        Overexpression of Chalcone Isomerase (CHI) Increases Resistance Against Phytophthora sojae in Soybean

        Chalcone isomerase (CHI) is a key enzyme ofthe flavonoid pathway that plays crucial roles in plant responsesto various pathogens. Our previous study using comparativetranscriptome analysis of soybean inoculated with or withoutPhytophthora sojae revealed Glyma.20G241500, a homologof CHI1A in Arabidopsis thaliana, to be induced by P.sojaein soybean. In the present study, we isolated the gene fromsoybean cultivar Nannong 10-1, designating it GmCHI1A. GmCHI1A transcripts were detected in soybean roots, stemsand leaves, with the highest levels in roots. Interestingly,GmCHI1A localizes to the cortical endoplasmic reticulum(ER). GmCHI1A was overexpressed in hairy roots to furtheruncover the roles of CHIs in soybean. Compared to emptyvector-expressing hairy roots (EV) infected with P. sojae,hairy roots overexpressing GmCHI1A (OE) exhibited decreasedP. sojae biomass accumulation, shortened the lesion lengthsand reduced zoospores germination. These results demonstratethat GmCHI1A plays positive roles in the response ofsoybean to P. sojae.

      • Combined Screening of Cervical Cancer, Breast Cancer and Reproductive Tract Infections in Rural China

        Objectives: To investigate the current prevalence and knowledge of cervical cancer, breast cancer and reproductive tract infections (RTIs) in rural Chinese women, and to explore the acceptance and feasibility of implementing a combined screening program in rural China. Methods: A population-based, cross-sectional study was conducted among women aged 30 to 59 years old in Xiangyuan County, Shanxi Province from 2009 to 2010. Socio-demographic characteristics, knowledge of cervical cancer, breast cancer and RTIs, and the attitude toward single or combined screening were collected by an interview questionnaire. Each participant received a clinical examination of the cervix, breast and reproductive tract. Examinations included visual inspection, mammography, laboratory tests and pathological diagnosis. Results: A total of 1,530 women were enrolled in this study. The prevalence of cervical precancerous lesions, suspicious breast cancer, suspicious benign breast disease and RTIs was 1.4%, 0.2%, 14.0% and 54.3%, respectively. Cervicitis, trichomonas vaginitis, and bacterial vaginitis were the three most common RTIs among our participants. Television, radio broadcast, and public education during screening were the major source of healthcare knowledge in rural China. Moreover 99.7% of women expressed great interest in participating in a combined screening project. The affordable limit for combined screening project was only 50 RMB for more than half of the rural women. Conclusion: A combined screening program would be more effective and popular than single disease screening projects, while appropriate accompanied education and a co-pay model for its successful implementation need to be explored, especially in low-resource settings.

        Gut Microbiota Community and Its Assembly Associated with Age and Diet in Chinese Centenarians

        Increasing evidence suggests that gut microbiota underpin the development of health and longevity. However, our understanding of what influences the composition of this community of the longevous has not been adequately described. Therefore, illumina sequencing analysis was performed on the gut microbiota of centenarians (aged 100-108 years; RC) and younger elderlies (aged 85-99 years; RE) living in Bama County, Guangxi, China and the elderlies (aged 80-92 years; CE) living in Nanning City, Guangxi, China. In addition, their diet was monitored using a semiquantitative dietary questionary (FFQ 23). The results revealed the abundance of Roseburia and Escherichia was significantly greater, whereas that of Lactobacillus, Faecalibacterium, Parabacteroides, Butyricimonas, Coprococcus, Megamonas, Mitsuokella, Sutterella, and Akkermansia was significantly less in centenarians at the genus level. Both clustering analysis and UniFraq distance analysis showed structural segregation with age and diet among the three populations. Using partial least square discriminate analysis and redundancy analysis, we identified 33 and 34 operational taxonomic units (OTUs) as key OTUs that were significantly associated with age and diet, respectively. Age-related OTUs were characterized as Ruminococcaceae, Clostridiaceae, and Lachnospiraceae, and the former two were increased in the centenarians; diet-related OTUs were classified as Bacteroidales, Lachnospiraceae, and Ruminococcaceae. The former two were deceased, whereas the later one was increased, in the high-fiber diet. The age and high-fiber diet were concomitant with changes in the gut microbiota of centenarians, suggesting that age and high-fiber diet can establish a new structurally balanced architecture of gut microbiota that may benefit the health of centenarians.

