MiR-886-5p Inhibition Inhibits Growth and Induces Apoptosis of MCF7 Cells

Background and Aims: To explore the molecular mechanisms of miR-886-5p in breast cancer., we examined roles in inhibiting growth and migration of MCF-7 cells. Methods: MiR-886-5p mimics and inhibitors were used to express or inhibit MiR-886-5p, respectively, and MTT and clone formation assays were used to determine the survival and proliferation. Hoechst 33342/ PI double staining was applied to detect apoptosis. The expression of caspase-3, caspase-8, caspase-9, MT1-MMP, VEGF-C and VEGF-D was detected by Western blotting, and the levels of MMP2 and MMP9 secreted from MCF-7 cells were assessed by ELISA. MCF-7 cell migration was determined by wound healing and Transwell assays. Results: We found that the growth of MCF-7 cells was inhibited upon decreasing miR-886-5p levels. Inhibiting miR-866-5p also significantly induced apoptosis and decreased the migratory capacity of these cells. The expression of VEGF-C, VEGF-D, MT1-MMP, MMP2, and MMP9 was also found to be decreased as compared to controls. Conclusions: Our data show that downregulation of miR-886-5p expression in MCF-7 cells could significantly inhibit cell growth and migration. This might imply that inhibiting miR-886-5p could be a therapeutic strategy in breast cancer.

The effects of exogenous abscisic acid (ABA) on phenolic characteristics of grapes and wines were investigated in Vitis vinifera cvs. Merlot and Cabernet Sauvignon. Exogenous ABA treatment at veraison significantly improved phenolic contents, mainly anthocyanins and flavonols, and antioxidant properties of the grape skins, but had no effects on total phenolics and antioxidant activities in the seeds or on basic fruit qualities. The wines made from ABA-treated grapes were also consequently enhanced in total phenolics, anthocyanins, flavonols and antioxidant activities. The proportions of methylated anthocyanins in the skins and acylated anthocyanins and derived pigments in the wines were decreased to some degree by exogenous ABA treatment. No distinct relationships were observed between ABA concentrations and phenolic characteristics, and the effects were observed even with 200 mg/L ABA. The results revealed that exogenous ABA applied at veraison offered opportunities to improve phenolic contents and nutritional values of grape skins and wines.

We investigated the small fatigue crack behavior of alloy GH4169 by using single-edge-notch tension specimens. Residual stress introduced by machining process was taken into consideration, and two stress levels were selected. A comparison was made between the experimental results of specimens with and without machining-induced residual stress. The results indicated that fatigue cracks of the two types of specimens initiated from surface inclusions or grain boundaries. For both types of specimens, small cracks grew very slowly when the crack lengths were less than 500 μm. The small crack growth might decelerate and retard temporarily for the existence of grain boundaries. The residual stress effect on crack growth can be identified at σ max = 380 MPa, i.e., compressive residual stress might impede the crack growth. However, this phenomenon was indistinguishable at σ max = 410 MPa.

In this study, ZnSe-TiO2 composites were synthesized by a facile hydrothermal-assisted sol-gel process and characterized by nitrogen adsorption isotherms (77 K), X-ray diffraction (XRD), scanning electron microscopy (SEM) with energy dispersive X-ray (EDX) analysis, transmission electron microscopy (TEM) and UV-vis diffuse reflectance spectrophotometry. The photocatalytic activity was investigated by decoloration methylene blue (MB), methyl orange (MO), and rhodamine B (Rh.B) in an aqueous solution under visible light irradiation. The results revealed that the photocatalytic activity of the ZnSe-TiO2 photocatalyst was much higher than that of pureTiO2. The ZnSe nanoparticles, which act as a photosensitizer, not only extend the spectral response of TiO2 to the visible region but also reduce charge recombinations.
Sonochemical Synthesis and Sonocatalysis Performance Behavior of Ag2Se and Ag2Se/TiO2 Nanocomposites

