisms of GSTP1 Ile105Val or RRM1 C37A-T524C, joint detection of both may be more helpful for patients with NSCLC to receive gemcitabine-cisplatin regimens as the first-line chemotherapy. Especially, genetic polymorphism of RRM1 is more likely to be used as an important biomarker to predict the response and toxicity of gemcitabine-cisplatin combination chemotherapy in NSCLC.

        Clinical and Pathologic Features of Multifocal and Multicentric Breast Cancer in Chinese Women: A Retrospective Cohort Study

        Purpose: This study aims to analyze the clinical-pathological characteristics of multifocal and multicentric breast cancer (MMBC) in Chinese women. Methods: Sixty-seven cases with MMBC were randomly collected and reviewed at seven hospitals in representative districts of China during 1999 to 2008. Results: The incidence of MMBC in breast cancer in China was 1.75%. Compared to those with unifocal breast cancer, women with MMBC were more likely to have larger tumor size, lymph node metastasis (59.70% vs. 45.62%) and stage III to IV (46.26% vs. 21.10%). The peak age at onset of MMBC was 40 to 49 years old and has been gradually increasing during 1999 to 2008. Most of the MMBC women were treated with surgery and adjuvant therapy. Conclusion: In China, the incidence of MMBC in breast cancer is significantly lower than that in Western countries. Compared to unifocal breast cancer, MMBC is biologically more aggressive. Most MMBC women underwent mastectomy, instead of breast conservation surgery.

        Prestressed concrete bridges with corrugated steel webs: Nonlinear analysis and experimental investigation

        Concrete bridges with corrugated steel webs and prestressed by both internal and external tendons have emerged as one of the promising bridge forms. In view of the different behaviour of components and the large shear deformation of webs with negligible flexural stiffness, the assumption that plane sections remain plane may no longer be valid, and therefore the classical Euler-Bernoulli and Timoshenko beam models may not be applicable. In the design of this type of bridges, both the ultimate load and ductility should be examined, which requires the estimation of full-range behaviour. An analytical sandwich beam model and its corresponding beam finite element model for geometric and material nonlinear analysis are developed for this type of bridges considering the diaphragm effects. Different rotations are assigned to the flanges and corrugated steel webs to describe the displacements. The model accounts for the interaction between the axial and flexural deformations of the beam, and uses the actual stress-strain curves of materials considering their stress path-dependence. With a nonlinear kinematical theory, complete description of the nonlinear interaction between the external tendons and the beam is obtained. The numerical model proposed is verified by experiments.

      • Degree of hydration-based thermal stress analysis of large-size CFST incorporating creep

        With the span and arch rib size of concrete-filled steel tube (CFST) arch bridges increase, the hydration heat of pumped mass concrete inside large-size steel tube causes a significant temperature variation, leading to a risk of thermal stressinduced cracking during construction. In order to tackle this phenomenon, a hydration heat conduction model based on hydration degree was established through a nonlinear temperature analysis incorporating an exothermic hydration process to obtain the temperature field of large-size CFST. Subsequently, based on the evolution of elastic modulus based on hydration degree and early-age creep rectification, the finite element model (FEM) model and analytical study were respectively adopted to investigate the variation of the thermal stress of CFST during hydration heat release, and reasonable agreement between the results of two methods is found. Finally, a comparative study of the thermal stress with and without considering early-age creep was conducted.

        Genetic variations in the bitter taste receptor gene TAS2R38 are related to cigarette smoking behavior in Han Chinese smokers

        Background: Smoking behavior is influenced by multiple genes, including the bitter taste gene TAS2R38. It has been reported that the correlation between TAS2R38 and smoking behavior has ethnicity-based differences. However, the TAS2R38 status in Chinese smokers is still unclear. Objective: This study aims to investigate the possible relationship between genetic variations in TAS2R38 (A49P, V262A and I296V) and smoking behaviors in the Han Chinese population. Methods: The haplotype analyses were performed and smoking behavior questionnaire was completed by 1271 individuals. Genetic association analyses for smoking behavior were analyzed using chi-square test. Further, for investigating the molecular mechanism of TAS2R38 variants effect on smoking behavior, we conducted TAS2R38-PAV and TAS2R38-AVI expression plasmids and tested the cellular calcium assay by cigarette smoke compounds stimulus in HEK293. Results: Significant associations of genetic variants within TAS2R38 were identified with smoking behavior. We found a higher PAV/PAV frequency than AVI/AVI in moderate and high nicotine dependence (FTND ≥ 4; X2 = 4.611, 1 df, p = 0.032) and strong cigarette smoke flavor intensity preference (X2 = 4.5383, 1 df, p = 0.033) in participants. Furthermore, in the in vitro cellular calcium assay, total particle matter (TPM), N-formylnornicotine and cotinine, existing in cigarette smoke, activated TAS2R38-PAV but not TAS2R38-AVI-transfected cells. Conclusion: Our data highlights that genetic variations in TAS2R38 are related to smoking behavior, especially nicotine dependence and cigarette smoke flavor intensity preference. Our findings may encourage further consideration of the taste process to identify individuals susceptible to nicotine dependence, particularly Han Chinese smokers.

