        Characteristics of 5-Hydroxytryptamine Receptors Involved in Contraction of Feline Ileal Longitudinal Smooth Muscle

        A number of studies have demonstrated that 5-hydroxytryptamine (5-HT) can induce muscle contraction or relaxation response and enhance secretion in the gastrointestinal tract via a multiplicity of 5-HT receptor subtypes. In the present study, we investigated the pharmacological characterization of the 5-HT-induced contractile response in longitudinal smooth muscle isolated from the feline ileum. Addition of 5-HT into muscle chambers enhanced the basal tone and spontaneous activity in a concentration-dependent manner. The neurotoxin tetrodotoxin did not alter the 5-HT-induced contraction of the longitudinal muscles. Neither atropine nor guanethidine affected the contraction. The 5-HT agonists, 5-methylserotonin hydrochloride and mosapride, also evoked concentration-dependent contractions. The 5-HT-induced contraction was enhanced by the 5HT2 receptor antagonist ketanserin and the 5-HT3 receptor antagonist ondansetron but was inhibited by the 5-HT1 receptor antagonist methysergide and 5-HT4 receptor antagonist GR113808. These results indicate that 5-HT1 and 5-HT4 receptors may mediate the contraction of the 5-HT-induced response and 5-HT2 and 5-HT3 receptors may mediate 5-HT-induced relaxation in feline ileal longitudinal smooth muscles.

      • Quantitative Analysis of Eugenol in Clove Extract by a Validated HPLC Method

        <B>Abstract</B><P>Clove (Eugenia caryophyllata) is a well-known medicinal plant used for diarrhea, digestive disorders, or in antiseptics in Korea. Eugenol is the main active ingredient of clove and has been chosen as a marker compound for the chemical evaluation or QC of clove. This paper reports the development and validation of an HPLC-diode array detection (DAD) method for the determination of eugenol in clove. HPLC separation was accomplished on an XTerra RP18 column (250 4.6 mm id, 5m) with an isocratic mobile phase of 60 methanol and DAD at 280 nm. Calibration graphs were linear with very good correlation coefficients (r2 &gt; 0.9999) from 12.5 to 1000 ng/mL. The LOD was 0.81 and the LOQ was 2.47 ng/mL. The method showed good intraday precision (RSD 0.080.27) and interday precision (RSD 0.321.19). The method was applied to the analysis of eugenol from clove cultivated in various countries (Indonesia, Singapore, and China). Quantitative analysis of the 15 clove samples showed that the content of eugenol varied significantly, ranging from 163 to 1049 ppb. The method of determination of eugenol by HPLC is accurate to evaluate the quality and safety assurance of clove, based on the results of this study.</P>

      • Unplanned ICU readmission is related more medical care cost and poorer prognosis

        The purposes of this study were to compare the difference in mortality rate and medical care cost between the patients with unplanned ICU readmission(RA) group and non-readmission(non-RA) group, and to evaluate whether unplanned ICU readmission rate is considered to be one of surrogates to qualify the ICU. This study is at questionnaire and the health insurance statement by the Korean Health Insurance Review & Assessment Service. A total of 39,576 patients were analyzed. ‘Unplanned ICU readmission’ was defined as the admission within 72 hours without planning the readmission in advance after discharge form the ICU. Unplanned ICU RA rate was 1.5%(589 patients among 39,576 patients admitted at the ICUs). The mortality rate in the ICU was 19.19% and 10.87% in the unplanned ICU RA group and the non-RA group, respectively. The unplanned ICU RA rates were higher in the institutes with more the number of total beds(p=0.001) more ICU beds(p=0.002) RRT(p=0.016) lower number of ICU patients to nurses(p=0.010) and full time intensivist(p=0.001). The institutes placed full-time intensivist or RRT had more severe patients, but lower mortality rate. The incidence of unplanned ICU RA is increased in ventilator use(OR=2.143, p= <.0001), cardiovascular comorbidity (OR=2.961, p=<.0001), more ICU beds(OR=1.004, p=0.009). The risk factors for unplanned ICU readmission were ventilator use, cardiovascular comorbidity, more ICU beds. The average of medical care cost was $19013 in the unplanned ICU RA group, which was 1.75 folds higher than that in the non-RA group(p=<.0001). The medical care cost per days was $660 in the unplanned ICU readmission group, which was 12%(1.12 folds) higher than that in the non-readmission group(p=<.0001). Unplanned ICU readmission was associated negative impacts on patient outcomes, such as more hospital days and increasing medical care cost. Unplanned ICU readmission rate is considered to be one of surrogates to evaluate the quality of ICU.

      • Isolation of adipose-derived stem cells by using a subfractionation culturing method

        <P><B><I>Objective:</I></B> Adipose-derived stem cells (ASCs) isolated from subcutaneous adipose tissue have been tested in clinical trials. However, ASCs isolated by enzyme digestion and centrifugation are heterogeneous and exhibit wide variation in regenerative potential and clinical outcomes. Therefore, we developed a new method for isolating clonal ASCs (cASCs) that does not use enzyme digestion or centrifugation steps.</P><P><B><I>Research design and methods:</I></B> In addition to cell surface markers and differentiation potential, we compared the mitogenic, paracrine and hair growth-promoting effects of ASCs isolated by the gradient centrifugation method (GCM) or by the new subfractionation culturing method (SCM).</P><P><B><I>Results:</I></B> We selected three cASCs isolated by SCM that showed high rates of proliferation. The cell surface markers expressed by ASCs isolated by GCM or SCM were very similar, and SCM-isolated ASCs could potentially differentiate into different cell lineages. However, cASC lines exhibited better mitogenic and paracrine effects than ASCs isolated by GCM. The expression of Diras3, Myb, Cdca7, Mki67, Rrm2, Cdk1 and Ccna2, which may play a key role in cASC proliferation, was upregulated in cASCs. In addition, cASCs exhibited enhanced hair growth-promoting effects in dermal papilla cells and animal experiments.</P><P><B><I>Conclusions:</I></B> SCM generates a highly homogeneous population of ASCs via a simple and effective procedure that can be used in therapeutic settings.</P>