In this study, novel Ag2Se sensitized TiO2 nanocomposites were prepared by facile sonochemical assisted synthesis method. The as-prepared products were characterized by X-ray diffraction (XRD), Scanning electron microscopy (SEM) with energy dispersive X-ray (EDX) analysis, transmission electron microscopy (TEM), and N2 adsorption BET analysis. The as-prepared Ag2Se/TiO2 nanocomposites simultaneously possessed great adsorptivity for organic dyes and efficient charge separation properties. In the decolorization of rhodamine B, a significant enhancement in the reaction rate was observed for the Ag2Se/TiO2 nanocomposite compared to the cases of using pure P25 or TiO2. The sonocatalysis activity was higher due to the greater formation of reactive radicals, as well as to the increase of the active surface area of the Ag2Se/TiO2 nanocomposite.

The preferentially oriented In2O3 thin films were prepared on glass substrates by conventional magnetron sputtering with Ar+ plasma exposure at room temperature. Based on the x-ray diffraction, x-ray photoelectron spectroscopy, and UV photoelectron spectroscopy results, it was found that the Ar+ plasma exposure not only enhanced the low-temperature crystallization of In2O3 thin films, but also led to a dramatic improvement in the work function. Furthermore, it demonstrated that the shift mechanism of the work function in In2O3 thin films mainly combined with theelimination of oxygen defects and the change of the preferential orientation of In2O3 film surface.

Nanoscale ZnS-photosensitized TiO2 particles supported on acid functionalized multi-wall carbon nanotubes (MWCNTs) were prepared via a mild hydrothermal route. The new optical properties and porous structure of the as-prepared samples were characterized by transmission electron microscopy (TEM), scanning electron microscopy (SEM), BET surface area analysis,UV-vis DRS and X-ray diffraction (XRD). The photocatalytic activities of the composite catalysts were evaluated by the decomposition of an MB solution under visible light irradiation. The results indicate that the addition of an appropriate amount of MWCNTs as supports for TiO2 and ZnS efficiently inhibit the agglomeration of ZnS/TiO2 thus improving the dispersion of nanoparticles and the activity of visible photodegradation was remarkably improved.

This paper reported a simple deposition-precipitation method, introducing the metal (Ni, Ag and Sn) and $Na_2S{\cdot}5H_2O$ to preparedispersion metal sulfide nanoparticles on the surface of the Multi-walled carbon nanotube for synthesis of CNT-$M_xS_y$ ($NiS_2$, $Ag_2S$, SnS) composite photocatalysts. The characterization of the prepared CNT-$M_xS_y$ ($NiS_2$, $Ag_2S$, SnS) composites was performed by X-ray diffraction, scanning electron microscopy with energy dispersive X-ray analysis and BET analysis. Furthermore, the MB degradation rate constant for CNT-SnS composite was $5.68{\times}10^{-3}$ under visible light irradiation, which was much higher than the corresponding values for other samples. The detailed formation and photocatalytic mechanism are also provided here.

To improve the visible-light induced photocatalytic application performances of $TiO_2$, in this study, the $NiS_2$ modied $TiO_2$ composites were prepared by two methods: hydrothermal method and sol-gel method. The composites were denoted as hs-$NiS_2$/$TiO_2$, and sg-$NiS_2$/$TiO_2$ and characterized by XRD, UV-vis absorbance spectra, SEM, TEM, EDX, and BET analysis. The photocatalytic activities under visible light were investigated by the degradation of methyl orange (MO). The photodegradation rate of methyl orange under visible light with $NiS_2$/$TiO_2$ composites was markedly higher than that of pure $TiO_2$, and the effect of hs-$NiS_2$/$TiO_2$ composites was better than that of sg-$NiS_2$/$TiO_2$. The results indicate that the hydrothermal process could partly inhibit the agglomeration of $NiS_2$/$TiO_2$. Thus, the dispersion of nanoparticles was improved, and that the promoting effect of $NiS_2$ could extend the light absorption spectrum toward the visible region.