        A Stability Study of the Longest Steel Truss Deck Cable-stayed Bridge during Construction

        For long-span cable-stayed bridges, the stability performance during construction is vital from the point view of structural safety. This paper presents an extensive study of stability performance during construction for the world longest steel truss deck cable-stayed bridge-Yachihe Bridge. Based on the finite element analysis, the Linear Elastic Stability (LES), Nonlinear Elastic Stability (NLES) and Nonlinear Inelastic Stability (NLIES) of the bridge have been investigated to identify the effects of geometry nonlinearity and material nonlinearity on the structural stability performance. In addition, the influences of temperature variations and static lateral winds on stability performance are also focused. The results show that: 1) Under design ambient temperature, the corresponding design of steel truss deck yields the minimum stability coefficients of LES, NLES and NLIES by 5.72, 4.64 and 2.96, respectively, corresponding to the completion of superimposed dead loads; 2) both the geometry nonlinearity and material nonlinearity have been identified to make negative effects on the stability performance and the material nonlinearity will reduce the structural stabilitycoefficient largely; 3) during construction, the rise and drop in temperature will increase and decrease the stability coefficients, respectively. NLIES is the most sensitive one to variations of temperature; and 4) the lateral static winds slightly reduce the stability coefficients of LES, NLES and NLIES.

        New Method for Analysis of Sliding Cable Structures in Bridge Engineering

        The Sliding Cable Structure (SCS) is widely used in bridge construction by cable crane method. The SCS generally consists of cables and fixed pulleys and the cables will slide around the pulleys when the cables are loaded until the balanced state is achieved. In this paper, an efficient Angle Bisector Method (ABM) is proposed for the mechanical analysis of SCS. The constraint effect of pulley on the cable is simulated by fixed hinge and the constraint force perpendicular to the angle bisector of cable elements adjacent to the pulley is therefore zero at balanced state. By utilizing “Unit Length Influence Matrix” (ULIM), the adjustment of unstressed length of each sub-cable can be determined. As the angle bisector at each pulley will change when the unstressed lengths of cables are changed, iterative procedures are necessary to find the convergence solutions. The proposed method is firstly verified by two examples and then applied to the analysis of cable crane during construction of the world’s longest steel truss cable-stayed bridge, namely Yachihe Bridge. Satisfactory numerical solutions can be obtained only with several iterations and are in good agreement with field measurements from the cable crane load tests, which confirms the accuracy and validity of the proposed method.

      • Aberrant DNA Methylation and Epigenetic Inactivation of hMSH2 Decrease Overall Survival of Acute Lymphoblastic Leukemia Patients via Modulating Cell Cycle and Apoptosis

        Objective: Altered regulation of many transcription factors has been shown to play important roles in the development of leukemia. hMSH2 can modulate the activity of some important transcription factors and is known to be a regulator of hematopoietic differentiation. Herein, we investigated epigenetic regulation of hMSH2 and its influence on cell growth and overall survival of acute lymphoblastic leukemia (ALL) patients. Methods: hMSH2 promoter methylation status was assessed by COBRA and pyrosequencing in 60 ALL patients and 30 healthy volunteers. mRNA and protein expression levels of hMSH2, PCNA, CyclinD1, Bcl-2 and Bax were determined by real time PCR and Western blotting, respectively. The influence of hMSH2 on cell proliferation and survival was assessed in transient and stable expression systems. Results: mRNA and protein expression of hMSH2 and Bcl-2 was decreased, and that of PCNA, CyclinD1 and Bax was increased in ALL patients as compared to healthy volunteers (P<0.05). hMSH2 was inactivated in ALL patients through promoter hypermethylation. Furthermore, hMSH2 hypermethylation was found in relapsed ALL patients (85.7% of all cases). The median survival of patients with hMSH2 methylation was shorter than that of patients without hMSH2 methylation (log-rank test, P=0.0035). Over-expression of hMSH2 in cell lines resulted in a significant reduction in growth and induction of apoptosis. Conclusions: This study suggests that aberrant DNA methylation and epigenetic inactivation of hMSH2 play an important role in the development of ALL through altering cell growth and survival.
