        한국인 및 중국 한족 정신분열병 환자의 5-HT2A 수용체 유전자 -1438A/G 다형성

        Objectives : The purpose of the present study was to investigate the association between -1438A/G polymorphism of 5-HT2A receptor gene and schizophrenia in Korean and Han Chinese population. Methods : A sample of 184 Korean patients with schizophrenia and 96 Korean healthy normal controls and 96 Han Chinese patients with schizophrenia and 96 Han-Chinese healthy normal controls were genotyped for a single nucleotide polymorphism with in 5-HT2A receptor gene (promoter region, A-1438G) by Msp I Resthction Fragment Length Polymorphism (RFLP). Results : There was no difference in allelic frequencies and genotype frequencies of -1438A/G polymorphism between Korean schizophrenics and controls (p=0.13) and Han Chinese schizophrenics and controls (p=0.40). Also, -1438A/G Poly-morphism did not show ethnical difference between Korean and Han Chinese controls. The Scale for the Assessment of Negative Symptoms (SANS) scores showed no significant differences between genotypes of -1438A/G polymorphism in both of Korean and Han Chinese schizophrenics. Conclusion : These results suggest that -1438A/G polymorphism of the 5-HT2A receptor gene is not causally related to the development of schizophrenia in Korean and Han Chinese population, and there no ethnic difference between Korean and Han Chinese population.

        Albumin-to-Fibrinogen Ratio as an Independent Prognostic Parameter in Untreated Chronic Lymphocytic Leukemia: A Retrospective Study of 191 Cases

        Purpose Chronic lymphocytic leukemia (CLL) is one of the most frequent type of B-cell chronic lymphoproliferative disorders and chronic inflammation takes part in the development of CLL. However, there has been no valid immune biomarker to predict the prognosis of untreated CLL patients. Materials and Methods In this retrospective study, we analyzed the clinical correlations and prognostic value of albumin-to-fibrinogen ratio (AFR) detected at diagnosis in 191 CLL patients. Results The cut-off value of AFR was 9.7 calculated by X-tile. Patients who were more than 65 years old were often accompanied by low level of AFR (p < 0.001). Survival analysis showed that patients with low level of AFR had shorter overall survival (OS) than patients with high level of AFR (p < 0.001). Multivariate analysis illustrated that AFR had a negative impact on OS (p=0.003) and was independent of parameters involved in CLL international prognostic index and other prognostic markers such as CD38 and ZAP-70. Conclusion These data provide a comprehensive view of AFR and shows that AFR at diagnosis is an adverse prognostic factor in untreated CLL patients.

        The Numerical Simulation of Unsteady Flow in a Mixed flow Pump Guide Vane

        In order to investigate the characteristics of unsteady flow in a mixed flow pump guide vane under the small flow conditions, several indicator points in a mixed flow pump guide vane was set, the three-dimensional unsteady turbulence numerical value of the mixed flow pump which is in the whole flow field will be calculated by means of the large eddy simulation (LES), sub-grid scale model and sliding mesh technology. The experimental results suggest that the large eddy simulation can estimate the positive slope characteristic of head & capacity curve. And the calculation results show that the pressure fluctuation coefficients of the middle section in guide vane inlet will decrease firstly and then increase. In guide vane outlet, the pressure fluctuation coefficients of section will be approximately axially symmetrical distribution. The pressure fluctuation minimum of section in guide vane inlet is above the middle location of the guide vane suction surface, and the pressure fluctuation minimum of section in which located the middle and outlet of guide vane. When it is under the small flow operating condition, the eddy scale of guide vane is larger, and the pressure fluctuation of the channel in guide vane being cyclical fluctuations obviously which leads to the area of eddy expanding to the whole channel from the suction side. The middle of the guide vane suction surface of the minimum amplitude pressure fluctuation to which the vortex core of eddy scale whose direction of fluid's rotation is the same to impeller in the guide vane adhere.

        Investigation of surface pressures on CAARC tall building concerning effects of turbulence

        This paper presents an experimental investigation on the surface pressures on the CAARC standard tall building model concerning the effects of freestream turbulence. Two groups of incidence turbulence are generated in the wind tunnel experiment. The first group has an approximately constant turbulence intensity of 10.3% but different turbulence integral scale varying from 0.141 m to 0.599 m or from 0.93 to 5.88 in terms of scale ratio (turbulence integral scale to building dimension). The second group presents similar turbulence integral scale but different turbulence intensity ranging from 7.2% to 13.5%. The experimental results show that the mean pressure coefficients on about half of the axial length of the side faces near the leading edge slightly decrease as the turbulence integral scale ratio that is larger than 4.25 increases, but respond markedly to the changes in turbulence intensity. The root-mean-square (RMS) and peak pressure coefficients depend on both turbulence integral scale and intensity. The RMS pressure coefficients increase with turbulence integral scale and intensity. As the turbulence integral scale increases from 0.141 m to 0.599 m, the mean peak pressure coefficient increases by 7%, 20% and 32% at most on the windward, side faces and leeward of the building model, respectively. As the turbulence intensity increases from 7.2% to 13.5%, the mean value of peak pressure coefficient increases by 47%, 69% and 23% at most on windward, side faces and leeward, respectively. The values of cross-correlations of fluctuating pressures increase as the turbulence integral scale increases, but decrease as turbulence intensity increases in most cases.

