<P><I>Clostridium difficile</I> is the major cause of hospital-acquired infectious diarrhea and colitis in developed countries. The pathogenicity of <I>C. difficile</I> is mainly mediated by the release of 2 large potent exotoxins, toxin A (TcdA) and toxin B (TcdB), both of which require neutralization to prevent disease occurrence. We have generated a novel chimeric protein, designated mTcd138, comprised of the glucosyltransferase and cysteine proteinase domains of TcdB and the receptor binding domain of TcdA and expressed it in <I>Bacillus megaterium</I>. To ensure that mTcd138 is atoxic, 2 point mutations were introduced to the glucosyltransferase domain of TcdB, which essentially eliminates toxicity of mTcd138. Parenteral immunizations of mice and hamsters with mTcd138 induced protective antibodies to both toxins and provided protection against infection with the hyper-virulent <I>C. difficile</I> strain UK6.</P>
Adsorption Behavior of Environmental Hormone Bisphenol A onto Mesoporous Silicon Dioxide

Mesoporous silicon dioxide (meso-SiO)2) was prepared using cetyltrimethylammonium bromide as the structure-directing reagent and tetraethyl orthosicate as the silicon source. The influence of pH value on the adsorption behavior of bisphenol A (BPA) was investigated. The adsorption capacity of BPA onto meso-SiO_2increases slightly with pH value from 2 to 6, and then gradually decreases as further improving pH value. The effect of temperature was also studied, and the adsorption capacity of BPA gradually declines with increasing temperature. The adsorption kinetics and thermodynamics of BPA were examined. It is found that the adsorption of BPA onto meso-SiO_2 is in good agreement with Langmuir adsorption model. The rate constant of adsorption is 5.17 × 10^−3 g mg−1 min−1, and the maximum adsorption capacity is as high as 353.4 mg g^−1 at 20 ^oC.

Algorithm of Gaussian Sum Filter based on High-order UKF for Dynamic State Estimation

In this work we consider the state estimation problem in nonlinear/non-Gaussian systems. A new version of Gaussian sum estimation algorithm is developed here based on high-order unscented Kalman filter (HUKF). A sigma point selection method, high-order unscented transformation (HUT) technique is proposed for the HUKF, which can approximate the Gaussian distributions more accurately. We present the systematic formulation of Gaussian filters and develop efficient and accurate numerical integration of the optimal filter. We then go on to extend the use of the HUKF to discrete-time, nonlinear systems with additive, possibly non-Gaussian noise. The resulting filtering algorithm, called the Gaussian sum high-order unscented Kalman filter (GS-HUKF) approximates the predicted and posterior densities as a finite number of weighted sums of Gaussian densities. It is corroborated in the theoretical analysis and the simulation that the proposed Gaussian sum HUKF has integrated advantages with respect to computational accuracy and time complexity for nonlinear non-Gaussian filtering problems.
Liquidity Constraints, the Composition of Government Expenditure, and Economic Growth

This paper examines the impacts of liquidity constraints on economic growth and social welfare by incorporating the role of government expenditure into the overlappinggenerations model developed by Jappelli and Pagano in 1990s. In our model, the government can provide funds to the young faced with liquidity constraints. Our theoretical findings are as follows: (1) with exogenous technical progress, liquidity constraints on households raise the saving rate; (2) with endogenous technical progress, liquidity constraints and economic growth rate show an inverted U-shaped relationship; (3) with both exogenous and endogenous technical progress, the steady state per capita income first increases and then declines with the increase of liquidity constraints. Our empirical analysis with cross-country data supported this conclusion; (4) given certain values of the model parameters, social welfare in steady state may decrease with the reduction of liquidity constraints.

The extracellular inulinase of the marine yeast Pichia guilliermondii strain 1 was purified to homogeneity resulting in a 7.2-fold increase in specific inulinase activity. The molecular mass of the purified enzyme was estimated to be 50.0 kDa. The op-timal pH and temperature for the purified enzyme were 6.0 and 60C, respectively. The enzyme was activated by Mn²+, Ca²+, K+, Li+, Na+, Fe³+, Fe²+, Cu²+, and Co²+, but Mg²+, Hg²+, and Ag+ inhibited activity. The enzyme was strongly inhibited by phenylmethanesulphonyl fluoride (PMSF), iodoacetic acid, EDTA, and 1, 10-phenanthroline. The Km and Vmax values of the purified inulinase for inulin were 21.1 mg/mL and 0.08 mg/min, respectively. A large number of monosaccharides were de-tected after the hydrolysis of inulin. The deduced protein sequence from the cloned P. guilliermondii strain 1 inulinase gene contained the consensus motifs R-D-P-K-V-F-W-H and W-M-N-D-P-N-G, which are conserved among the inulinases from other microorganisms.
