포항시의 철강공단에 인접한 하천인 냉천을 대상으로 식물상과 식생을 비롯하여 저서성 대형무척추동물의 군집구조와 하천의 일반 현황을 조사하였다. 이 조사 결과를 바탕으로 냉천의 특성에 적절한 친환경적 정비 방안을 제시하였다. Flora, vegetation, community structure of benthic invertebrates as well as general condition of Naengcheon stream near by the Pohang industrial complex have been investigated, On the basis of this investigation, a scheme for environment-friendly management is suggested.

In the present, batch test was conducted to evaluate the neutralization and adsorption of heavy metals from wastewater using waste egg shells. Neutralization and removal rate of heavy metals were excellent in the increase of waste concrete amounts and a small size. It seemed that adsorption efficiencies of heavy metals were influenced by solubility. As a result on the experiments of Freundlich isotherm, the adsorption capacities(k) were Cr 3.11, Cu 2.61, Mn 3.02 and Pb 0.95, respectively and the adsorption intensities(1/n) were Cr 0.35, Cu 0.44, Mn 0.4 and Pb 0.41, respectively. In view of these results, it showed that wastes containing the similar compositions as waste egg shells could utilize the neutralization and adsorption of heavy metals in wastewater.

A FET type micro CO_(2) sensor based on a Severinghaus type arrangement has been prepared and its response characteristics were examined. The sensor was fabricated by formation of hydrogel layer containing bicarbonate/chloride ions on the PH-ISFET and the Ag/AgCl reference electrode prepared by the thermal evaporation and chemical chloridation methods. And then gas-permeable membrane was dip-coated on the hydrogel layer of the sensor. Gas-permeable membranes were fabricated with silicon rubber and polyurethane dissolved in toluene and THF, respectively. The CO_(2) sensor with the silicon rubber membrane showed better sensitivity and lower detection limit than the CO_(2) sensor with the polyurethane membrane for dissolved CO_(2). The sensor with silicon rubber membrane at optimal conditions showed a good response: linear range of 1.60X10^(-4) ~ 4.40X10^(-2) M CO_(2), sensitivity of 57.30 mV/decade, and detection limit of 7.50x10^(-4) M.

민주지산 일대의 곤충상 파악을 위하여 1997년 7월부터 1997년 10월까지 조사를 실시하였다. 그 결과 총 18목 151과 540종이 조사되었다. 딱정벌레목의 곤충들이 176종으로 가장 풍부하게 나타났으며, 다음으로 나비목, 노린재목, 벌목 등의 순으로 조사되었다. 또한 보호가 필요한 종, 감소종, 및 환경부 보존 지정 곤충을 확인하였다. Insect fauna of Mt. Minjujisan was investigated from July to October in 1997. As the result, a total of 540 species of 151 families under 18 orders were identified in thr present of study. The number of species were the most abundantly revealed in the order Coleoptera, 176 species and the Order Lepidoptera, Order Hemiptera, Order Hymenoptera were followed in order.

한천, 키틴, 셀룰로오스, 자일란, 만난과 같은 복합다당류들에 대한 분해능을 나타내는 효소들을 생산하는 해양미생물을 홍조류인 Porphyra dentata로부터 분리하였다. 분리균 BK1은 그람음성, 호기성 간균으로 DNA의 G+C함량은 51.6 mol%를 나타내었다. 주요 isoprenoid quinine 구성 성분은 ubiquinone-8로 확인되었고, 주요 세포 지방산은 C16:0, C16:1 w6c, C18:1 w7c로 밝혀졌다. 16S rRNA서열의 비교분석 결과는 분리균 BK1이 Pseudomonas 속의 일원인 것으로 확인되었다. 이러한 결과를 바탕으로 분리균 BKl은 Pseudomonas sp. BKl으로 명명하였다. A marine bacterium (strain BKl) that produces extracellular enzymes capable of decomposing complex polysac-charides, such as agar, chitin, carboxymethylcellulose, xylan and mannan, was isolated from the marine red alga Porphyra dentata. Strain BKl was gram-negative, aerobic, catalase- and oxidase-positive, polarly flagellated bacilli that produce gelatinase and urease, but not decarboxylases. The G+C content of the DNA was 51.6 mol%. The major isoprenoid quinone component was identified as an ubiquinone-8, and the major cellular fatty acids were C16:0, C16:1 w6c and C18:1 w7c. Comparative 16S rRNA sequence analysis placed strain BK1 with members of the genus Pseudomonas. On the basis of phenotypic and genotypic data, the strain BK1 was shown to be a member of the subgroup of Pseudomonas, and named as Pseudomonas sp. BK1.
Production and Characterization of Alkaline Protease of Micrococcus sp. PS-1 Isolated from Seawater

