        In Vitro Evaluation and Monitoring of the Expression Level and Localization of Aldose Reductase Using Functionalized Quantum Dots and EGFP

        The optimization of aldose reductase (AR) expression levels and tracking of the AR expression sites within the cell is an essential step in developing a platform for the effective production of aldose reductase inhibitors (ARIs). In this study, we have demonstrated the use of both immunocytochemistry and quantum dots-based immunofluorescence techniques for observing and detecting the expression level and localization of AR in the cytoplasm and cell membrane of a eukaryotic cell model with high levels of AR protein expression. Our results show that high expression levels of human AR can be achieved using the eukaryotic cell model that we have developed. The overexpressed AR can be used for translational studies of hAR and the screening of ARIs. More importantly, the use of the established quantum dots-based immunofluorescence technique in the intracellular labeling of AR allows the determination of the expression and distribution of the AR gene. Overall, the use of the interdisciplinary approach of both genetic engineering and quantum dot-based immunofluorescence allows not only the effective production of a desired protein, but also the determination of the cellular localization of such an expressed protein.

        Studies on the Soybean Cyst Nematode (Heterodera glycines) in China

        Soybean has been cultivated in China for 5,000 years. The soybean cyst neamtode (SCN), Heterodra glycines, was recongnized in Northeastern China in 1899. Currently, it is known to occur in 12 provinces. The biology of SCN was investigated in several provinces. Six races of SCN were identified (race 1, 2, 3, 4, 5 and 7). About f10,000 soybean germplasm lines were evaluated for their resistance to race 1, 3, 4 and 5 of SCN. At least two black-seeded cultivars are resistant to all four races. Several tolerant soybean cultivars with yellow seed coat were released and are in production. Additional resistant cultivars are being developed. Nematicides were not applied in production Potential biocontrol agents and related aspects are being investigated.

      • Array-based Nano-amplification Technique Was Applied in Detection of Hepatitis E Virus

        A rapid method for the detection of Hepatitis E Virus (HEV) was developed by utilizing nano-gold labeled oligonucleotide probes, silver stain enhancement and the microarray technique. The 5'-end -$NH_2$ modified oligonucleotide probes were immobilized on the surface of the chip base as the capture probe. The detection probe was made of the 3'-end -SH modified oligonucleotide probe and nano-gold colloid. The optimal concentrations of these two probes were determined. To test the detection sensitivity and specificity of this technique, a conservative fragment of the virus RNA was amplified by the RT-PCR/PCR one step amplification. The cDNA was hybridized with the capture probes and the detection probes on microarray. The detection signal was amplified by silver stain enhancement and could be identified by naked eyes. 100 fM of amplicon could be detected out on the microarray. As the results, preparation of nano-gold was improved and faster. Development time also was shortened to 2 min. Thus, considering high efficiency, low cost, good specificity and high sensitivity, this technique is alternative for the detection of HEV.

      • Multiple Sexual Partners as a Potential Independent Risk Factor for Cervical Cancer: a Meta-analysis of Epidemiological Studies

        It's known that having multiple sexual partners is one of the risk factors of human papillomavirus (HPV) infection which is a major cause of cervical cancer. However, it is not clear whether the number of sexual partners is an independent risk factor for cervical cancer. We identified relevant studies by searching the databases of MEDLINE, PubMed and ScienceDirect published in English from January 1980 to January 2014. We analyzed those studies by combining the study-specific odds ratios (ORs) using random-effects models. Forty-one studies were included in this meta-analysis. We observed that the number of sexual partners was associated with the occurrence of non-malignant cervical disease (OR=1.82, 95%CI 1.63-2.00) and invasive cervical carcinoma (OR=1.77, 95%CI 1.50-2.05). Subgroup analyses revealed that the association remained significant after controlling for HPV infection (OR=1.52, 95%CI 1.21-1.83 for non-malignant disease; OR=1.53, 95%CI 1.30-1.76 for invasive cervical carcinoma). We found that there was a non-linear relation of the number of sexual partners with both non-malignant cervical disease and invasive cervical carcinoma. The risk of both malignant and non-malignant disease is relatively stable in women with more than 4-7 sexual partners. Furthermore, the frequency-risk of disease remained significant after controlling for HPV infection.The study suggested that h aving multiple sexual partners, with or without HPV infection, is a potential risk factor of cervical cancer.

