        Inhibition of MicroRNA-92a Improved Erectile Dysfunction in Streptozotocin-Induced Diabetic Rats via Suppressing Oxidative Stress and Endothelial Dysfunction

        Purpose: To determine whether microRNA could be a therapy target of erectile dysfunction (ED) and the underlying mecha-nisms. Materials and Methods: Eight-week-old fasting male SD rats were intraperitoneally injected with streptozotocin to construct diabetic rat models. Diabetic ED rats were treated with miRNA-92a inhibitor. The cavernous nerves were electrically stimu-lated to measure the intracavernous pressure and mean arterial pressure of rats in each group. After the detection, the penile cavernous tissues are properly stored for subsequent experiments. Rat aortic endothelial cells were used in in vitro studies. Results: The expression of miR-92a was significantly increased in the corpus cavernosum of Streptozocin (STZ)-induced di- abetic rats and injection of miR-92a antagomir into the corpus cavernosum of diabetic rats significantly increased eNOS/NO/ cGMP signaling pathway activities, cavernous endothelial cell proliferation, endothelial cell-cell junction protein expression and decreased the levels of oxidative stress. These changes restored erectile function in STZ-induced diabetic rats. Moreover, in vitro study demonstrated that the miR-92a expression increased significantly in endothelial cells treated with high glucose, inhibiting AMPK/eNOS and AMPK/Nrf2/HO-1 signaling pathways in rat aortic endothelial cells via targeting Prkaa2, causing endothelial dysfunction and overactive oxidative stress, miR-92a inhibitor can improve the above parameters. Conclusions: miRNA-92a inhibitor could exert an inhibition role on oxidative stress and endothelial dysfunction to improve diabetic ED effectively.

        Effects of 1-MCP on Reactive Oxygen Species, Polyphenol Oxidase Activity, and Cellular Ultra-structure of Core Tissue in ‘Yali’ Pear (Pyrus bretschneideri Rehd.) during Storage

        1-methylcyclopropene (1-MCP) has been shown to delay fruit ripening and senescence during storage. In this study, we investigated the effects of 1.0 μL·L-1 1-MCP on the biochemical changes and cellular ultra-structure of the core tissue in ‘Yali’ pear (Pyrus bretschneideri Rehd.) fruits stored at 25 ± 2°C. Our results showed that 1-MCP treatment significantly decreased the rates of respiration and ethylene production before their climacteric peaks, inhibited the development of core browning, lowered the chlorogenic acid content and polyphenol oxidase activity before the onset of core browning, and preserved high levels of superoxide dismutase activity, ascorbic acid, and glutathione contents. Furthermore, it suppressed the accumulation of hydrogen peroxide, superoxide anion, and malondialdehyde in the core tissue, maintained the integral structure of the cell membrane and organelle, and delayed the accumulation of electron-opaque matters in the vacuoles of core cells. These results suggested that 1-MCP treatment could reduce the injury caused by reactive oxygen, inhibit the oxidation of phenolics, and keep the cell membrane intact in core cells, thus, delaying the core browning in ‘Yali’ pear during storage.

      • PBK/TOPK Expression During TPA-Induced HL-60 Leukemic Cell Differentiation

        Objective: This study concerns expression of PBK/TOPK during differentiation of HL-60 leukemic cells induced by tetradecanoyl phorbol acetate (TPA). Methods: Wright-Giemsa staining was performed to observe morphological changes in the HL-60 cells, and flow cytometry was used to assess the cell cycle and CD11b, CD14, CD13, and CD33 expression. PBK/TOPK levels were determined by Western blot analysis. Results: After treating HL60 cells with $5.1{\times}10^{-9}$ mmol/L of TPA for three days, the number of nitroblue-tetrazolium-positive cells and CD11b, CD13, and CD14 expression increased, whereas the PBK/TOPK levels decreased. Conclusions: TPA can inhibit proliferation and induce differentiation of HL60 cells of the granulocytic or monocytic lineage. PBK/TOPK expression was downregulated during this process, whereas the Pho-PBK/TOPK expression was increased.

