교잡종 찰옥수수에 대한 연구 III : Ⅲ. 국내 옥수수 유전자원 수집종에 대한 주요 특성 분포

2006년 국내 각 지역으로부터 수집된 재래종옥수수 유전자원을 관행방법에 의해 파종하고 주요특성별로 분포도를 살펴본 결과는 다음과 같다. 1. 수집된 유전자원에 대한 발아율은 대부분 80∼100%이상의 분포를 보였으나, 일부 수집계통에는 30% 미만의 극히 저조한 발아율을 보였다. 2. 수집자원에 대한 초기생육을 살펴보면 극히 생육이 저조한 것이 40% 정도를 보였고, 극히 우수한 자원은 3%미만으로 나타났다. 3. 간장은 대부분이 150∼199cm범위에 55%가 분포하고 있었으나, l00cm이하가 5%, 250cm이상이 8%로 나타나 수집종간에 큰 차이를 보였다. 4. 주당 분얼수는 1개 미만이 40%의 분포를 보였으나, 일부 수집종에서 2 내지 3개의 분얼수를 보이는 계통이 확인되었다. 5. 개화소요일수는 61∼64일의 범위에서 40%, 57∼60일의 범위에서 30%의 분포를 보였으며, 특히 일부 수집종의 경우 55일 이하의 극조생종도 10% 정도의 분포를 보였다. 6. 이삭길이는 대부분 11∼15cm범위에 60%가 포함되어 있으나, 일부계통에서 5cm미만이 2%, 16∼20cm의 이삭길이도 10%가 분포하였다. Germination ratio on the 300 collected genetic resources showed 80 to 100 percentage, but those of some accessions were very low as below 30 percentage. Early growth phase of most of these lines were very poor while some collected lines were very vigor. Range of stem height showed from 150 to 199cm, but that of some accessions showed below 100cm as five percentage and some of them was over 250cm as eight percentage. Number of tillers per plant appeared less one but some collected lines have two to three tillers per plant. In days to tasseling, some accessions showed more four per plant. Ear length of these accession showed above 60 percentage as 11 to 15cm, while that of some collected lines showed over 10 percentage as 16 to 20cm.

Previous studies have demonstrated that repeated restraint stress in rodents produces increases in depression and anxietylike behaviors and alters the expression of corticotrophinreleasing factor (CRF) in the hypothalamus. The current study focused on the impact of Bupleurum falcatum (BF) extract administration on repeated restraint stress-induced behavioral responses using the forced swimming test (FST) and elevated plus maze (EPM) test. Immunohistochemical examinations of tyrosine hydroxylase (TH) expression in rat brain were also conducted. Male rats received daily doses of 20, 50, or 100 mg/kg (i.p.) BF extract for 15 days, 30 min prior to restraint stress (4 h/day). Hypothalamicpituitary- adrenal axis activation in response to repeated restraint stress was confirmed base on serum corticosterone levels and CRF expression in the hypothalamus. Animals that were pre-treated with BF extract displayed significantly reduced immobility in the FST and increased open-arm exploration in the EPM test in comparison with controls. BF also blocked the increase in TH expression in the locus coeruleus of treated rats that experienced restraint stress. Together, these results demonstrate that BF extract administration prior to restraint stress significantly reduces depression and anxiety-like behaviors, possibly through central adrenergic mechanisms, and they suggest a role for BF extract in the treatment of depression and anxiety disorders.
Deoxycytidine Production by Metabolically Engineered Corynebacterium ammoniagenes

Corynebacterium ammoniagenes N424 was metabolically modified to isolate overproducers of deoxycytidine. Inosine auxotrophy (ino-) was initially introduced to prevent the flow of PRPP (phosphoribosyl pyrophosphate)into the purine biosynthetic pathway by random mutagenesis using N-methyl-N'-nitro-N-nitrosoguanidine. Following that, mutants possessing hydroxyurea resistance (HUr) were isolated to increase the activity of ribonucleoside diphosphate reductase, which catalyzes the reduction of ribonucleoside diphosphate to deoxyribonucleoside diphosphate. Then, in order to block the flow of dCTP into the TMP biosynthetic pathway via dUTP, thymine auxotrophy (thy-) was introduced into the mutant IH30 with ino- and HUr. The resulting mutant IM7, possessing the characteristics of ino-, HUr, and thy-, was deficient in dCTP deaminase and produced significantly higher amounts of deoxycytidine (81.3 mg/L) compared to its mother strain IH30(6.2 mg/L). Deoxycytidine productivity was further enhanced by isolating the mutant IU19, which was resistant to 5-fluorouracil, an inhibitor of carbamoyl phosphate synthase. This enzyme catalyzed the synthesis of carbamoyl phosphate from glutamine, HCO3-, and ATP. 5-Fluorouracil also inhibited aspartate transcarbamoylase,catalyzeing the condensation of carbamoyl phosphate and aspartate. Finally, 5-fluorocytosine resistance (FCr) was introduced into the mutant strain IU19 to relieve the repression caused by accumulation of pyrimidine nucleosides. The mutant strain IC14-C6 possessing all the five characteristics described above produced 226.3 mg/L of deoxycytidine, which was at least 2,000 fold higher compared to the wild type,and accumulated only a negligible amount of other pyrimidines under shake flask fermentation.