본 연구는 부산 인근의 해수욕장에서 얻은 해수에서 protease를 생산하는 균주를 분리하여 동정하고 균주의 배양학적인 특성과 protease의 효소학적 특성을 확인하였다. 해수에서 분리한 protease를 생산하는 미생물은 16S rDNA sequencing을 통해 Micrococcus sp. PS-1으로 동정하였다. Protease 생산의 최적조건은 2% skim milk와 1% NaCl이 포함된 pH 7.0의 LB배지에 48시간 배양이었다. 효소의 부분정제를 위해 ultrafiltration 과 acetone 침전법을 사용하였고, zymography를 통해 분자량이 35.0 kDa과 37.5 kDa인 protease를 확인하였다. 또한 효소의 최적 활성은 pH 9.0와 37℃에서 나타났고, 효소는 pH 8.0에서 11.0까지, 25℃에서 37℃까지 80% 이상의 효소활성이 유지되어 안정한 것으로 확인되었으며 PMSF, EDTA 처리시 protease가 저해되는 것을 통해 alkaline metallo-serine protease로 확인되었다. The purpose of this research was to investigate the production and characterization of alkaline protease from Micrococcus sp. PS-1 newly isolated from seawater. Micrococcus sp. PS-1 was grown in Luria- Bertani (LB) medium. Its optimal temperature and pH for growth were 30°C and 7.0, respectively. The effect of nitrogen sources was investigated on optimal enzyme production. A high level of alkaline protease production occurred in LB broth containing 2% skimmed milk. The protease was purified in a 3-step procedure involving ultrafiltration, acetone precipitation, and dialysis. The procedure yielded a 16.43-purification fold, with a yield of 54.25%. SDS-PAGE showed that the enzyme had molecular weights of 35.0 and 37.5 kDa. Its maximum protease activity was exhibited at pH 9.0 and 37°C, and its activity was stable at pH 8.0?11.0 and 25?37°C. The protease activity was strongly inhibited by PMSF, EDTA, and EGTA. Taken together, the results demonstrate that the protease enzyme from Micrococcus sp. PS-1 probably belongs to a subclass of alkaline metallo-serine proteases.

To identify genes involved in the decolorization of crystal violet, we isolated random mutants generated by transponson insertion in crystal violet-declorizing bacterium, Citrobacter sp. The resulting mutant bank yielded mutants with six distinct phenotypes, and Southern hybridization with a Tn5 fragment as a probe showed a single hybridized with six distinct phenotypes, and Southern hybridization with a Tn5 fragment as a probe showed a single hybridized band in the mutants Ctg 2, 5 an 6, whereas two and three bands were detected in Ctg1, 4 and 3, respectively. Tn5-inserted genes were isolated and the DNA sequence flanking Tn5 was determined. From comparison with a sequence database, putative protein product encoded by ctg 5 was identified as E. coli maltose transproter(Mal G) homolog, whereas the deduced amino acid sequence of the other ctg genes did not show any significant similarity with any DNA or protein sequency. Therefore, these results indicate that the other ctg genes except ctg 5 encode new proteins responsible for decolorization of crystal violet.
