      • 국내 분리 폐렴 구균의 항균제 내성 현황 및 분자역학 조사를 통한 내성 전파의 규명

        목 적 : 최근 폐렴구균의 항균제 내성은 전 세계적인 문제가 되고 있다. 국내에서도 폐렴구균의 페니실린 내성율이 70%를 넘어 세계 최고 수준의 내성을 나타내고 있다. 본 연구는 국내 분리 폐렴 구균의 페니실린 내성 및 다제 내성 현황을 확인하고, 국내의 내성율이 외국에 비하여 지나치게 높은 원인으로서 내성 전파의 가능성을 규명하고자 항균제 감수성 검사 및 pulsed-field gel electrophoresis(PFGE)와 penicillin-binding protein(PBP) profile을 분석하였다. 대상 및 방법 : 국내 2개의 3차 병원에서 1989년부터 1995년까지 153명의 환자 검체에서 분리된 173균주의 폐렴 구균을 대상으로 하였다. 항균제 감수성 검사는 oxacillin disk로 페니실린 내성 여부를 검색한 후, 한천희석법을 이용하여 11개 항균제의 성장억제최소농도를 구하였다. 항균제 감수성 검사 결과 모든 β-lactam 제제에 동시 내성을 보인 다제 내성 균주 42 균주를 대상으로 PFGE 와 PBP profile 분석을 시행하였다. PFGE는 SmaI과 ApaI을 제한효소로 사용하였으며, CHEF DR-Ⅱ system을 이용하여 전기영동하였다. PBP profile은 ³H-benzylpeniclillin으로 균주를 label하여 분석하였다. 결 과 : 항균제 감수성 결과 전체의 67.8%가 중등도 이상의 내성을 보였으며 (중등도 8.2%, 내성 59.6%), 3가지 종류 이상의 항균제에 동시 내성을 보이는 다제 내성율은 25.4%였다. 페니실린에 내성을 보이는 균주는 대부분 다른 항균제 특히 cephalosporin 제제에 내성을 보였다. 시험 균주의 혈청균은 19와 23이 가장 많았다. PFGE의 결과 35개의 다제 내성 균주 및 29 균주 (83%)가 동일한 제한 절편 양상을 보였으며, 총 5가지의 PFGE pattern 을 나타내었다. PBP profile 분석에서도 PFGE상 동일한 양상을 보인 다제내성 균주들은 PBP profile이 동일하였다. 결 론 : 본 연구는 기존의 국내 보고와 마찬가지로 폐렴구균의 페니실린 내성율이 세계 최고 수준임을 확인하였고, 아울러 다제 내성율도 현저히 증가하여 있음을 확인하였다. PFGE를 통한 유전자 절편 분석과 PBP의 변화 양상 분석을 통하여 다제 내성 균주의 80% 이상이 유전적 상관성이 있음을 확인함으로써, 내성의 전파가 국내에서 일어나고 있으며, 이 현상이 비교적 단기간에 국내에서 폐렴구균의 내성율을 급증하게 한 원인의 일부일 것으로 생각하였다. 향후 내성전파의 기전 및 근원에 대한 후속 연구가 내성 문제에 대한 장기적인 대책 마련에 필수적일 것으로 사료된다. Background : During recent three decades, the resistance of Streptococcus pneumoniae to penicillin has been rapidly increasing in many parts of the world. To characterize the epidemiology of multidrug-resistant S. pneunoniae as well as to investigate the possible spread of multiresistant clones which could partly explain the unusually high resistance rate in Korea, we conducted the antimicrobial susceptibility tests of pneumococcal isolates and performed pulsed-field gel electrophoresis (PFGE) and penicillin-binding protein (PBP) profile analysis of multiresistant strains from Korea. Methods : A total of 174 pneumococcal isolates obtained from 153 patients who visited or were admitted to two tertiary care hospitals during the period from 1989 to 1995 were studied. Antimicrobial susceptibility tests for 173 isolates were performed using agar dilution method to determine minimal inhibitory concentrations of 11 antimicrobial agents. A total of 42 isolates with multidrug-resistance were subjected to PFGE and PBP profile analysis to investigate the genetic relatedness between multiresistant strains, SmaI and ApaI were used to digest the genomic DNAs in PFGE. For PBP profile analysis, 5 mCi³H-benzylpenicillin were used to label PBPs of pneumococci. Results : Antimicrobial susceptibility tests for 173 isolates showed that 67.8% of isolates were not susceptible to penicillin, exhibiting either intermediate (8.2%) of resistance (59.6%). Multidrug-resistance to three or more classes of antibiotics was observed in 25.4% of the isolates. Almost all isolates with resistance to penicillin were resistant to cephalosporins. PFGE showed that 29 of 35 Korean isolates with multidrug-resistance had an identical PFGE pattern. These strains also shared a common PBP profiles with decreased affinity of PBPs 1a, 2x, and 2b. Conclusion : These data demonstrated the extremely high rates of penicillin- and multidrug-resistance in pneumococcal isolates in Korea. Molecular epidemiologic studies suggest the spread of a single epidemic clone of resistant pneumococci within Korea, which could partly explain the unusually high rate of resistance in Korea. Future investigation with regards to source and mode of transmission is warranted.