      • Ultra-sensitive graphene based mid-infrared plasmonic bio-chemical sensing using dielectric beads as a medium

        <P>Graphene is moving beyond the realm of simple electronic devices toward areas such as advanced biochemical sensing. The infrared (IR) response of graphene, characterized by collective long-lived charge-carrier oscillations, could be applied in IR-absorption spectroscopy, typically used for bio-chemical analysis. However, direct light absorption by propagating plasmons in graphene is forbidden due to the large momentum mismatch. Proposed methods to overcome this bottleneck come at a cost, the use of noble metal particles on graphene reduces the spectral bandwidth and nano-structuring graphene is expensive. Here, we propose a simple and cheap method to fabricate large scale ultra-sensitive graphene based mid-IR biosensors, by introducing dielectric beads to excite mid-IR range plasmons. Interference from waves scattered by the beads excite surface plasmon polaritons, which propagate several micrometers in graphene and enhance the interaction between the molecules and mid-IR light.         An Analysis of the Regional Art and Cultural Characteristics of Manchu Paper-Cutting in Northern China -The Manchu Paper-Cutting in Xiuyan as an Example-

        진정한 의미에서의 민간예술작품에서 구현하는 것은 겉면에 표출된 화려한 표상할 뿐만 아니라 그 내면을 탐구하는 것, 더욱 이런 예술 풍격을 단조해 낸 불굴의 민족의식과 정감이다. 예로부터 사회의 발전과 계급 분화로 원시 예술의 변천을 거쳐 분리되어 형성된 문인예술, 궁정예술, 종교예술, 민간예술에 이르기까지 이러한 다른 관념을 기반으로 한 예술 문화 종류가 역사의 흐름 속에서 갈등하고 또 끊임없이 서로 연계하며 기반으로 하고 모순, 연계와 융합에서 하나의 완전한 민족 문화를 육성하였다. 오늘에 이르기까지 민간예술문화에서의 순수한 예술 신념은 여전히 줄곧 이른바 고상하고 우아한 예술 문화에 대해 확고한 기초적 역할을 하고있다. 수암만족 민간 전지(剪紙)는 동북 백산흑수사이에 만족 노동 인민이 창조한 역사적 예술 문화의 완벽한 결정체일 뿐만아니라 또한 수암의 전통적 만족 민간 예술 특유한 민족 예술문화 표식이고 동북 지역의 만족 의향 및 옥도 특색의 전통 민간예술의 걸출한 대표이다. 이것은 동북 만족 인민의 지혜의 예술을 포함하고 있으며 중요한 역사적 가치와 문화적 가치가 있을 뿐만 아니라 또한 중요한 예술적 가치와 과학적 가치를 지니고 있으며 전통문화의 결정체이자 화하문명의 보물이다. 지금은 어떤 의미에서 이미 문화브랜드로 된 민간 전지예술이 점차 문화의 수요품으로 되고 있다. 특유의 가치와 방식으로 문화가 널리 전파되는 또 다른 경로로 되고 있다. 그러나 북방 만족 전지를 놓고 말할 때 기나긴 과정에서 아주 많은 문제도 나타났다. 이를테면 제작 형식과 예술 풍격의 혼동, 관련 문화자료내용의 귀납 정리와 발전 전승 등이다. 본 논문은 북방 만족 문화의 대표 특징을 지닌 수암만족 전지를 연구 대상으로 지역 역사, 문화 신앙 등등 방면에 대해 중국 북방 만족 종이 오림이 형성된 근원성 원인을 심도 있게 분석하였다. 이와 동시에 종이 오림 작품이 드러낸 예술 상태에 있어서 다른 지역의 동류 작품들과 전체적 및 개별적 비교를 하여 예술풍격상의 지역문화 차이를 분명히했다. 또 현재를 결부시켜 전지 예술이 사람들에게 대한 의의를 서술하고 전통문화를 발전시키고 전승할 것을 호소했다. A true folk artwork is not only a gorgeous appearance, but also an unyielding national consciousness and emotion that forged this art style. Since ancient times, due to social development and class differentiation, primitive art has developed into literati art, court art, religious art, and then folk art, all of which were based on different concepts of art and culture, entangled in the long river of history and were constantly interconnected with each other, creating a whole national culture after having contradictions, connections. To this day, the pure artistic beliefs of folk art culture continue to provide a solid foundation for a relatively elegant art culture. The Manchu paper-cuttings in Xiuyan are not only the perfect crystallization of the historical art and culture created by the Manchu people living in the northeast; they are also the unique national art and cultural identity of the traditional Manchu folk art in Xiuyan. It is also an outstanding representative of the traditional folk art that characterizes Manchu’s homeland and Yudu in the northeast. These artistic treasures, which contain the wisdom of the people in the northeast, not only have important historical and cultural values but also have important artistic and scientific values. They are also the treasures of Chinese civilization. Nowadays, the paper-cutting art which has become a cultural brand, is quietly becoming a cultural commodity. It is providing an unique way for wide culture transmission. However, many different problems have also emerged throughout the development of the northern Manchu paper-cutting, such as the confusion in the form and artistic style, the induction of the content of relevant cultural materials and the development of heritage, etc. This paper analyzes the root causes of the formation of Manchu paper-cutting in northern China from the perspective of regional history, cultural beliefs, and so on, taking Xiuyan Manchu paper-cutting, which is representative of northern Manchu culture, as the subject. At the same time, it compares the artistic feature of Xiuyan Manchu paper-cutting with other similar works from other regions to clarify the regional cultural differences in the artistic style of paper-cutting in Xiuyan. The significance of paper-cutting is highlighted and the importance of developing and preserving traditional culture is also pointed out.