The fed-batch culture system was employed to enhance production of α-ketoglutarate (α-KG) by the strainsof Corynebacterium glutamicum, whose genes encoding the key enzymes responsible for the biosynthesis of L-glutamate from α-KG were deleted. In a shake flask fermentation, C. glutamicum JH110 in which the 3 genes, gdh (encoding glutamate dehydrogenase), gltB (encoding glutamate synthase), and aceA (encoding isocitrate lyase) were disrupted showed the highest production of α-KG (12.4 g/L) compared to the strains JH102 (gdh mutant), JH103 (gltB mutant), and JH107 (gdh gltB double mutant). In the fed-batch cultures using a 5 L-jar fermenter, the strain JH107 produced more α-KG (19.5 g/L), but less glutamic acid (23.3 g/L) than those produced by the parent strain HH109, as well as JH102. The production of α-KG was significantly enhanced and the accumulation of glutamicacid was minimized by the ammonium-limited fed-batch cultures employing C. glutamicum JH107. Further improvement of α-KG production by the strain JH107 was achieved through the ammonium-limited fed-batch culture with the feeding of molasses, and the levels of α-KG and glutamic acid produced were 51.1 and 0.01 g/L, respectively.
뇌기능 장애 환자의 가상영상(Moving Sorround) 자극에 따른 자세 균형 제어

뇌기능 장애 환자에서 자세균형 제어능력의 저하는 보행 및 일상생활동작 수행 등에 어려움을 초래하며 이에 대한 정확한 평가 및 치료를 위하여 일상의 환경변화와 유사한 상황을 제공하고 이예 따른 자세제어력을 파악하는 것이 중요하다. 본 연구는 뇌기능 장애환자에서 움직이는 환경에 따른 자세균형제어기능을 정확히 평가하는 환경의 움직임이 자세균형제어에 미치는 영향을 분석하였다. 15명의 뇌기능 장애 환자들과 정상인 15명을 대상으로 실생활과 유사한 환경의 조성을 위하여 HMD를 이용한 가상영상 환경변화(Moving Surround)를 네가지 움직임 종류에 따라 제공하였다. 자세동요의 정도는 힘판을 이용하여 신체압력중심의 변화를 전체이동거리, 동요주파수, 최대 빈도 COP 위치 등으로 측정하였으며 환경의 차이에 따른 변화를 비교 분석하였다. 연구결과 검사 재검사 신뢰도 평가에서 일관된 분석결과를 나타냈고 뇌기능장애 환자와 정상인간의 분석에서는 두 그룹간의 차이를 확인할 수 있었다. 특히 전후로 빠르게 변하는 가상영상 환경에서 가장 큰 자세동요를 나타내었고 통계적으로 유의한 차이가 있었다. 본 연구를 통해 뇌기능 장애 환자에서 가상영성 환경변화가 자세균형제어에 미치는 영향을 확인할 수 있었으며 이러한 환자들을 위한 치료환경 조성 등에도 유용한 자료로 쓰일 수 있을 것으로 사료된다.

The distribution of barbiturates in the model membranes of total lipids (SPMVTL) and total phospholipids (SPMVPL) extracted from synaptosomal plasma membrane vesicles was determined by employing a fluorescent probe technique. The two fluorescent probes 2-(9-anthroyl)stearic acid and 12-(9-anthroyl)stearic acid were utilized as probes for the surface and the hydrocarbon interior of the outer monolayer of the SPMVTL and SPMVPL, respectively The Stern-Volmer equation of fluorescent quenching was modified to calculate the relative distribution. The analysis of preferential quenching of these probes by barbiturates indicates that pentobarbital, hexobarbital, amobarbital and Phenobarbital are predominantly distributed on the surface area, while thiopental sodium has an accessibility to the hydrocarbon interior of the outer monolayer of the SPMVTL and SPMVPL. From these results, it is strongly suggested that the more effective penetration into the hydrocarbon interior of the outer monolayer of the membrane lipid bilayer could result in a higher general anesthetic activity
