        Identification and Characterization of a New Alkaline SGNH Hydrolase from a Thermophilic Bacterium Bacillus sp. K91

        est19 is a gene from Bacillus sp. K91 that encodes a new esterase. A comparison of the amino acid sequence showed that Est19 has typical Ser-Gly-Asn-His (SGNH) family motifs and could be grouped into the SGNH hydrolase family. The Est19 protein was functionally cloned, and expressed and purified from Escherichia coli BL21(DE3). The enzyme activity was optimal at 60°C and pH 9.0, and displayed esterase activity towards esters with short-chain acyl esters (C2-C6). A structural model of Est19 was constructed using phospholipase A1 from Streptomyces albidoflavus NA297 as a template. The structure showed an α/β-hydrolase fold and indicated the presence of the typical catalytic triad Ser49-Asp227-His230, which were further investigated by site-directed mutagenesis. To the best of our knowledge, Est19 is a new member of the SGNH hydrolase family identified from thermophiles, which may be applicable in the industrial production of semisynthetic β-lactam antibiotics after modification.

        Characterization and mechanism analysis of graphite/C-doped TiO2 composite for enhanced photocatalytic performance

        Graphite/C-doped TiO2 composite was synthesized through a modified sol–gel reaction. DRS resultsindicated that light absorbance edge of graphite/C-doped TiO2 was clearly red-shifted to visible lightregion by doping with C. SPS results displayed that separation efficiency of photoinduced charge carriersof graphite/C-doped TiO2 were greatly increased by coupling with graphite. Moreover, the photocatalyticperformance of composite was evaluated by the degradation of methyl orange (MO). Results showedthat graphite/C-doped TiO2 exhibited higher photocatalytic activity for MO than that of pristine TiO2 andP25, which was mainly attributed to the high crystallization, large surface hydroxyl groups, intensevisible light absorption and effective separation of photoinduced charge carriers.

        Effects of hydrothermal parameters on the physicochemical property and photocatalytic degradation of bisphenol A of Ti-based TiO2 nanomaterials

        Effects of hydrothermal parameters on morphology, crystal structure, light absorption, separation efficiencyof photo-generated charge carriers, and photocatalytic removal of Bisphenol A (BPA) of Ti-basedTiO2 nanomaterials were systematically investigated. Through changing hydrothermal parameters,TiO2 nanobelts, TiO2 nanosheets and TiO2 nanowires were prepared. With increasing NaOH concentration,hydrothermal temperature, and hydrothermal time, more TiO2 with (101) crystal plane grew onTi substrate, resulting in higher crystallinity. The UV-light absorption enhanced with increasing NaOHconcentration, but decreased with improving hydrothermal temperature, hydrothermal time, and HClconcentration. Variation of UV-light absorption was mainly affected by morphology, and UV-light absorptionof TiO2 nanomaterials with different morphologies was arranged in order ofnanobelts > nanosheets > nanowires. The hydrothermal growth of TiO2 nanomaterials on Ti plate conformedto Ostwald ripening mechanism. Variation trend of photo-generated current was consistent withthat of BPA degradation, they both first increased and then decreased within investigated range. The optimalNaOH concentration, hydrothermal temperature, hydrothermal time, and HCl washing concentrationwere 1 M, 170℃, 28 h, and 0.1 M, respectively. Under this condition, Ti-based TiO2 nanosheets exhibitedthe highest BPA removal efficiency (92.7%), which was due to highly ordered nanosheet structure, goodcrystallinity, appropriate UV-light absorption and high separation efficiency of electron-hole pairs.