        Novel Microwave Absorption Materials of Porous Flower-Like Nickel Oxide@Polyaniline in the X-Band

        Porous flower nickel oxide@polyaniline (NiO@PANI) composites as excellent microwave absorption (MA) materials in the X-band were synthesized via a two-step strategy in this work. The porous NiO flower is uniformly dispersed and homogeneous in particle size after Ostwald ripening process. Coating conductive PANI on the surface of porous NiO microspheres could improve interfacial polarization and dielectric loss property that will lead to a great improvement of MA properties. Electromagnetic (EM) parameters of NiO@PANI composites with different NiO contents were investigated by a vector network analyzer and the reflection loss (RL) values with varied thickness were also calculated. The results showed that the effective absorption bandwidths (RL < -10 dB) of all NiO@PANI composites can cover the whole X-band. Especially, the NiO@PANI0.1 composite is able to attenuate microwave energy in the X-band with the thickness of 2.5mm. The NiO@PANI0.2 has a maximum RL of -32.8 dB at 10.1GHz and the effective absorption bandwidths cover 4.64GHz (11.12–15.76GHz) at 2.0mm. The excellent MA absorption performance may be ascribed to the polarization effect, dielectric loss and structure of porous flower-like NiO@PANI. Our work confirms that the synthesized NiO@PANI composite is an attractive candidate as a highly efficient MA material in the X-band.

      • CD44v3 and VEGF-C Expression and its Relationship with Lymph Node Metastasis in Squamous Cell Carcinomas of the Uterine Cervix

        Background: To investigate the expression of CD44v3 and vascular endothelial growth factor-C (VEGF-C) and their relationship with lymph node metastasis in squamous cell carcinomas (SCC) of the uterine cervix. Materials and Methods: Expression of CD44v3 and VEGF-C was analyzed in 109 cases of cervical SCC by immunohistochemistry (IHC). The relationship was analyzed between expression and the patient age, histological differentiation, formation of tumor emboli in lymphoid vessels, lymph node metastasis, FIGO staging, and TNM classification. Results: Expression rates for both CD44v3 and VEGF-C were 43.1% in cervical SCC. The cells with positive immunohistochemical staining of CD44v3 were distributed mainly around the keratin pearls in well differentiated carcinomas, but distributed diffusely in the moderately and poorly differentiated lesions. VEGF-C was found stained positively in most of the tumor cells. There were differences in expression between normal epithelium and atypical hyperplasia as well as carcinoma. Both CD44v3 and VEGF-C were found to be associated positively with lymph node metastasis and TNM classification (both p=0.000). Neither CD44v3 nor VEGF-C was found to be associated with patient age, histological differentiation, formation of tumor emboli in lymphoid vessels and FIGO staging. CD44v3 was found to be associated with VEGF-C positively (p=0.000). Conclusions: Abnormal expression of CD44v3 and VEGF-C is associated closely with the lymph node metastasis in cervical SCC, and these agents may cooperate in carcinogenesis and development of metastatic lesions.

        Efficient degradation of methylene blue dye by catalytic oxidation using the Na_8Nb_6O_19.13H_2O/H_2O-2 system

        Na_8Nb_6O_19.13H_2O particles were synthesized by a simple hydrothermal method. The catalysts were characterized by X-ray diffraction (XRD), scanning electronic microscopy (SEM) and thermogravimetric and differential scanning (TG-DSC). The XRD and TG-DSC analyses indicated that Na_8Nb_6O_19.13H-2O was an intermediate hexaniobate during the preparation of NaNbO_3 powders. Methylene blue (MB) dye degradation using Na_8Nb_6O_19.13H_2O/H_2O_2,Nb_2O_5/H_2O_2 and NaNbO_3/H_2O_2 systems were investigated, respectively. Among the catalytic oxidation systems, Na_8Nb_6O_19·13H_2O showed the highest activity for degradation of MB in the presence of H_2O_2. The results indicated that the dye degradation efficiency could be 93.5% at 30 ℃ after 60 min in the presence of the Na_8Nb_6O_19·13H_2O/H_2O-2 system. It was also found that the degradation of MB over the catalytic systems followed pseudo-first-order kinetics, and the degradation rate was 0.02376 min−1 in the Na_8Nb_6O_19.13H_2O/H_2O_2 system, which was higher than that in the Nb_2O_5/H_2O_2and NaNbO_3/H_2O_2 systems. A possible mechanism for MB catalytic oxidation degradation using the Na_8Nb_6O_19.13H-2O/H_2O_2 system was proposed.
