        STUDIES ON BOUNDARY VALUE PROBLEMS FOR BILATERAL DIFFERENCE SYSTEMS WITH ONE-DIMENSIONAL LAPLACIANS

        Existence results for multiple positive solutions of two classes of boundary value problems for bilateral difference systems are established by using a fixed point theorem under convenient assumptions. It is the purpose of this paper to show that the approach to get positive solutions of boundary value problems of finite difference equations by using multi-fixed-point theorems can be extended to treat the bilateral difference systems with one-dimensional Laplacians. As an application, the sufficient conditions are established for finding multiple positive homoclinic solutions of a bilateral difference system. The methods used in this paper may be useful for numerical simulation. An example is presented to illustrate the main theorems. Further studies are proposed at the end of the paper.

        This paper is concerned with a kind of non-homogeneous boundary value problems for singular second order differential systems with Laplacian operators. Using multiple fixed point theorems, sufficient conditions to guarantee the existence of at least three solutions of this kind of boundary value problems are established. An example is presented to illustrate the main results.

        Study on the Stability of Accumulated Layer Landslide under the Coupling Action of Earthquake and Rainfall

        Tectonic movement, heavy rainfall and other environmental factors are important factors causing the slope instability of accumulation body. Aiming at jiangdingya H1 landslide in Bailong River Basin, Zhouqu County, Gansu Province, the deformation law and stability of accumulation landslide under the coupling condition of earthquake and rainfall were systematically analyzed by means of laboratory physical model test, numerical simulation and theoretical derivation. The results show that: 1) The crack development law of landslide model test under the coupling effect of earthquake and rainfall is analyzed, and the stability characteristics of landslide deformation and failure in three stages are obtained. 2) According to the model test and numerical simulation results, it is found that the coupling effect has the greatest influence on the stability of the slope toe of the accumulation landslide, and the failure mode of the accumulation landslide changes from “pull-shear” failure to “compression-shear” failure, and the failure of the accumulation landslide occurs in a local location. 3) Based on the unbalanced thrust method and GA model, a landslide stability calculation model of loose accumulation body under the coupling effect of earthquake and rainfall was improved by comprehensively considering the effects of complex factors such as seismic inertia force and seepage force.The research results can provide reference for landslide prevention, monitoring and early warning in Zhouqu area of Gansu Province.

        The Arabidopsis Mediator Complex Subunit MED19a is Involved in ABI5-mediated ABA Responses

        Arabidopsis Mediator complex subunit 19a(MED19a), which mediates interactions between transcriptionalregulators and RNA polymerase II, plays a critical role inplant response to infection by pathogens. However, the rolesof MED19a in other signaling pathways are unknown. Here,we report that MED19a plays an important role in regulationof abscisic acid (ABA)-mediated transcriptional regulation inArabidopsis. Plants deficient in MED19a showed reducedsensitivity to ABA inhibition of seed germination, cotyledongreening, root growth, and stomatal opening. MED19adeficientmutants also had reduced resistance to droughtstress, evidenced by high water-loss rates and low survivalrates. Molecular genetic analysis revealed that MED19amutants had down-regulated ABA-induced genes, includingEm1, Em6, and RD29B, and MED19a could occupy thepromoters of Em1 and Em6 in an ABA-dependent manner. Furthermore, MED19a interacted with the transcription factorABA-insensitive 5 (ABI5) in split-luciferase complementationassays and co-immunoprecipitation assays. An analysis ofdouble mutants (med19a-2 and abi5-7) suggested that theaction of MED19a in ABA signaling was dependent uponABI5. Furthermore, MED19a and ABI5 influenced eachother in recruiting the promoters of the target genes Em1 andEm6, which are involved in embryonic development. Altogether,these results indicate that MED19a acts as a positive regulator inABI5-mediated ABA responses.