      • ^40Ca(d,p)^41Ca 직접 핵반응에서의 결합채널방정식에 의한 편극율 계산

        The polarization ratio of ^40Ca(d,p)^41Ca direct reaction was calculated by coupled channel method with Woods-Saxon optical potential. And the polarization ratio for incident particle energy of 10.8 and 20.8 MeV were calculated in the ground state f and first excited state p of ^41Ca nucleus respectively. The result of coupled channel method was compared with experimental data and a result of distorted wave Born approximation method. It was found that the result of calculation by coupled channel equation agreed with the experimental data and that it was shown CC calculation to be better than DWBA calculation.

      • 혐기-무산소-호기공정에 의한 도시하수의 질소·인제거에 관한 연구

        본 연구는 질소와 인을 동시에 제거할 수 있는 생물학적 질소-인제거 공정인 혐기-무산소-호기공정을 산업폐수가 포함된 도시하수에 적용한 경우의 유기물 및 질소·인의 제거특성에 대하여 조사하였다. 원수의 특성은 유기물의 농도가 CODCr 120∼230mg/ι로 평균 178mg/ι을 나타내었으나 무기성 탄소원의 농도가 매우 높았으며 이에 따라 생물학적 제거 가능한 유기물의 비율은 낮아 충분한 질산화와 탈질화가 이루어지지 않았다. CODCr/NH_(3)-N이 평균 10이상으로 높았지만 무기성 탄소원이 높아 탈질에 필요한 추가적인 에너지원의 투입을 고려해야 하며, 또한 alkalinity/NH_(3)-N의 비가 2.3정도로 낮게 나타나 완전한 질산화를 위해서는 alkalinity의 공급이 필요한 것으로 조사되었다. 유기물에 대한 평균 제거율은 BOD 84%, CODCr 53%를 나타내었으며, NH_(3)-N의 제거율은 평균 76%이었고 T-N의 제거율은 60%이었다. 그리고 T-P 제거율은 50%정도를 나타내었다. The anaerobic-anoxic-aerobic process was used for biological nitrogen and phosphorus removal of municipal sewage. The CODCr concentration of municipal sewage was 120∼230mg/ι and its average CODCr concentration was 178mg/ι. But because of the high inorganic concentration of municipal sewage, the biological nitrification and denitrification were not accomplished successfully. The alkalinity/NH_(3)-N was 2.3 and under those conditions the additional carbon source and lime should be added The removal efficiency BOD and CODCr were 84% and 53% respectively. The removal efficiencies T-N and T-P were 60% and 50% respectively.

Also the internet connected around the world completely diminished the limit of time and distance and company have enveloped keen competition out of each nation and continent in the world market. To fulfill these demands of customer, they need a concept of eCRM(Web based Customer Relationship Management), and go from selling products and services, or gathering customer requests, up to the phase of solving customer's problem by real time or previous action, With the help of internet, the frequency and speed of the problem solving has improved greatly. In this paper provide concept of eCRM and Axiom Design. And with the target company of study, it was built eCRM system using Axiom design to insert design for customer request.