        Cavitation cloud dynamic characteristics of dual-chamber self-excited oscillatory waterjet

        Aiming to enhance self-excited oscillating cavitation jet performance, the effect of the dual-chamber nozzlestructure on the jet dynamical characteristics was designed and investigated. With high-speed camera technology,the cavitation phenomenon was investigated to analyze the area pattern and shedding period of the cavitation cloudunder different nozzle structures. The results showed that the dual-chamber nozzle significantly improved the jet cavitationstrength, and the cavitation cloud area increased by 76% and decreased the shedding period by 90% comparedwith the single-chamber nozzle. In the upstream chamber, the upper shrinkage ratio had a more drastic effect on thecavitation cloud area and shedding frequency than the lower shrinkage ratio with a more sensitive effect on the sheddingfrequency. In the downstream chamber, the outlet diameter ratio and chamber diameter were more sensitive tothe regulation of cavitation cloud shedding frequency and area, respectively, with the optimal regulation at the outletdiameter ratio of 1 and chamber length of 6 mm. The chamber diameter modulated the cavitation cloud most drasticallywith a comprehensive performance optimum at 12mm, which the area fluctuation reached 76.8%. The resultsprovide a basis for further research and application of dual-chamber nozzles.

      • Decreased Expression of FADS1 Predicts a Poor Prognosis in Patients with Esophageal Squamous Cell Carcinoma

        FADS1 (fatty acid desaturase 1) plays a crucial role in fatty acid metabolism, and it was recently reported to be involved in tumorigenesis. However, the role of FADS1 expression in esophageal squamous cell carcinoma (ESCC) remains unknown. In the current study, we investigated the expression and clinical pathologic and prognostic significance of FADS1 in ESCC. Immunohistochemical analyses revealed that 58.2% (146/251) of the ESCC tissues had low levels of FADS1 expression, whereas 41.8% (105/251) exhibited high levels of FADS1 expression. In positive cases, FADS1 expression was detected in the cytoplasm of cells. Correlation analyses demonstrated that FADS1 expression was significantly correlated with tumor location (p=0.025) but not with age, gender, histological grade, tumor status, nodal status or TNM staging. Furthermore, patients with tumors expressing high levels of FADS1had a longer disease-free survival time (p<0.001) and overall survival time (p <0.001). Univariate and multivariate analyses revealed that, along with nodal status, FADS1 expression was an independent and significant predictive factor (p<0.001). In conclusion, our study suggested that FADS1 might be a valuable biomarker and potential therapeutic target for ESCC.

      • Matrine Reduces Proliferation of Human Lung Cancer Cells by Inducing Apoptosis and Changing miRNA Expression Profiles

        Matrine, a main active component extracted from dry roots of Sophora flavecens, has been reported to exert antitumor effects on A549 human non-small lung cancer cells, but its mechanisms of action remain unclear. To determine effects of matrine on proliferation of A549 cells and assess possible mechanisms, MTT assays were employed to detect cytotoxicity, along with o flow cytometric analysis of DNA content of nuclei of cells following staining with propidium iodide to analyze cell cycle distribution. Western blotting was performed to determined expression levels of Bax, Bcl-2, VEGF and HDAC1, while a microarray was used to assessed changes of miRNA profiles. In the MTT assay, matrine suppressed growth of human lung cancer cell A549 in a dose- and timedependent manner at doses of 0.25-2.5 mg/ml for 24h, 48h or 72h. Matrine induced cell cycle arrest in G0/G1 phase and decreased the G2/M phase, while down-regulating the expression of Bcl2 protein, leading to a reduction in the Bcl-2/Bax ratio. In addition, matrine down regulated the expression level of VEGF and HDAC1 of A549 cells. Microarray analysis demonstrated that matrine altered the expression level of miRNAs compared with untreated control A549 cells. In conclusion, matrine could inhibit proliferation of A549 cells, providing useful information for understanding anticancer mechanisms.

        Effect of Nd Substitution for Mischmetal on Magnetic Properties in (MM,Nd)-Fe-B Ribbons

        Mischmetal-based permanent magnets of (MM,Nd)12Fe82B6 were prepared by melt-spinning method via varying the relative content of mischmetal and Nd. The coercivity is low, and the squareness of hysteresis loop is poor in MM12Fe82B6 ribbons. The x-ray diffraction pattern, maximum of δm value as well as the temperature dependence of magnetization indicates that the partial substitution of Nd for MM could improve the crystallinity in the mischmetal-based magnets, and both the coercivity and squareness of hysteresis loop increase with Nd substitution for mischmetal. The increase of squareness should be partially attributed to the improvement in the crystallinity of R₂Fe14B crystal phase. For more than 4 at.% Nd substitution the coercivity is not less than that in corresponding (Ce,Nd)-based magnets, and the coercivity of 7.81 kOe and the maximum energy product of 15.41 MGOe were obtained in MM8Nd₄Fe82B6 ribbons. These investigations show that it is reasonable to use the low cost mischmetal to prepare the resource-saving rare-earth magnets, and that optimizing the addition amount of Nd is necessary to improve the crystallinity and enhance the magnetic properties.