        Distribution and characteristics of radioactivity$(^{232}Th,\;^{226}Ra,\;^{40}K,\;^{137}Cs\;and\;^{90}Sr)$ and radiation in Korea

        The concentrations of natural and artificial radionuclides in soil and gamma ray dose rate in air at 233 locations in Korea have been determined. The national mean concentrations of $^{232}Th,\;^{226}Ra,\;^{40}K,\;^{137}Cs\;and\;^{90}Sr$ in soil were $60{\pm}31,\;33{\pm}14,\;673{\pm}238,\;35{\pm}9.3\;and\;5.0{\pm}3.4\;Bq\;kg^{-1}$, respectively. The mean gamma-ray dose rate at 1 m above the ground was $7918\;nGy\;h^{-1}$. $^{137}Cs$ concentration had highly significant correlation with organic matter content and cation exchange capacity. $^{90}Sr$ concentration had slightly coherent with pH. The results have been compared with other global radioactivity and radiation measurements.

        Aeromonas hydrophila KH-54 가 분비하는 유기폐수처리용 생물응집제 생산조건의 최적화

        For the need of bio-degradable flocculant in stage of wastewater treatment, some cultural conditions of bioflocculant production were optimized with Aeromonas hydrophila KH-54. About 260 strains of type culture and bacteria isolated from marsh, pond, activated sludge, etc were examined for their ability to flocculate kaolin particles and swine wastewater. Among them, KH-54 showed the highest flocculating activity and was identified as Aeromonas hydrophila according to the cultural, morphological and physiological properties. The maximum production of the flocculant secreted by Aeromonas hydrophila KH-54 was observed in culture medium containing 2.0% mannitol, 0.05% ammonium chloride, 0.02% potassium phosphate dibasic, 0.01% MgSO₄· 7H₂O and 0.05% yeast extract at initial pH 7.0 when cultured on rotary shaker controlled at 25℃ and 150 rpm. Under the optimized condition, the flocculating ability reached to 770 units/㎖ of kaolin flocculating activity and 81% of NTU removal efficiency against swine wastewater after 4 days cultivation. The bioflocculant was also effective on various organic wastewaters other than swine wastewater, showing NTU removal rate ranging from 92% to 34%.

        고온호기성소화의 모델에 관한 연구

        슬럿지의 안정화와 위생처리를 위한 연속흐름식 및 반 연속식 반응조에 반응열을 이용한 친열호기성 소화에 대한 Model이 설정되었다. 반응속도식은 생물학적 분해가능한 유기고형물 농도에 대한 1차반응식을 적용하였으며 산소농도에 대한 영향을 파악하기 위하어 하나의 항을 추가로 삽입하였다. simulation을 위하여 계수나 운전요건들은 대부분 실측된 자료들이 사용되었으며 발효액에서 실측된 mixing unit의 에너지 주입 및 산소 전달특징을 고려하였다. 관측된 결과들은 아래와 같다. (1) 효율적인 슬럿지의 안정화의 위생처리를 위하어 글럿지의 농도가 높아야 되며 단열상태가 좋아야 한다. (2) impeller의 회전수와 공기주입속도에 대한 적절한 운전점은 유기물제거속도(r_(eff))의 증가뿐만 아니라 효율(E) 즉 단위에너지 주입에 대한 유기물 제거량도 고려하여 선정되어야 한다. (3) Batch와 연속흐름식 그리고 반 연속식 운전의 비교로부터 반 연속식 운전이 슬럿지의 안정화와 위생처리에 효과적이며 이것은 Batch 식보다 체류기간은 길지만 저조류가 필요없고 또 외부조건(독성물질등)에 덜 민감하며 연속식운전보다 체류기간이 짧다는 장점을 지닌다. (4) 이론치와 실측치의 비교는 일부 추측에 의한 값들도 적용되었지만 해당 글럿지에 대한 반응계수들이 측정된다면 이론에 의한 Model로 실측값에 상당히 접근될 수 있을 것이다. Models of the autothermal thermophilic aerobic digestion process for stabilization and pasteurization of sewage sludge were proposed. They can be applied to the continuous and semi-continuous operations in completely stirred tank reactor. Oxygen concentration term was added to the first order reaction kinetic to investigate the influence of the oxygen concentration on the reaction rate. Sludge was considered as a nonnewtonian (pseudoplastic) fluid. For the effective operation of the process the sludge concentration must be high and the thermal protection of the reactor with isolation material should be required. The RPM of turbine impeller and the air supply velocity must be selected on the basis not only of the organic removal rate but also of the efficiency; quantity of the removal organics per unit energy input. Also the semi-continuous operation appears one of the most effective process for the stabilization and pasteurization of the sludge.
