In this paper, two statistical methods were applied to optimize medium components to improve the production of the milk-clotting enzyme by Bacillus amyloliquefaciens D4. First, wheat bran juice, skim milk powder, and Na2HPO4 were shown to have significant effects on D4 enzyme production using the Plackett?Burman experimental design. Subsequently, an optimal medium was obtained using the Box?Behnken method, which consisted of 3.31 g/l of skim milk powder, 5.0 g/l of sucrose, 0.1 g/l of FeSO4?7H2O, 0.1 g/l of MgSO4?7H2O, 0.1 g/l of MnSO4?2H2O, 0.1 g/l of ZnSO4?7H2O, 1.52 g/l of Na2HPO4, and 172.45 g/l of wheat bran juice. With this optimal medium, the milk-clotting enzyme production was remarkably enhanced. The milk-clotting enzyme activity reached 3,326.7 SU/ml after incubation of 48 h, which was 1.76-fold higher than that of the basic medium, showing that the Plackett?Burman design and Box?Behnken response surface method are effective to optimize medium components, and B. amyloliquefaciens D4 possessed a high rennet-producing capacity in the optimal medium.

Background/Aims:This study aimed to explore the effect of gut microbiota-regulated Kupffer cells (KCs) on colorectal cancer (CRC) liver metastasis. Methods: A series of in vivo and in vitro researches were showed to demonstrate the gut microbiota and its possible mechanism in CRC liver metastasis. Results: Fewer liver metastases were identified in the ampicillin-streptomycin-colistin and colistin groups. Increased proportions of Parabacteroides goldsteinii, Bacteroides vulgatus, Bacteroides thetaiotaomicron, and Bacteroides uniformis were observed in the colistin group. The significant expansion of KCs was identified in the ampicillin-streptomycin-colistin and colistin groups. B. vulgatus levels were positively correlated with KC levels. More liver metastases were observed in the vancomycin group. An increased abundance of Parabacteroides distasonis and Proteus mirabilis and an obvious reduction of KCs were noted in the vancomycin group. P. mirabilis levels were negatively related to KC levels. The number of liver metastatic nodules was increased in the P. mirabilis group and decreased in the B. vulgatus group. The number of KCs decreased in the P. mirabilis group and increased in the B. vulgatus group. In vitro, as P. mirabilis or B. vulgatus doses increased, there was an opposite effect on KC proliferation in dose- and time-dependent manners. P. mirabilis induced CT26 cell migration by controlling KC proliferation, whereas B. vulgatus prevented this migration. Conclusions: An increased abundance of P. mirabilis and decreased amount of B. vulgatus play key roles in CRC liver metastasis, which might be related to KC reductions in the liver.

Background: A common genetic variant rs3757318, located in intron of C6orf97, was firstly identified to be associated with breast cancer (BC) risk by a genome-wide association (GWA) study. However, subsequent validation studies with different ethnicities have yielded conflicting results. Materials and Methods: We performed a meta-analysis to synthesize all available data for evaluating the precise effect of this variant on BC susceptibility. Results: A total of 8 articles containing 11 studies with 62,891 cases and 65,635 controls were included in this meta-analysis. When compared to the G allele, the rs3757318-A allele was significantly associated with BC risk with the pooled OR of 1.21 (95% CI=1.15 - 1.29, P<0.001) but with obvious between-study heterogeneity (P=0.040). Stratified analysis suggested that diversity of ethnicity along with control source may explain part of the heterogeneity. Similarly, significant associations were also identified in heterozygote, homozygote, dominant and recessive genetic models. Sensitivity and publication bias analyses indicated robust stability of our results. Conclusions: Our present meta-analysis demonstrated that the variant rs3757318 is associated with increased BC risk. Nevertheless, further studies are needed to clarify the underlying biological mechanisms.

Beclin 1 is a key factor for initiation and regulation of autophagy, which is a cellular catabolic process involved in tumorigenesis. To investigate the role of alternative splicing of Beclin1 in the regulation of autophagy in leukemia cells, Beclin1 mRNA from 6 different types of cell lines and peripheral blood mononuclear cells from 2 healthy volunteers was reversely transcribed, subcloned, and screened for alternative splicing. New transcript variants were analyzed by DNA sequencing. A transcript variant of Beclin 1 gene carrying a deletion of exon 11, which encoded a C-terminal truncation of Beclin 1 isoform, was found. The alternative isoform was assessed by bioinformatics, immunoblotting and subcellular localization. The results showed that this variable transcript is generated by alternative 3' splicing, and its translational product displayed a reduced activity in induction of autophagy by starvation, indicating that the spliced isoform might function as a dominant negative modulator of autophagy. Our findings suggest that the alternative splicing of Beclin 1 might play important roles in leukemogenesis regulated by autophagy.

