        Isolation and Anti-Inflammatory Activity of Bakuchiol from Ulmus davidiana var. japonica

        The bark of the root and stem of Ulmus davidiana var. japonica has been used as a traditional Korean medicine to treat inflammatory disorders. This plant reportedly exhibits antioxidant, anticancer, and anti-inflammatory effects. A search for biologically active compounds in U. davidiana var. japonica extracts yielded bakuchiol, which we structurally identified on the basis of spectral data, including two-dimensional nuclear magnetic resonance spectroscopy and distortionless enhancement by polarization transfer. In our study, bakuchiol (50μM) inhibited lipopolysaccharide-induced nitric oxide and prostaglandin E2 production in RAW 264.7 macrophages by 53.7% and 84.2%, respectively. These results suggested that bakuchiol is one of the potent anti-inflammatory components of U. davidiana var. japonica.

      • 김치를 이용한 타블렛 조성물이 고지방식이로 유도된 흰쥐의 비만 억제에 미치는 효과

        Anti-obesity effects of kimchi tablet composition (KTC) were investigated with male Sprague-Dawley rats fed high-fat diet for 8 weeks. Fifty rats were classified to 4 groups: normal diet (ND), high fat diet (HFD), HFD + KTC 5% and HFD + KTC 10% for feeding experiments. This research showed that the final weight, weight gain, food efficiency ratio and organ weight were decreased by the addition of KTC compared to those of HFD group. Amount of subcutaneous fat of KTC groups were 13.6% and 21.3%, respectively. The plasma triglyceride concentration of KTC groups was lower than that of high fat diet group. Plasma cholesterol concentration of all four groups were not significantly different. HDL-cholesterol concentration of KTC groups was higher than those of high fat diet group. In addition, triglyceride and cholesterol concentrations in the liver and heart were reduced by the addition of KTC. Moreover, triglyceride concentration of fecal and hepatic HMG-CoA reductase activity were increased in KTC group. These results suggested that KTC might be useful for obesity control and good source of functional kimchi tablet.

        김치를 이용한 타블렛 조성물이 고지방식이로 유도된 흰쥐의 비만 억제에 미치는 효과

        동결건조된 김치분말을 주성분으로 하여 부형제로 glucose, 고춧가루, 식이섬유소 등을 혼합하여 제조된 김치 타블렛 제조용 조성물(KTC)의 항비만 효과를 알아보기 위하여 4주령의 SD계 수컷 흰쥐를 식이에 따라 정상대조군, 고지방식이군, 고지방 식이에 KTC 5%와 10% 첨가군의 4군으로 나누어 8주간 사육하고 체중, 피하지방, 혈당, 혈 중 지질농도, 간조직의 지질 농도와 HMG-CoA reductase 활성, 분변으로의 지질 배설능 등을 측정한 결과는 다음과 같다. 고지방 식이에 KTC 첨가량이 늘어날수록 흰쥐의 혈중 중성지질, 총콜레스테롤치를 농도 의존적으로 감소시켜 혈액내 지방의 축적을 저해하였고, 복부 피하지방의 양은 KTC 5%, 10% 첨가군이 고지방군에 비해 13.6%, 21.3% 감소시켰다(p<0.05). 또한 간과 심장, 고환의 중성지방, 콜레스테롤 농도는 KTC 첨가군이 고지방식이군에 비해 농도 의존적으로 감소하였고(p<0.05), HMG-CoA reductase 활성 및 혈당 농도를 고지방식이군에 비해 유의적으로 감소시켜 KTC가 흰쥐의 비만을 억제함을 확인할 수 있었다.         차나무(Camellia sinensis) 추출물이 아급성 알코올 투여 마우스의 항산화 및 알코올 분해 효소 활성에 미치는 영향

