<P>Epithelial-mesenchymal transition (EMT) is an important process appearing in embryo development and tumor migration or progression, which is influenced by 17β-estradiol (E2). Bisphenol A (BPA) and nonylphenol (NP) are suspected as endocrine disrupting chemicals (EDCs) because they can exert estrogenic properties. In this study, we examined whether E2, BPA, and NP can lead to the EMT process in BG-1 ovarian cancer cells expressing estrogen receptors (ERs). To confirm the effect of E2, BPA, and NP, BG-1 cells were cultured under treatment with E2, BPA, or NP, and the alteration of EMT markers such as vimentin was examined at mRNA levels by using real-time and reverse-transcription (RT)-PCR. The expressions of snail, slug, and vimentin were enhanced by the treatment of E2, BPA, or NP compared to a control (DMSO). In protein levels, vimentin protein was increased by E2 and two EDCs, while E-cadherin was decreased. In addition, the expression of snail protein was enhanced by the treatment of E2 and the two EDCs in comparison with that of the control. Since EMT response in cancer cells can affect metastasis, we also performed a scratch assay and Western blot assay to show the migration ability caused by E2, BPA, or NP. Consequently, E2, BPA, and NP enhanced the migration capability of BG-1 cells and increased the expression of MMP-9 protein. Furthermore, to examine whether EMT and migration of BG-1 cancer cells are induced by BPA or NP via the ER dependent pathway, we cotreated the cells with ER-antagonist, ICI 182,780, in the presence of E2, BPA, or NP. As a result, the expressions of E-cadherin, vimentin, snail, and slug were reversed following treatment with an ER antagonist. Moreover, we confirmed that ICI 182,780 reduced the migration ability of BPA and NP to the control level. Taken together, these results indicate that BPA and NP, the potential EDCs, may have the ability to influence ovarian cancer metastasis via regulating EMT markers and migration in ER-expressing BG-1 ovarian cancer cells.</P>

국내 과학관 수는 지난 10여년 동안 양적으로는 팽창하고 있으나, 대부분이 중소규모의 과학관으로 인력부족 및 과학전시 컨텐츠 부족에 따라 운영에 어려움을 겪고 있다. 이에 따라 과학관이 보유하고 있는 전시 컨텐츠의 DB구축 및 정보 공유를 통해 과학관간 상호 교환 및 교류를 촉진할 수 있는 과학관 전시 플랫폼 구축이 요구되고 있는 실정이다. 국외 선진 과학관의 경우 자연사 컨텐츠의 전산화 및 DB 구축을 통해 대국민 검색서비스를 제공하여 전시품 등을 다양하게 활용하고 있는 추세이다. 따라서 본 연구에서는 과학관에서 활용가능한 자연사 컨텐츠 중, 곤충을 중심으로 이들의 전시 컨텐츠 발굴 및 DB확보 현황과 향후 플랫폼 구축 계획에 대해 소개하고자 한다.
Ustekinumab, an effective therapeutic option for erythrodermic psoriasis

Erythrodermic psoriasis is a severe form of psoriasis, presenting as prominent erythema, affecting the whole body. Ustekinumab (Stelara®) is a human monoclonal antibody targeting the p40 subunit shared by interleukin IL-12 and IL-23. Erythrodermic psoriasis has been an exclusion criteria in studies looking at the efficacy of ustekinumab against psoriasis, but recently, a few reports have proven its efficacy on erythrodermic psoriasis. Herein, we report 2 cases of erythrodermic psoriasis that successfully improved with ustekinumab. A 42-year-old man presented with a 7-year history of erythrodermic psoriasis and psoriatic arthritis. The patient was treated with ustekinumab 45 mg subcutaneously at 0, week 4, and every 12 weeks thereafter. At week 28, his Psoriasis Areaand Severity Index (PASI) score decreased from 64.8 to 9.6. The second case is a 26-year-old man suffering from erythrodermic psoriasis for 10 years. Four months after the first administration of ustekinumab, the patient achieved 75% improvement of the PASI score and 90% improvement after 112 weeks. Although further studies are required, it is likely that ustekinumab can be an effective therapeutic option for patients affected by erythrodermic psoriasis.
FC 2-7 : Elevated KAI1 protein expression identified in malignant melanoma