        Double-Blind Compact E-cash from Bilinear Map

        Compact E-cash is the first scheme which can withdraw 2l coins within (1) operations and then store them in ( ) bits. Because of its high efficiency, a lot of research has been carried out on its basis, but no previous research pay attention to the privacy of payees and in some cases, payees have the same privacy requirement as payers. We propose a double-blind compact E-cash scheme, which means that both the payer and the payee can keep anonymous while spending. In our scheme, the payer and the bank cannot determine whether the payees of two different transactions are the same one and connect the payee with transactions related to him, in this way, the privacy of the payee is protected. And our protocols disconnect the received coin from previous transaction, then, the coin can be transferred into an unspent coin which belongs to the payee. The proposed e-cash scheme is secure within y-DDHI and LRSW assumption.

        The Complete Genome Sequence of Southern rice black-streaked dwarf virus Isolated from Vietnam

        We determined the complete genome sequence of a Vietnamese isolate of Southern rice black-streaked dwarf virus (SRBSDV). Whole genome comparisons and phylogenetic analysis showed that the genome of the Vietnamese isolate shared high nucleotide sequence identities of over 97.5% with those of the reported Chinese isolates, confirming a common origin of them. Moreover, the greatest divergence between different SRBSDV isolates was found in the segments S1, S3, S4 and S6, which differs from the sequence alignment results between SRBSDV and Rice black streaked dwarf virus (RBSDV), implying that SRBSDV evolved in a unique way independent of RBSDV. This is the first report of a complete nucleotide sequence of SRBSDV from Vietnam and our data provides new clues for further understanding of molecular variation and epidemiology of SRBSDV in Southeast Asia.

        Functional significance of cholesterol metabolism in cancer: from threat to treatment

        Cholesterol is an essential structural component of membranes that contributes to membrane integrity and fluidity. Cholesterol homeostasis plays a critical role in the maintenance of cellular activities. Recently, increasing evidence has indicated that cholesterol is a major determinant by modulating cell signaling events governing the hallmarks of cancer. Numerous studies have shown the functional significance of cholesterol metabolism in tumorigenesis, cancer progression and metastasis through its regulatory effects on the immune response, ferroptosis, autophagy, cell stemness, and the DNA damage response. Here, we summarize recent literature describing cholesterol metabolism in cancer cells, including the cholesterol metabolism pathways and the mutual regulatory mechanisms involved in cancer progression and cholesterol metabolism. We also discuss various drugs targeting cholesterol metabolism to suggest new strategies for cancer treatment.

        Evaluation of the YOLO models for discrimination of the alfalfa pollinating bee species

        Identifying insect pollinators and their roles in mediating pollen flow is critical to understand the potential gene flow risks of insect pollination-dependent crop species, such as alfalfa. This study was conducted to evaluate and compare the feasibility of You Only Look Once (YOLO) version 3 (YOLOv3), YOLOv5, YOLOv7, and YOLO Representation (YOLOR) to discriminate the three most common alfalfa pollinating bee species, including honeybee, bumblebee, and leafcutting bee. The metrics comparison results showed YOLOv3 and YOLOv5 out performed YOLOv7 and YOLOR regarding model precision, recall, F 1 score, and mAP50 values. YOLOv3 and YOLOv5 could successfully discriminate the three different bee species with an accuracy of almost 100% (99.9%, 99.8%, and 100% accuracy for honeybee, bumblebee, and leafcutting bee for the two models, respectively). Comparatively, YOLOv7 could discriminate honeybee with an accuracy of 95% but was more likely to mistakenly discriminate bumblebee and leafcutting bee due to the relatively lower discriminating accuracy (87.3% and 66.2%, respectively). While the values of determined parameters for YOLOR were lower than YOLOv3 and YOLOv5, the higher precision (0.99680) along with recall (0.98721), F 1 (0.99198), mAP50 (0.99323), and mAP50-100 (0.89076) values indicate that this model could be able to obtain a favorable performance in discriminating the three bee species. In summary, the proposed method in this study has the potential for identifying the alfalfa pollinating bee species, studying the bees’ flower-visiting behaviors, evaluating the risks of insect-mediated pollen flow, and thus contributing to the management of genetically engineered (GE) alfalfa transgene flow.