        Mouse에 알코올을 투여한 결과 항산화 효소의 활성이 감소하는 것을 볼 수 있으며 ADH 및 ALDH활성도 모두 감소하였다. 녹차, 홍차, 우롱차 및 보이차의 알코올 섭취 mouse에서의 항산화 및 알코올 분해효소에 미치는 영향을 측정하여 비교한 결과 네 가지 차 모두에서 투여하지 않은 군에 비하여 항산화 및 알코올 분해 촉진효과를 나타내었으며 그 중 보이차가 간에서의 SOD 및 GR활성을 크게 증가시켰고 MDA함량을 감소시켰다. 또한 우롱차는 혈액에서의 SOD 및 GSH-PX활성을 크게 증가시켰고 MDA함량을 감소시켰으며 녹차와 홍차의 항산화 효과는 이들에 비하여 비교적 낮았다. 또한 알코올 분해에 관여하는 효소인 ADH, ALDH 활성은 녹차를 투여한 군에서 매우 높게 나타났으며 녹차 다음으로는 우롱차가 ADH 활성측정에서, 보이차가 ALDH 활성측정에서 높은 수치를 보였고 홍차 투여 군에서 가장 낮았다. 따라서 네 가지 차 중 우롱차와 보이차를 섭취하였을 때 높은 항산화 효과를 기대할 수 있으며 알코올 분해에는 녹차의 섭취가 가장 효율적일 것으로 사료된다. This study was conducted to investigate the effects of four kinds of tea (Camellia sinensis) extracts on the antioxidant defense systems as well as the activities of alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) in ethanol administered ICR mice. According to the results, treatment with puerh tea significantly increased the superoxide dismutase activity and glutathion reductase activity in liver. In addition, the group treated with oolong tea exhibited higher superoxide dismutase activity and glutathion reductase activity in serum than those of puerh tea, green tea and black tea treated groups. The oolong tea and puerh tea also reduced malondealdehyde contents in both liver and serum. These results suggested that puerh tea and oolong tea were the most effective against alcohol-induced oxidative damage among the Camellia sinensis teas. On the other hand, in the measurement of alcohol break-down enzyme activities, the group treated with green tea exhibited the highest hepatic ADH and ALDH activities, suggesting that the group treated with green tea might be useful for alcohol down-regulation.

        Immunomodulatory Effect of Scutellaria radix in Balb/c Mice

        Scutellaria radix (SR), the root of Scutellaria baicalensis is one of the most widely used herbal medicines against bacterial infections of the respiratory and gastrointestinal tract, and various inflammatory diseases. No previous studies have investigated SR-induced alterations in the immune system of Balb/c mice. In the present study, SR was administered by gavage for 14 days at 0,50, 100, 200 and 400 ㎎/㎏ to female Balb/c mice. Immunomodulatory activity was evaluated by assessment of body and organ weight, lymphocyte blastogenesis, antibody-forming cell (AFC) assay and lymphocyte subpopulation by flowcytometry. Hematological and histopathological changes were also assessed. Even though the change of body weight was not observed in SR-administered group, spleen weight and cellularity of spleen are slightly increased at the dose groups of SR (100, 200 and 400 ㎎/㎏). The T lymphocyte blastogenesis by concanavalin A (Con A) was slightly increased, but B-lymphocyte blastogenesis by lipopolysaccharide (LPS) was significantly increased in SR-treated group. In the case of lymphocyte subpopulation, the population of CD3⁺ T cells and CD3⁺ T cell/CD45R⁺ B cell ratio were significantly increased at high dose of SR-treated mice. However, there were no significant changes in the hematological findings of SR-treated mice. The antibody response to T lymphocyte dependent antigen, sheep red blood cell (SRBC) was increased at all doses of SR-treated group. These results indicate that SR could induce the immunomodulation in Balb/c mice.

        백삼과 홍삼의 페놀성 성분 함량 및 멜라닌 생성억제효과

        백삼과 홍삼의 quercetin, cinnamic acid, p-coumaric acid, maltol, esculetin 함량을 측정한 결과 백삼에는 cinnamic acid가 가장 높은 함량을 보였고, 홍삼에는 증숙과정 중에 생성되는 특이 성분인 maltol이 가장 높은 함량을 나타냈다. 인삼시료 중 백삼의 ethyl acetate 분획물은 melan-a 세포주에서의 세포생존율 대비 멜라닌 생성억제활성 측정, UV-흡수도 측정, tyrosinase 억제활성 측정에서 모두 우수한 활성을 나타내었으며, 이의 활성 성분으로는 cinnamic acid가 주요한 역할을 하는 것으로 판단되어진다. 한편, maltol을 다량 함유한 홍삼 ethyl acetate 분획물은 UV-흡수 활성을 보였으나 다른 유의적인 활성을 나타내지 않았다. 따라서 인삼소재 중 백삼 ethyl acetate 분획물은 피부미백소재로써의 기능을 가지는 것으로 판단된다. Quantitative difference in five phenolic acids between white and red ginsengs was measured in this study. As the results, white ginseng has higher contents of cinnamic acid, quercetin and p-coumaric acid than red ginseng. Maltol was mainly included in red ginseng. These five compounds were recently reported to have tyrosinase inhibitory effects. These reports led us to investigate the de-pigmenting effect of ginseng products. In our examination of effect on tyrosinase activity, UV-protection and melanin production in melan-a cells, ethylacetate fraction of white ginseng extract and cinnamic acid showed potent de-pigmenting properties. The results indicated that white ginseng might be useful as skin whitening material and cinnamic acid proved to be one of active ingredient.