        A Case Study of the Effects of a Synoptic Situation on the Motion and Development of Warm-Sector Mesoscale Convective Systems over South China

        The effects of a synoptic situation on the motion and development of warm-sector mesoscale convective systems (MCSs) in a heavy rainfall event occurred in South China on 8 May 2014 are investigated using high-resolution observational data and ERA-Interim data. The results show that the blocking of an eastward moving low-level trough over southwestern China by a stable anticyclone over eastern China induced strong southerly winds ahead of the trough and an eastward vertical slant of the wind speed maximum. Consequently, a southeastward low-level wind shear formed in the region under the southerly jet, which caused a southeastward turning of the motion of a large inland MCS. Meanwhile, a northeastward/eastward low-level wind shear formed in the region under the east side of the southerly jet, leading to northeastward/eastward movement of some small MCSs in the coastal region. The continuous merging of the southeastward-moving MCS with the northeastward/eastward-moving MCSs resulted in heavy rainfall along the coastal region. On the other hand, the intensification and expansion of associated low and high mean-sea-level pressure (MSLP) centers caused southeastward migration of strong boundary-layer moisture flux convergence and high convective available potential energy zones, which promoted the persistent development of the large MCS and the later development of the small MCSs. Moreover, the strong boundary-layer southeasterly winds associated with the high MSLP center continuously enhanced the frontal ascent of the large MCS. These southeasterly winds also triggered the small MCSs through the convergence induced by differential surface friction between the land and sea or the convergence of them with the southwesterly winds from the South China Sea.

      • Efficacy and Survival-associated Factors with Gefitinib Combined with Cisplatin and Gemcitabine for Advanced Non-small Cell Lung Cancer

        Objective: To analyze the efficacy and survival associated factors of gefitinib combined with cisplatin and gemcitabine for advanced non-small cell lung cancer. Materials and Methods: A total of 57 patients with advanced non-small cell lung cancer (NSCLC), who received platinum-based chemotherapy regimens for more than 1 cycle, were treated with gefitinib combined with cisplatin and gemcitabine until disease progression. Efficacy, survival time and adverse reactions were observed. The Kaplan-Meier method was adopted for analysis of survival and Cox regression for associated influencing factors. Results: The patients were followed up until October 31, 2013, and the median follow-up time was 19 months. Of 57 patients, there were 4 (7.0%) with complete remission (CR), 8 (14.0%) with partial remission, 31 (54.4%) with stable disease, and 14 (24.6%) with disease progression. The remission rate was 21.1% and the disease control rate was 75.4%. The median progression-free survival (PFS) time and the median overall survival time were 10 months and 15.2 months. The one-year, two-year and three-year survival rates were 47.4%, 23.3% and 10.0%. Gender and pathological types were the independent risk factors influencing PFS time (P=0.028, P=0.009). Tumor pathological type and early efficacy were independent factors for the prognosis (P=0.018, P=0.000). Adverse reactions were mostly rashes of I~II degree and diarrhea and slightly increasing level of aminopherase. The skin adverse event incidence of III degree or above was 1.8% (1/57) and brain metastasis was foudn in 31.6% (18/57). Conclusions: Gefitinib combined with cisplatin andgemcitabine, is effective for patients with IIIb~IV NSCLC who received multiple cycles of chemotherapy.

        Numerical Study of the Role of Microphysical Latent Heating and Surface Heat Fluxes in a Severe Precipitation Event in the Warm Sector over Southern China