<P>This paper focuses on the improvement of the diagnostic accuracy of focal liver lesions by quantifying the key features of cysts, hemangiomas, and malignant lesions on ultrasound images. The focal liver lesions were divided into 29 cysts, 37 hemangiomas, and 33 malignancies. A total of 42 hybrid textural features that composed of 5 first order statistics, 18 gray level co-occurrence matrices, 18 Law's, and echogenicity were extracted. A total of 29 key features that were selected by principal component analysis were used as a set of inputs for a feed-forward neural network. For each lesion, the performance of the diagnosis was evaluated by using the positive predictive value, negative predictive value, sensitivity, specificity, and accuracy. The results of the experiment indicate that the proposed method exhibits great performance, a high diagnosis accuracy of over 96% among all focal liver lesion groups (cyst vs. hemangioma, cyst vs. malignant, and hemangioma vs. malignant) on ultrasound images. The accuracy was slightly increased when echogenicity was included in the optimal feature set. These results indicate that it is possible for the proposed method to be applied clinically.</P>

Twenty natural compounds isolated fromtraditional Chinese medicines were examined for theirantiviral activities against PCV2 in vitro. Antiviral activitywas analyzed by the assays of blocking of virus entry,inhibition of virus replication and inactivation of virusparticles in the virus suspension. 50% cytotoxic concentration(CC50) and 50% effective concentration (EC50) for eachcompound were determined by using the MTT method andimmunofluorescence assay (IFA), respectively. The resultsobtained from the inhibition of virus replication andinactivation of virus particles showed that the maximuminhibition rates for matrine and scutellarin were 57 and72.69% and their SI were 29.87 and 18.66, respectively. However, the results from the assays of inhibition of virusentry and inhibition of virus replication demonstrated thatboth aesculetin and arecoline hydrochloride stimulatedPCV2 replication, with the maximum stimulation rate of387.3 and 219.4%. In summary, both matrine and scutellarinhad antiviral activity against PCV2, while aesculetin andarecoline hydrochloride promoted PCV2 infection replicationin vitro.

In this study, a green and efficient ultrasound-assisted enzymatic extraction (UAEE) method was establishedfor total polyphenols (TPP) extraction from E. nigrum aerial parts. Fractional Factorial design (FFD)was initially implemented for the selection of significant parameters affecting TPP yield and then Box-Behnken design-response surface methodology (BBD-RSM) was employed to optimize the parameters. The optimum extraction conditions for UAEE were as follows: incubation temperature of 38.39 ℃, incubationtime of 3.39 h, tannase of 386.53 U/g of sample (U/g), and cellulase of 224.42 U/g. Under the optimumconditions, the TPP yield was 52.17 ± 0.39 mg/g, which was 1.62 and 1.73 times those of themethods of ultrasound-assisted extraction (UAE) and enzyme-assisted extraction (EAE), respectively. The crude extract was purified by HPD-600 microporous resin, and the purity was improved from8.11% to 22.56%. In addition, the results of antioxidant and antibacterial activity assays showed thatthe purified extract has stronger bioactivities in contrast with the crude extract. This study indicated thatUAEE was an effective technique for the extraction of TPP from the aerial parts of E. nigrum, and TPPextracted by this extraction method could be applied in pharmaceuticals, cosmetics, and food industries.

<P><B>Abstract</B></P> <P>For accurate force tracking of an electro-hydraulic servo system (EHSS), this paper presents a state-space representation of model predictive control (MPC). EHSS is highly nonlinear because of the transmission nonlinearities, variations of the trapped fluid volume, internal friction, and nonlinear load-based nonlinearities. A nonlinear mathematical model is built, and model parameters are identified using a grey-box system identification method. Then MPC is firstly designed, and to improve the robustness, a nominal mathematical model with proportional integral control (PIC) is constructed for MPC design. Three implementation scenarios are investigated in MATLAB/Simulink and real-time LabVIEW-based experimental rig. The control performance is compared between and among PIC, MPC, and hybrid MPC–PIC controller.</P>
