        Antioxidative and Hepatoprotective Effects of Magnolol on Acetaminophen-induced Liver Damage in Rats

        Acute liver failure (ALF), an often fatal condition characterized by massive hepatocyte necrosis, is frequently caused by drug poisoning, particularly with acetaminophen (N-acetyl-p-aminophenol/APAP). Hepatocyte necrosis is consecutive to glutathione (GSH) depletion and mitochondrial damage caused by reactive oxygen species (ROS) overproduction. Magnolol, one major phenolic constituent of Magnolia officinalis, have been known to exhibit potent antioxidative activity. In this study, the anti-hepatotoxic activity of magnolol on APAP-induced toxicity in the Sprague-Dawley rat liver was examined. After evaluating the changes of several biochemical parameters in serum, the levels of aspartate aminotransferase (AST), alanine aminotransferase (ALT), and lactate dehydrogenase (LDH) were elevated by APAP (500 mg/kg) intraperitoneal administration (8 and 24 h) and reduced by treatment with magnolol (0.5 h after APAP administration; 0.01, 0.1, and 1 μg/kg). Histological changes around the hepatic central vein, lipid peroxidation (thiobarbituric acid-reactive substance/TBARS), and GSH depletion in liver tissue induced by APAP were also recovered by magnolol treatment. The data show that oxidative stress followed by lipid peroxidation may play a very important role in the pathogenesis of APAP-induced hepatic injury; treatment with lipid-soluble antioxidant, magnolol, exerts anti-hepatotoxic activity. Our study points out the potential interest of magnolol in the treatment of toxic ALF.

      • Effect of Eck Fistula on the Gastric Mucisa and the Liver in Rats

        Among the complications of clinical portacaval shunt, high incidence of peptic ulcer was well known especially in patients with hepatic disease. It is the logical explanation of the gastric hyperfunction that secretagogue, acid secretory stimulator or endogenous hormones are normally inactivated by the liver during cir-culating through portal system. The establishment of a portasystemic shunt provide a systemic diversion of these materials and caused acid hypersecretion in stomach. It has been known that Eck fistula in normal animals might produce deterious effects. With advancement of microvascular surgical technique, Eck fistula on the small animal became feasible and many investigators have reported the effect of Eck fistula in rats were not fatal in contrast with the large animals such as dogs, monkeys and pigs, etc. Authors performed Eck fistula in rats as an experimental model for evaluation of a long time effect on gastric mucosa.

        The mitogen-activated protein kinase phosphatase-1 (MKP-1) gene is a potential methylation biomarker for malignancy of breast cancer

        The mitogen-activated protein kinase (MAPK) phosphatase-1 (MKP-1) belongs to the MAPK cascades which are central to cell proliferation and apoptosis. The carcinogenic role of MKP-1 has been reported in many types of cancer but it has rarely been investigated in breast cancer. The present study was designed to evaluate the MKP-1 mRNA expression and its possible regulation by methylation of MKP-1 promoter in the model of several breast cancer cell lines and tissues as well as controls. Our data demonstrate MKP-1mRNA expression significantly decreased in five breast cancer cell lines compared to breast controls (P <0.01). Using the methylation-specific PCR (MSP) analysis,the unmethylated reaction (U) is dominant in both normal cell lines and benign breast tumors (100% vs. 86.2%), whereas the methylated reaction (M) is dominant in both breast cancer cell lines and invasive breast tumors (100% vs. 57.2%). In terms of methylation ratio (M/M+U), methylation level in MKP-1 promoter is significantly higher in the invasive breast tumor tissues (n = 152) than in benign breast tumor tissues (n = 29)(P < 0.0001). Assessing the methylation ratio of the promoter of MKP-1 gene to diagnose the breast malignancy (invasive vs. benign), the area under the receiver-operating characteristic (ROC) curve was 0.809(95% CI: 0.711-0.906, P < 0.001). The best performance for this prediction has a sensitivity of 76.32% and a specificity of 82.76% at the cutoff value of 0.38. Taken together, we firstly demonstrated that the promoter methylation of MKP-1 gene is a potential breast cancer biomarker for breast malignancy.

        Prediction of the Duration to Next Admission for an Acute Affective Episode in Patients with Bipolar I Disorder

        Objective: Predicting disease relapse and early intervention could reduce symptom severity. We attempted to identify potential indicators that predict the duration to next admission for an acute affective episode in patients with bipolar I disorder. Methods: We mathematically defined the duration to next psychiatric admission and performed single-variate regressions using historical data of 101 patients with bipolar I disorder to screen for potential variables for further multivariate regressions. Results: Age of onset, total psychiatric admissions, length of lithium use, and carbamazepine use during the psychiatric hospitalization contributed to the next psychiatric admission duration positively. The all-in-one found that hyperlipidemia during the psychiatric hospitalization demonstrated a negative contribution to the duration to next psychiatric admission; the last duration to psychiatric admission, lithium and carbamazepine uses during the psychiatric hospitalization, and heart rate on the discharge day positively contributed to the duration to next admission. Conclusion: We identified essential variables that may predict the duration of bipolar I patients’ next psychiatric admission. The correlation of a faster heartbeat and a normal lipid profile in delaying the next onset highlights the importance of managing these parameters when treating bipolar I disorder.

