        Optimization of preparation and properties of Gardenia yellow pigment-loaded alginate beads

        Gardenia yellow pigment (GYP) loaded alginate beads were prepared by the ionic gelation technique, and the preparation parameters were optimized by response surface methodology for high encapsulation efficiency. The optimized parameters were alginate concentration of 3.3%, CaCl2 concentration of 2.4%, and GYP concentration of 3.2mg/mL, under which the encapsulation efficiency was 73.61%. The surface morphology and bead size analysis showed that the GYP-loaded alginate beads had a roughly spherical morphology with a wrinkled surface, and their average diameter was 0.87 mm. In vitro release test revealed that the GYP release had a pH-dependent release profile and a two-step release process. The Rigter-Peppas model was the most proper model to assess the GYP release from alginate beads. The release mechanism of GYP at pH 1.2 and 7.4 was non-Fickian transport and case-II transport, respectively. The 2,2-diphenyl-1-picrylhydrazyl assay indicated that the encapsulated GYP had effectively maintained 82.56% of the antioxidant activity.

        Chronic Kidney Disease and Associated Cardiovascular Risk Factors in Chinese with Type 2 Diabetes

        Background: To determine the frequency of chronic kidney disease (CKD) and its associated risk factors in Chinese type 2 diabetic patients, we conducted a cross-sectional study in Nanjing, China, in the period between January 2008 and December 2009. Methods: Patients with type 2 diabetes under the care by Jiangsu Province Official Hospital, Nanjing, China were invited for assessment. CKD was defined as the presence of albuminuria or estimated glomerular filtration rate <60 mL/min/1.73 m2. Albuminuria was defined as urinary albumin-to-creatinine ratio ≥30 mg/g. Results: We recruited 1,521 urban Chinese patients with type 2 diabetes (mean age, 63.9±12.0 years). The frequency of CKD and albuminuria was 31.0% and 28.9%, respectively. After adjusted by age and sex, hypertension, anemia and duration of diabetes were significantly associated with CKD with odds ratio (95% confidence interval) being 1.93 (1.28 to 2.93), 1.70 (1.09 to 2.64), and 1.03 (1.00 to 1.06), respectively. Conclusion: In conclusion, CKD was common in the urban Nanjing Chinese with type 2 diabetes. Strategies to prevent or delay progression of kidney disease in diabetes should be carried out at the early disease course of type 2 diabetes.

        Analysis of the Optical Constants of Aluminum-doped Zinc-oxide Films by Using the Single-oscillator Model

        A group of aluminum-doped zinc-oxide (AZO) films were prepared on glass substrates by directcurrent reactive magnetron sputtering at substrate temperatures (Ts) ranging from 170 ℃ to 210 ℃. The optical constants of the AZO films defined by using Caughy model were fitted in terms of a two-layer model by using the measured spectroscopic ellipsometric parameters. The refractive index dispersion data below the interband absorption edge of the AZO films were analyzed by using a single-oscillator model. The optical energy gap as-fitted by using the single-oscillator model demonstrated a blue and a red shift as T<SUB>s</SUB> increased from 170 ℃ to 200 ℃ and above 200 ℃, respectively. This could be attributed to a change in the free electron concentration, which was related to a change in the effective Al-doping efficiency. The calculated parameter β, related to the crystalline structure, indicated that the as-deposited AZO films fell into an ionic class even though β had a slight deviation from the ionic value. The β of the as-deposited AZO film at 200 ℃ largely remained in the range of ionic values, indicating a minimum deviation from the wurtzite structure. This denoted that Al is a very effective substitute for the zinc sites. Additionally, the calculated plasma frequency, hw<SUB>p</SUB>, remained in the violet region.

      • MAGED4 Expression in Glioma and Upregulation in Glioma Cell Lines with 5-Aza-2'-Deoxycytidine Treatment

        Melanoma-associated antigen (MAGE) family genes have been considered as potentially promising targets for anticancer immunotherapy. MAGED4 was originally identified as a glioma-specific antigen. Current knowledge about MAGED4 expression in glioma is only based on mRNA analysis and MAGED4 protein expression has not been elucidated. In the present study, we investigated this point and found that MAGED4 mRNA and protein were absent or very lowly expressed in various normal tissues and glioma cell line SHG44, but overexpressed in glioma cell lines A172,U251,U87-MG as well as glioma tissues, with significant heterogeneity. Furthermore, MAGED4 protein expression was positively correlated with the glioma type and grade. We also found that the expression of MAGED4 inversely correlated with the overall methylation status of the MAGED4 promoter CpG island. Furthermore, when SHG44 and A172 with higher methylation were treated with the DNA demethylating agent 5-aza-2'-deoxycytidine (5-AZA-CdR) reactivation of MAGED4 mRNA was mediated by significant demethylation in SHG44 instead of A172. However, 5-AZA-CdR treatment had no effect on MAGED4 protein in both SHG44 and A172 cells. In conclusion, MAGED4 is frequently and highly expressed in glioma and is partly regulated by DNA methylation. The results suggest that MAGED4 might be a promising target for glioma immunotherapy combined with 5-AZA-CdR to enhance its expression and eliminate intratumor heterogeneity.