        Degradation Characteristics and Metabolic Pathway of 17β-estradiol (E2) by Rhodococcus sp. DS201

        In this study an E2-degrading bacterium was isolated from the activated sludge of a municipal treatment plant that treats the waste from a contraceptive medicineprocessing factory in Beijing, China. Using the observed morphological and physiological features of the bacterium and 16S rRNA sequence analysis, this bacterial strain was identified as Rhodococcus sp. DS201. Using single-factor experiments and orthogonal tests, it was demonstrated that, when strain DS210 bacteria were inoculated into MM medium at an initial concentration of 1 mg/L with an initial pH of 7 and an inoculum amount of 1%, complete degradation of E2 by this strain was achieved within 3 days at 30oC. After strain DS201 had degraded the E2, several E2 metabolites were detected in the culture extracts using high-performance liquid chromatography (HPLC); they were then further identified using HPLC with tandem mass spectrometry (LC-MS/MS). Mass spectrum analysis of the E2 degradation identified the following products: pent- 4-enoic acid; 2-ethyl-3-hydroxy-6-methylcyclohexane-1- carboxylic acid; 3-(7a-methyl-1,5-dioxooctahydro-1H-inden- 4-yl) propanoic acid; and 5-hydroxy-4-(3-hydroxypropyl)- 7a-methyloctahydro-1H-inden-1-one. These products have not previously been reported as parts of a mechanism for microbial E2 degradation and were suspected to be new metabolite products. Therefore, the E2 degradation pathway by strain DS201 is proposed herein.

        長安古都大多表現在其曆史發展中建設基地的遺存,而建築物、構築物構成的空間格局及其所依托的自然地景環境,依然留存至今,타反映了中國傳統景觀文化"籠山水爲苑""相地地形數理"和"君子山水比德"的原型特點和文化寓意。基於這種理念,本文評述華淸池風景區的景觀文化價値以及現狀問題,通過景象空間和賦文景觀文化內涵的營造,保護和展示人工建築與自然山水空間融爲一體的曆史景觀。 Chang`an, an ancient historical city with its construction basement remains. Chinese Traditional Landscape Culture comes from the ancient civilization of the Yellow River and the environment of the central Shaanxi plain forms its ideology characteristic. Basing on the fast urbanization and growth of economies, there is a growing emphasis on Urban Landscape Cultural Heritage. The spatial pattern and traditional landscape should be protected and exhibit because it reflects the original characteristic and implied meaning of Chinese traditional landscape culture. This paper presents the case of Huaqing Palace(華淸宮) scenery area as an example; evaluate the value of its original landscape culture, showing the way of the conservation and regulation.

      • Relation between Ki-67, ER, PR, Her2/neu, p21, EGFR, and TOP II-α Expression in Invasive Ductal Breast Cancer Patients and Correlations with Prognosis

        The aim of the present study was to investigate the expression of the transcription factor Ki-67, ER, PR, Her2/neu, p21, EGFR, and TOP II-${\alpha}$ in the tumor tissue of patients with invasive ductal carcinoma(IDC); in addition, we examined correlations between these markers. Two hundred and sixteen IDC patients, who were not previously been treated with chemo- or radiotherapy, were included in the study. All tumors were grade I-III. Expression of molecular markers was determined by immunohistochemical analysis on paraffin-embedded tissue sections. Follow-up data were collected for 3 months to 10 years and analyzed for tumor recurrence, survival time, and prognostic risk factors. We determined Ki-67 expression correlates with the expression of ER, PR, HER-2, EGFR, and TOP-${\alpha}$, as well as lymph node involvement, high tumor grade, lymphovascular invasion, high tumor stage, and high TNM stage in IDC. Positive Ki-67 expression was a risk factor for rapid tumor recurrence and may help tumor progression, leading to poor prognosis in IDC. Ki-67 was directly correlated with EGFR, TOP II-${\alpha}$, lymph node involvement, high tumor grade, lymphovascular invasion, high tumor stage, and high TNM stage in the hormone receptor subtypes of breast cancer. In triple negative breast cancer, Ki-67 correlated with TOP II-${\alpha}$. Expression of Ki-67 correlated with that of ER, PR, HER-2, EGFR, TOP II-${\alpha}$, and p21. In addition, the biomarker Ki-67 has a role as a prognostic factor and indicates a poor prognosis in IDC.