        Galangin Prevents Acute Hepatorenal Toxicity in Novel Propacetamol-Induced Acetaminophen-Overdosed Mice

        Acetaminophen (APAP) overdose causes severe liver and kidney damage. APAP-induced liver injury (AILI) represents the most frequent cause of drug-induced liver failure. APAP is relatively insoluble and can only be taken orally; however, its prodrug, propacetamol, is water soluble and usually injected directly. In this study, we examined the timedependent effects of AILI after propacetamol injection in mice. After analyses of alanine aminotransferase and aspartate aminotransferase activities and liver histopathology, we demonstrated that a novel AILI mouse model can be established by single propacetamol injection. Furthermore, we compared the protective and therapeutic effects of galangin with a known liver protective extract, silymarin, and the only clinical agent for treating APAP toxicity, N-acetylcysteine (NAC), at the same dose in the model mice. We observed that galangin and silymarin were more effective than NAC for protecting against AILI. However, only NAC greatly improved both the survival time and rate consequent to a lethal dose of propacetamol. To decipher the hepatic protective mechanism(s) of galangin, galangin pretreatment significantly decreased the hepatic oxidative stress, increased hepatic glutathione level, and decreased hepatic microsomal CYP2E1 levels induced by propacetamol injection. In addition, propacetamol injection also reproduced the probability of APAP-induced kidney injury (AIKI), appearing similar to a clinical APAP overdose. Only galangin pretreatment showed the protective effect of AIKI. Thus, we have established a novel mouse model for AILI and AIKI using a single propacetamol injection. We also demonstrated that galangin provides significant protection against AILI and AIKI in this mouse model.

        3D Scaffold with PCL Combined Biomedical Ceramic Materials for Bone Tissue Regeneration

        Three-dimensional porous biodegradable polymer scaffolds have been widely used for tissue engineering of bone repair or regeneration. The primary function of scaffolds is to provide structure support for the cells adhesion and proliferation. This study selects the Poly-ε-caprolactone (PCL) as material, NaCl mixed with hydroxyapatite (HA) or nano-aluminum oxide (nAl2O3) for porous scaffold. This study uses the solvent casting/particulate leaching method to fabricate the porous scaffold. The authors discuss the compression mechanical properties, physical properties (porosity, moisture content, contact angle) of a pure PCL, PCL/mHA,PCL/nHA and PCL/nAl2O3 scaffolds. In vitro cell culture is used for osteoblast cell (MG63) and the microculture tetrazolium test (MTT) is undertaken in the scaffold. The scaffolds are implanted to the femur of rats and histological examination is attempted after 2 weeks. The experimental results indicate that HA and nAl2O3 can improve the hydrophilic property. In conclusion, the PCL/nHA scaffold exhibits splendid in vivo biocompatibility and osteogenesis.

      • Cryobanking of Semen, Embryo and Oocytes for Animal Industry

        Genetic resources of buffalo, cattle, goat, sheep, deer, rabbit, pig, chicken, duck, goose, turkey and swan in Taiwan are conserved in living form or frozen genetic materials. Preserving farm animals in living form must conserve a considerable number of mature individuals with reproductive potential, and continued funding and breeding space are necessary. Based on the afore‐mentioned restraints and consideration of risk diversification, living animals are preserved in Taiwan Livestock Research Institute (TLRI) branches and breeding stock reproduction grounds throughout Taiwan. Genetic materials preserved in frozen form include germ cell (sperm, egg and embryo), tissue, somatic cell, cell line, DNA and gene pool. Those preserved for long term more than 10 years are conserved in liquid nitrogen storage tank at ‒185 to ‒196℃. Animal industry on utilization of farm animal genetics emphasizes the importance of in situ conservation and considers ex situ conservation as an essential complementary activity to in situ. In situ and ex situ utilization are complementary, not mutually exclusive. The exact strategy of germplasm cryobanking will clearly depend on the conservation objectives of TLRI. In situ and ex situ strategies differ in their capacity to achieve the different bio‐utilization objectives. Based on the objectives for bio‐utilization and conservation, the existing national technical capacity and infrastructure for cryoconservation, and amount of capital to invest in developing and maintaining a gene bank for food animal genetic resources (FAnGR), each country should determine for which FAnGR, if any, national cryobanking program should be undertaken. The health and sanitary issues of animals that must be considered when establishing and operating gene banks for animal genetic resources to help prevent the conservation of potentially dangerous pathogens along with the valuable genetic material in the ultra‐low temperature cell repository. Bio‐utilization of material stored in the bio‐bank must eventually be thawed and used to create new animals and therefore, national organization and annotation of the stored material is critical to ensure its proper utilization. Stock animals are the source of semen, embryo and oocytes are usually privately owned. This ownership may or may not change during the gene banking process, but the terms of agreements between bio‐banks and donors must be explicitly defined. Taiwan Animal Germplasm Center now conserves genetic resources of 77 livestock breeds and strains, including 19 native and 38 foreign species as well as 20 new breeds produced by inbreeding in poultry or breed‐crossing with the artificial insemination in livestock. Cryobanking of semen of native animals is for genetic diversity but also for risk management to long‐term global food security. Ex situ conservation program involving in vitro storage of germplasm cryobanking can contribute to ensure that it will be available to allow livestock keepers and animal breeders to confront future changes in animal production and economic environments.
