To obtain robust and thermo-stable enzyme aggregates, p-benzoquinone was used as cross-linker and bovine serum albumin (BSA) as crowding macromolecules to prepare cross-linked enzyme aggregates (CLEAs) of lipase. Effects of cross-linking time and cross-linker content on the activity, thermal stability and characteristics of enzyme aggregates were examined carefully. It was observed that when the content of p_benzoquinone was 5 mM and amount of BSA was 125% of that of lipase (w/w), the specific activity of cross-linked co-aggregates of lipase and BSA was 79.8 U mg^−1, 2.44-fold of that of cross-linked enzyme aggregates of lipase without BSA. Moreover, after heat treatment for 96 h at 50℃ , the CLEAs prepared with this facile routine kept 75.18% of their initial activity, 5.01-fold more than that of the just CLEAs using glutaraldehyde. Furthermore, BSA macromolecules in lipase CLEAs enhanced the catalytic efficiency of free and just lipase CLEAs without BSA by 1.45 and 2.83 times, respectively. The proposed crosslinking technique would rank among the potential strategies for efficiently preparing robust and thermo-stable enzyme aggregates.
High-Conductivity SiO2-Matrix B-Doped Si-NC Thin Films by Following Ion-Beam Treatment

In this work, further ion-beam was performed on SiO2-matrix B-doped Si-NC (SBC) thin films in order to enhance conductivity. The effect of ionbeamtype on the electrical properties of SBC thin films was investigatedsystematically. The results indicated that the conductivities of SBC thinfilms were significantly improved by both argon and hydrogen ion-beamtreatments, and the higher the hydrogen ion ratio, the higher theconductivity of SBC thin films. The conductivity of SBC thin films wasincreased from 1.82 × 10−6 S/cm to 3.2 × 10−3 S/cm with followinghydrogen-ion-beam treatment. The change in conductivity of SBC thinfilms was most possibly resultant from the ion-beam treatment facilitatingthe formation of higher superficial order and lower defects. An alternativemethod was proposed to prepare high-conductivity SBC thin films, whichmay be applied to other heterogeneous thin films.
Fatigue analysis of overlapped tubular joints

The increasing demand of on the use of wind energy during recent years, have led to installation of more durable, safe and stable offshore structures in deep waters. The circular hollow section tubular joints are generally preferred in offshore structures. The overlapped joints are considered better than simple gap K-joints because of their higher axial strength, high stiffness and high durability. These tubular joints being subjected to combination of wind and waves load during their design period, the constantly acting loads deteriorates the structure, which often results in total collapse of offshore structures. In order to allow for cost effective and reliable design, the substantial requirement is to do accurate and reliable simulation. In this study in order to understand the behavior of crack initiation and the fatigue life of overlapped joints, the 3D fatigue FEM analysis of overlapped joint was carried out. The 3D fatigue FEM study based on continuum damage mechanics and constitutive model, was carried out in two steps namely 3D thermo elastic-plastic analysis and 3D non-steady heat conduction analysis. The fatigue life and crack initiation positions were determined. Finally, S-N curves were plotted and the results from FEM analysis were compared with Hot Spot Stress and with S-N curves recommended by Eurocode 3.
Fatigue Analysis of Tripod wind turbine support structure

The offshore wind turbines in deep waters requires building large foundations, which transfers loads from wind turbine to ground. The tripod being considered the most robust and secure supporting structure due to its good strength and stability. The welded joints in the structure being exposed to different environmental loads such as wave load, ice load, wind load and earthquake for longer periods increases the stress concentration at hotspot positions which in turn leads to crack initiation and ultimately the failure of whole structure. In this work the main objective is to investigate the fatigue life and crack initiation positions of welded joint in Tripod structure by using 3D fatigue FEM analysis. Firstly, in 3-D Fem analysis, welding residual stresses and welding deformation is calculated by employing sequentially coupled 3-D thermo-mechanical analysis. Secondly, a nonlinear damage model based on continuum damage mechanics for multiaxial high cycle fatigue is used to predict the fatigue life and fatigue crack initiation. The results obtained from analysis were quite satisfying, so 3D fatigue FEM can predict the fatigue life of structures with great accuracy.

The extracellular inulinase of the marine yeast Pichia guilliermondii strain 1 was purified to homogeneity resulting in a 7.2-fold increase in specific inulinase activity. The molecular mass of the purified enzyme was estimated to be 50.0 kDa. The op-timal pH and temperature for the purified enzyme were 6.0 and 60C, respectively. The enzyme was activated by Mn²+, Ca²+, K+, Li+, Na+, Fe³+, Fe²+, Cu²+, and Co²+, but Mg²+, Hg²+, and Ag+ inhibited activity. The enzyme was strongly inhibited by phenylmethanesulphonyl fluoride (PMSF), iodoacetic acid, EDTA, and 1, 10-phenanthroline. The Km and Vmax values of the purified inulinase for inulin were 21.1 mg/mL and 0.08 mg/min, respectively. A large number of monosaccharides were de-tected after the hydrolysis of inulin. The deduced protein sequence from the cloned P. guilliermondii strain 1 inulinase gene contained the consensus motifs R-D-P-K-V-F-W-H and W-M-N-D-P-N-G, which are conserved among the inulinases from other microorganisms.