        Investigating chemical features of Panax notoginseng based on integrating HPLC fingerprinting and determination of multiconstituents by single reference standard

        Background: Panax notoginseng is a highly valued medicine and functional food, whose quality is considered to be influenced by the size, botanical parts, and growth environments. Methods: In this study, a HPLC method integrating fingerprinting and determination of multiconstituents by single reference standard was established and adopted to investigate the chemical profiles and active constituent contents of 215 notoginseng samples with different sizes, from different botanical parts and geographical regions. Results: Chemical differences among main root, branch root, and rotten root were not distinct, while rhizome and fibrous root could be discriminated from other parts. The notoginseng samples from Wenshan Autonomous Prefecture and cities nearby were similar, whereas samples from cities far away were not. The contents of major active constituents in main root did not correlate with the market price. Conclusion: This study provided comprehensive chemical evidence for the rational usage of different parts, sizes, and growth regions of notoginseng in practice.

        Analysis of the DNA Methylation of Maize (Zea mays L.) in Response to Cold Stress Based on Methylation-sensitive Amplified Polymorphisms

        DNA methylation plays a vital role in tuning geneexpression in response to environmental stimuli. Here,methylation-sensitive amplified polymorphisms (MSAP)were used to assess the effect of cold stress on the extent andpatterns of DNA methylation in maize seedlings. Overall,cold-induced genome-wide DNA methylation polymorphismsaccounted for 32.6 to 34.8% of the total bands at the differenttreatment time-points. It was demonstrated that the extentand pattern of DNA methylation was induced by cold stressthrough the cold treatment process and that thedemethylation of fully methylated fragments was the maincontributor of the DNA methylation alterations. The sequencesof 28 differentially amplified fragments relevant to stresswere successfully obtained. Under the cold stress, demethylationwas detected in most fragments. BLAST results indicate thatthe homologues of these fragments are involved in manyprocesses, including hormone regulation, cold response,photosynthesis, and transposon activation. The expressionanalysis demonstrated an increase in the transcription of fivedemethylated genes. Despite the fact that DNA methylationchanges and cold acclimation are not directly associated, ourresults may indicate that the specific demethylation of genesis an active and rapid epigenetic response to cold in maizeduring the seedling stage, further elucidating the mechanismof maize adaptation to cold stress.

        Dynamic Analysis of Slab Track with Crack Considering Rail Corrugation

        To investigate the dynamic response of ballastless slab track with crack in the rail corrugation area under the load of urban rail vehicle, this study establishes a vertical coupling dynamic model of the vehicle-rail-slab track, considering crack and rail corrugation. The exact closed-form solution of the vibration mode of the Euler Bernoulli beam with crack is introduced. Furthermore, a finite element model of the slab track is developed, and the vertical dynamic model's fastening force is applied to the slab track at the rail corrugation frequency to calculate the surface compressive stress of the slab track. The results indicate that, under rail corrugation conditions of varying magnitudes, the presence of crack induces higher vertical acceleration amplitudes in the slab track at low frequencies. As the wave depth increases, the acceleration of the slab track at the crack gradually intensifies. Moreover, with an increasing wavelength, the corresponding frequency to the acceleration amplitude of the slab track at the crack gradually decreases. Notably, when the wave depth is 0.2 mm and the wavelength is 50 mm, the axle load transfer rate exhibits the greatest increase. These research findings offer valuable insights for the maintenance of urban rail transit slab track systems.

        Hepatic Hippo signaling inhibits development of hepatocellular carcinoma

        Primary liver cancer is one of the most common cancer worldwide. Hepatocellular carcinoma (HCC) in particular, is the second leading cause of cancer deaths in the world. The Hippo signaling pathway has emerged as a major oncosuppressive pathway that plays critical roles inhibiting hepatocyte proliferation, survival, and HCC formation. A key component of the Hippo pathway is the inhibition of yes-associated protein (YAP)/transcriptional co-activator with PDZ-binding motif (TAZ) transcription factors by the Hippo kinase cascade. Aberrant activation of YAP or TAZ has been found in several human cancers including HCC. It is also well established that YAP/TAZ activation in hepatocytes causes HCC in mouse models, indicating that YAP/TAZ are potential therapeutic targets for human liver cancer. In this review, we summarize the recent findings regarding the multifarious roles of Hippo/YAP/TAZ in HCC development, and focus on their cell autonomous roles in controlling hepatocyte proliferation, differentiation, survival and metabolism as well as their non-cell autonomous in shaping the tumor microenvironment.