        김치저장성 향상을 위한 동결방법이 배추조직의 물성변화에 미치는 영향

        김치를 10˚C에서 8일간 숙성시켜 산도 0.6~0.7%로 숙성된 김치를 -70˚C와 -20˚C로 냉동하며 -20˚C에서 저장하면서 배추 조직의 elasticity, hardness, 세포 조직의 변화, 드립양을 실험한 결과는 다음과 같다. -70˚C에서 급속 동결한 것과 -20˚C에서 완만 동결한 냉동 김치 중 배추조직의 elasticity는 냉동 저장 15일까지 감소하다 일정하게 유지되었고 hardness는 거의 변화가 없었으며 냉동 방법에 따른 변화도 거의 나타나지 않았다. 투과 전자현미경으로 관찰한 결과 control의 경우 세포벽이 매우 두꺼우며 세포의 모양들이 잘 보존되어 잇는 것을 볼 수 있었고 -20˚C로 냉동 처리하여 해동시킨 세포벽들은 많이 손상되어 있음을 볼 수 있었으며 -70˚C로 급속 냉동시료의 경우 세포벽의 손상 정도가 -20˚C로 냉동 처리한 시료보다 덜 파괴되어 있음을 볼 수 있었다.한편 냉동 저장기간 동안 드립의 손실량의 변화는 -70˚C로 냉동 처리한 시료의 드립양은 3~4%정도로 -20˚C로 냉동 처리한 시료의 5~6%에 비해 적은 것을 알 수 있었다. This study was conducted to investigate the effect of freezing methods, show freezing at -20℃ and rapid freezing at -70℃, on physical properties of Chinese cabbage in frozen Kimchi during storage at -20℃. Elasticity of midrib of Chinese cabbage in frozen Kimchi was decreased until 15 days and did not changed thereafter during storage at -20℃, Hardness of that was not changed during storage. Those results of elasticity and hardness of slow frozen sample are similar to rapid frozen sample. By the morphological observation through transmission electron microscope, more of cellular structure of Chinese cabbage in slow frozen was destructed than that of rapid frozen sample. Drip loss was more in slow frozen sample than that in rapid one.

      • TSN 206 SA형 질량분석기의 고유특성에 관한 연구

        Some characteristics of Model TSN 206 SA Mass Spectrometer, especially, on the variation of ion population as a function of time at the ion source, response time of the magnetic field and stability of the field after step scanning at peaks have been investigated and the following results are obtained. 1) Magnetic field response is fast enough for a general experimental purpose so that waiting time longer than 50μsec. after step scanning at peaks gives sufficient accuracy. 2) Integration time is more important than waiting time and the former should be longer than 1sec. in order to have ordinary analytical accuracy. 3) Built-in clock system is designed such that it's increment is 0.2sec. For short integrating time experiment, it is very important that the integration should be set at duration of integar multiple of 0.2sec. 4) In order to obtain more accurate isotope ratio, data processing program must be modified considering the pattern of tatal ion population variation during a measurement in the ion source of the system.

        The purpose of this study is to examine the effects of combined swim training and run trauung. We divided 14 long distance runners of middle and high school to groups, swimming and run combined training group(7), and run training group(7). They was trained for 10 weeks and analyzed the change of cardiopulmonary function and endurance The results are follows. 1.There were no significant difference between combined group and run trained group of the value of HRrest and HRmax. 2.The value of VO2max did not increased meaningfully between two groups. 3.There were no significant difference between combined and run group in the value of VEmax. 4. There were no significant difference between combined and run group in the BF. 5.The change of VT was increased meaningfully in the combined group than that of run training group. 6.The change of VCO2 was slightly increased in the combined group that of run trained group, but significant difference were not seen. 7.The change of all-out time and running distance were significantly higher in the combined group than those of run trained group.

        The water quality in the Taechong dam basin was simulated for some conditions of flow and pollutant load with QUAL2E model verified to be available in several examples. The changes of water quality in 1996, 2001 and 2006 were predicted to evaluate the pollutant load contributions and aid to establish the water quality control plan. The flow rate for environment maintenance of upstream basin of Kum river was also proposed.