        Prolyl endopeptidase remodels macrophage function as a novel transcriptional coregulator and inhibits fibrosis

        Macrophages are immune cells crucial for host defense and homeostasis maintenance, and their dysregulation is involved in multiple pathological conditions, such as liver fibrosis. The transcriptional regulation in macrophage is indispensable for fine-tuning of macrophage functions, but the details have not been fully elucidated. Prolyl endopeptidase (PREP) is a dipeptidyl peptidase with both proteolytic and non-proteolytic functions. In this study, we found that Prep knockout significantly contributed to transcriptomic alterations in quiescent and M1/M2-polarized bone marrow-derived macrophages (BMDMs), as well as aggravated fibrosis in an experimental nonalcoholic steatohepatitis (NASH) model. Mechanistically, PREP predominantly localized to the macrophage nuclei and functioned as a transcriptional coregulator. Using CUT&Tag and co-immunoprecipitation, we found that PREP was mainly distributed in active cis-regulatory genomic regions and physically interacted with the transcription factor PU.1. Among PREP-regulated downstream genes, genes encoding profibrotic cathepsin B and D were overexpressed in BMDMs and fibrotic liver tissue. Our results indicate that PREP in macrophages functions as a transcriptional coregulator that finely tunes macrophage functions, and plays a protective role against liver fibrosis pathogenesis.

        Detection of Cucumber Powdery Mildew Based on Spectral and Image Information

        Purpose The aim of this study was to fi nd the optimal detection method for cucumber powdery mildew and improve the identifi cation effi ciency. Methods Image segmentation technology was used to extract spot images and grade classifi cation of powdery mildew. The relationship between powdery mildew spot and spectral refl ectance and intensity was studied. The powdery mildew detection model was established by using qualitative analysis and quantitative prediction methods combined with greenness ( a* ) indices of cucumber leaves. Results There were strong positive correlations between greenness and spectrum in some characteristic bands. Through the extraction of disease spot images and disease classifi cation, it was found that the higher the disease grade of leaves was, the higher the spectral refl ectivity and fl uorescence intensity. In the quantitative prediction model, the R 2 of the NIR spectrum was improved (0.8742) after MSC and SPA, and the eff ect of the fl uorescence spectrum model was not ideal. In the qualitative discriminant model, KNN and ensemble subspace discriminant were obtained for two kinds of spectra, and the identifi cation accuracy of the qualitative model was 97.5% after verifi cation. Conclusion An NIR spectral model can be used for the quantitative prediction of cucumber powdery mildew. The qualitative discriminant model had high accuracy for cucumber powdery mildew.

      • Association of XRCC3 Thr241Met Polymorphisms and Gliomas Risk: Evidence from a Meta-analysis

        The relationship between the X-ray repair cross-complementing group 3 (XRCC3) Thr241Met polymorphism and gliomas remains inclusive or controversial. For better understanding of the effect of XRCC3 Thr241Met polymorphism on glioma risk, a meta-analysis was performed. All eligible studies were identified through a search of PubMed, Elsevier Science Direct, Excerpta Medica Database (Embase) and Chinese Biomedical Literature Database (CBM) before May 2013. The association between the XRCC3 Thr241Met polymorphism and gliomas risk was conducted by odds ratios (ORs) and 95% confidence intervals (95% CIs). A total of nine case-control studies including 3,533 cases and 4,696 controls were eventually collected. Overall, we found that XRCC3 Thr241Met polymorphism was significantly associated with the risk of gliomas (T vs. C: OR=1.10, 95%CI=1.01-1.20, P=0.034; TT vs. CC: OR=1.30, 95%CI=1.03-1.65, P=0.027; TT vs. TC/CC: OR=1.29, 95%CI=1.01-1.64, P=0.039). In the subgroup analysis based on ethnicity, the significant association was found in Asian under four models (T vs. C: OR=1.17, 95%CI=1.07-1.28, P=0.00; TT vs. CC: OR=1.79, 95%CI=1.36-2.36, P=0.00; TT vs. TC/CC: OR=1.75, 95%CI=1.32-2.32, P=0.00; TT/TC vs. CC: OR=1.11,95% CI=1.02-1.20). This meta-analysis suggested that the XRCC3 Thr241Met polymorphism is a risk factor for gliomas, especially for Asians. Considering the limited sample size and ethnicities included in the meta-analysis, further large scale and well-designed studies are needed to confirm our results.