Background: KAI1/CD82 reportedly slows the metastatic process in a variety of tumors. However, its function is not fully clear. This study investigates the expression of KAI1 in correlation with malignant skin cancers. Objectives: To investigate the expression and clinical significance of KAI1 protein in malignant skin cancers. Methods: KAI1/CD82 expression was evaluated by western blotting in 24 specimens including 6 malignant melanoma (MM), 6 squamous cell carcinoma (SCC), 6 BCC and 6 normal skin specimens. Immunohistochemical staining was performed to measure KAI1/CD82 expression in 6 MM, 6 SCC and 6 BCC. Results: We observed over-expression of KAI1 in the malignant melanoma. The expressions of KAI1 in malignant melanoma were higher than those in SCC and BCC. The KAI1 expressions in malignant melanoma were higher in protein levels compared to SCC and BCC. Western blotting results agreed with immunohistochemistry. Conclusion: KAI1 is highly related to malignant melanoma and may be a useful indicator of malignant melanoma.

Background: Cancer survivors are at a higher risk of primary cancer recurrence and development of second primary cancer. In both cases, early disease detection is crucial. This cross-sectional study assessed cancer screening participation rates according to cancer history. Methods: Data were obtained from the 2010–2012 Korea National Health and Nutrition Examination Survey for 12,500 participants. Of these, 624 cancer survivors were enrolled in this study. Sampling weights were applied to maintain the representativeness of the Korean adult population. Results: Overall 2-year cancer screening rates prior to the survey in male and female cancer survivors were 59.9% and 73.7%, respectively, while opportunistic cancer screening rates were 33.5% and 52.1%, respectively. The odds ratios (95% confidence interval) of the overall cancer screening among the cancer survivors, compared to others, were 1.16 (0.79–1.72) in male and 1.78 (1.20–2.63) in female participants, after the adjustment for confounding variables. The odds ratios (95% confidence interval) for opportunistic cancer screening and National Cancer Screening Program among cancer survivors were 1.56 (1.07–2.27) and 0.80 (0.53–1.21) in males and 2.05 (1.46–2.88) and 0.66 (0.46–0.95) in females. Conclusion: Female cancer survivors showed a higher rate of overall and opportunistic cancer screening than did the male cancer survivors. Further efforts are required to improve cancer screening among male cancer survivors.

Collagen is decomposed by MMP-1, an enzyme induced by transcription factor activator protein 1 (AP-1) and matrix metalloproteinase-9 (MMP-9). Action of MMPs in inflammatory response promotes inflammatory cell movement and secretion, resulting in wrinkles on the skin. After using Bacillus sp. fermentation system and water, Artemisia vulgaris was fermented to prepare different solvent fractions using water, dichloromethane, hex ane, n-butanol, and ethyl acetate. These fractions were used to assess their effects on cell survival, wound healing, MMP-1/MMP-9 and procollagen type I C-peptide (PICP) expression, and skin turnover. MTT assay showed that cell viability of each treated group was 103% to 121%, indicating that A. vulgaris fractions were not toxic compared to control (cell viability: 100%). Wound healing assay revealed that wound healing ability in each treated group was 51% to 61%. This was lower than wound healing area in the control. Using RT-PCR, inhibition of MMP-1/MMP-9 gene expression was examined. As a result, each group treated with fraction showed reduced expression of both MMP-1 and MMP-9 compared to tumor necrosis factor-α (TNF-α) treatment group. Effects on collagen biosynthesis were analyzed using a PICP ELISA Kit. The group in which Artemisia vulgaris was treated increased collagen synthesis from 141 to 262ng/mL compared to the control group. Three-dimensional cell culture revealed that each fraction could increase skin wall formation. These results suggest that each fractions has anti-aging and anti-wrinkle effects on the skin, indicating its suitability as a functional material.