        Antinociceptive Effects of Prim-O-Glucosylcimifugin in Inflammatory Nociception via Reducing Spinal COX-2

        We measured anti-nociceptive activity of prim-o-glucosylcimifugin (POG), a molecule from Saposhnikovia divaricate (Turcz) Schischk. Anti-nociceptive or anti-inflammatory effects of POG on a formalin-induced tonic nociceptive response and a complete Freund``s adjuvant (CFA) inoculation-induced rat arthritis pain model were studied. Single subcutaneous injections of POG produced potent anti-nociception in both models that was comparable to indomethacin analgesia. Anti-nociceptive activity of POG was dose-dependent, maximally reducing pain 56.6% with an ED50 of 1.6 mg. Rats given POG over time did not develop tolerance. POG also time-dependently reduced serum TNFα, IL-1β and IL-6 in arthritic rats and both POG and indomethacin reduced spinal prostaglandin E2 (PGE2). Like indomethacin which inhibits cyclooxygenase-2 (COX-2) activity, POG dose-dependently decreased spinal COX-2 content in arthritic rats. Additionally, POG, and its metabolite cimifugin, downregulated COX-2 expression in vitro. Thus, POG produced potent anti-nociception by downregulating spinal COX-2 expression.

        Web Service technologies make the automation of business activities that are distributed across multiple enterprises possible. Existing extended transaction protocols typically resort to compensation actions to regain atomicity and consistency. A reservation-based transaction protocol is proposed to reduce high compensation risk. However, for a serial long running transaction processing, the resource that is reserved in the early stage may be released due to resource holding time expires. Therefore, our analysis theoretically illustrates a scheduling scheme that tries to prevent the loss of resource holding as well as gain an optimized execution plan with minimum compensation cost. In order to estimate cost of different schedules, we set up a costing model and cost metric to quantize compensation risk.

        CD4+CD25+regulatory T cells (Tregs) play a key role in regulation of immnue response and maintenance of self-tolerance. Studies have found Tregs could suppress tumor-specific T cell-mediated immune response and promote cancer progression. Depletion of Tregs can enhance antitumor immunity. Dendritic cells (DCs) are professional antigen-presenting cells and capable of activating antigen-specific immune responses, which make them ideal candidate for cancer immunotherapy. Now various DC vaccines are considered as effective treatment for cancers. The aim of this study was to evaluate variation of Tregs in BALB/C mice with hepatocellular carcinoma and investigate the interaction between tumor-derived Tregs, effector T cells (Teff) and splenic DCs. We found the percentages of Tregs/CD4+ in the peripheral blood of tumor-bearing mice were higher than in normal mice. Tumor-derived Tregs diminished the up-regulation of costimulatory molecule expression on splenic DCs, even in the presence of Teff cells and simultaneously inhibited IL-12 and $TNF-{\alpha}$ secretion by DCs.

        Genome‑wide identification of GMP genes in Rosaceae and functional characterization of FaGMP4 in strawberry (Fragaria × ananassa)

        Background GDP-D-mannose pyrophosphorylase (GMP) is one of the key enzymes determining ascorbic acid (AsA) biosynthesis. However, little information about GMP genes is currently available for the Rosaceae species, especially in the AsA-riched cultivated octoploid strawberry (Fragaria × ananassa). Objective To identify the all the GMP genes in Rosaceae, as well as the predominant homologues and the role of GMP genes in strawberry AsA accumulation. Methods In the present study, we performed genome-wide identifcation and comprehensive analysis of the duplicated GMP genes in strawberry and other Rosaceae species by bioinformatics methods, the expression of the GMP genes from cultivated strawberry (Fragaria × ananassa, FaGMP) was specifcally analyzed by qPCR. Finally, the FaGMP4 was transiently overexpressed in strawberry to estimate the role of GMP in regulating AsA accumulation in strawberry. Results As results, a total of 28 GMP genes were identifed in the fve Rosaceae species. The origins of duplication events analysis suggested that most GMP duplications in Rosaceae species were generated from whole genome duplication (WGD). The Ka/Ks ratio suggested that FaGMP genes underwent a stabilization selection. qPCR based expression analysis showed diferent patterns of FaGMP paralogs during fruit ripening, while FaGMP4 expressed higher in the variety containing higher AsA. Overexpression of FaGMP4 in strawberry signifcantly enhanced AsA accumulation. Furthermore, the expression of FaGMP4 under the treatment of blue and red light was largely increased in leaves while signifcantly inhibited in fruit. These results revealed the vital role of FaGMP4 in regulating AsA in strawberry.

        An Improved Reservation-based Protocol for Timely Web Service Transactions

        웹서비스 트랜잭션은 다원분산 기업간 장기수행, 완결합 그리고 복합서비스들로 구성되어 있다. 기존 트랜잭션 규약은 보상을 통하나 바람직하잖은 비용으로 전체적으로 일관성을 유지한다. 따라서 기존 연구들은 자원 대기시간 및 보상 위기를 줄이기 위하여 자원을 보존하도록 제안했었다. 반면에 예약은 또한 실질적인 잠금과 수행성능을 개선하는 병행충돌들을 가져온다. 본 논문에서 개선된 시간제약으로 예약기반규약을 제안한다. 그리고 측면효과를 없애는 것을 돕는다. 또한 병행충돌 탐지가 성능을 높이기 위하여 개발된다. 더구나 시물레이션은 이 자원중심적 접근을 SOA로 통합한다. 결국 적시적 웹서비스 트랜잭션용 성능평가는 성공율, 실행기간, 보상비용 관점에서 커다란 개선을 보여준다. Web Service Transactions consist of long-running, loosely coupled and complex services across multiple distributed enterprises. Existing transaction protocols maintain overall consistency through compensation but with undesirable cost. Thus, researchers have proposed to reserve resources to reduce resource waiting time and compensation risk. Meanwhile, reservation also brings substantial locks and concurrent conflicts that impair performance. In this paper, we propose a reservation-based protocol but with an imposed time constraint, which helps eliminate those side effects. Also, concurrent conflicts detection is developed as well to enhance performance. Moreover, the simulation integrates this resource-centric approach into Service-Oriented Architecture (SOA). Finally, performance evaluation for timely Web Service Transaction shows a great improvement in aspects of success rate, execution duration and compensation cost.