        Simulations of the severe precipitation event that occurred in the warm sector over southern China on 08 May 2014 are conducted using the Advanced Weather Research and Forecasting (WRF-ARWv3.5.1) model to investigate the roles of microphysical latent heating and surface heat fluxes during the severe precipitation processes. At first, observations from surface rain gauges and groundbased weather radars are used to evaluate the model outputs. Results show that the spatial distribution of 24-h accumulated precipitation is well reproduced, and the temporal and spatial distributions of the simulated radar reflectivity agree well with the observations. Then, several sensitive simulations are performed with the identical model configurations, except for different options in microphysical latent heating and surface heat fluxes. From the results, one of the significant findings is that the latent heating from warm rain microphysical processes heats the atmosphere in the initial phase of the precipitation and thus convective systems start by self-triggering and selforganizing, despite the fact that the environmental conditions are not favorable to the occurrence of precipitation event at the initial phase. In the case of the severe precipitation event over the warm sector, both warm and ice microphysical processes are active with the ice microphysics processes activated almost two hours later. According to the sensitive results, there is a very weak precipitation without heavy rainfall belt when microphysical latent heating is turned off. In terms of this precipitation event, the warm microphysics processes play significant roles on precipitation intensity, while the ice microphysics processes have effects on the spatial distribution of precipitation. Both surface sensible and latent heating have effects on the precipitation intensity and spatial distribution. By comparison, the surface sensible heating has a strong influence on the spatial distribution of precipitation, and the surface latent heating has only a slight impact on the precipitation intensity. The results indicate that microphysical latent heating might be an important factor for severe precipitation forecast in the warm sector over southern China. Surface sensible heating can have considerable influence on the precipitation spatial distribution and should not be neglected in the case of weak large-scale conditions with abundant water vapor in the warm sector.

        First-principles study of the switching characteristics of the 15,16-dinitrile DDP/CPD-based optical molecular switch with carbon nanotube electrodes

        We have studied the switching characteristics of an optical molecular switch based on the 15,16-dinitrile dihydropyrene/cyclophanediene (DDP/CPD) molecule with two single-walled carbon nanotube (SWCNT) electrodes using first-principles transport calculations. It is shown that the DDP shows an overall higher conductance than the CPD at low bias which is independently of the SWCNT chirality. Meantime, the conductance of the molecular switch can be tuned by the chirality of the SWCNT.         Microwave-assisted Approach for the Rapid Enzymatic Digestion of Rapeseed Meal

        This study demonstrates the use of a new microwave-assisted approach for accelerating the enzymatic digestion of rapeseed meal. The effects of different microwave parameters, such as the time, temperature, and power level, on the degree of hydrolysis (DH) were investigated by using response surface methodology (RSM). The maximum predicted DH value (10.2%) was in good agreement with the value obtained experimentally using an alkaline protease, which was 12.57% under optimal conditions. In only 7 min, the microwave-assisted method achieved a DH value similar to that obtained by the conventional enzymatic digestion method (4 hr). Therefore,this new technique for rapid enzymatic digestion will improve the application of rapeseed meal in the preparation of protein hydrolysates for use in food and feed.

      • Organo-layered double hydroxides for the removal of polycyclic aromatic hydrocarbons from soil washing effluents containing high concentrations of surfactants

        <P><B>Abstract</B></P> <P>Disposal of soil washing effluent (SWE) resulting from the surfactant-enhanced remediation of soil containing hydrophobic organic contaminants (HOCs)is complicated because of the presence of high levels of surfactants. The synthesized layered double hydroxides (LDHs), modified with sodium dodecyl sulfonate (SDS) in different loading amounts (organo-LDHs),were evaluated in this study as sorbents for the removal of two typical HOCs, phenanthrene (PHE) and pyrene (PYR),from a simulative SWE. The results showed that the organo-LDHs can effectively sorb PHE and PYR from the SWE within an equilibrium time of 2 h. All isotherms were linear and the sorption capabilities of the organo-LDHs increased almost linearly with the increase in the amount of SDS loaded on the LDHs. Besides, the surface areas of the organo-LDHs decreased sharply with the increase in SDS loading owing to the hindrance of the exposed surface of the LDHs by the incorporated SDS. These findings indicated that partitioning dominated the sorption process rather than adsorption, and the strong affinity of HOCs towards the organic phase in LDHs assisted in the effective removal of polycyclic aromatic hydrocarbons (PAHs) from the SWE. Furthermore, the sorption capabilities of organo-LDHs towards PHE and PYR at the higher loading amounts of SDS were much greater than that of commercial activated carbon at the higher concentration ranges of PAHs.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Dodecyl sulfonate anion modified LDHs are employed for the removal of PHE and PYR in soil washing effluent (SWE). </LI> <LI> Organo-LDHs showed superior performance in PHE removal from SWE than that of commercial activated carbon. </LI> <LI> Partitioning of PHE and PYR in the organic fraction of the organo-LDHs was the main mechanism. </LI> <LI> Using organo-LDHs for the separation of HOCs from surfactants in SWE facilitated reuse of the surfactants. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>
