        Identification of AFLP and SCAR Markers Linked to the Fertility Restorer Gene for the "Polima" CMS in Brassica napus

        Polima (Pol) system of cytoplasmic male sterility (CMS) has been widely used for hybrid breeding in Brassica napus L in China. Molecular markers tightly linked to the restorer gene would be not only a valuable tool for breeding new restorer lines but also important landmarks for cloning the gene based on the linkage map. In order to identify DNA markers linked to its restorer gene ( Rfp), a near-isogenic line (NIL) population segregating for male sterility ( rfprfp) /fertility ( Rfprfp) was subjected to amplified fragment length polymorphism (AFLP) analysis using the bulked segregant analysis (BSA) method. From the survey of 2,560 primer combinations, nine AFLP markers tightly linked to the target gene were identified. The distance between the two closest markers (ZQA and ZQH) and the target locus were both 0.4 cM, but they located at the same side of the Rfp locus. To exploit fast, cheap and reliable PCR-based markers for large-scale application in marker-assisted selection (MAS), polymorphic fragments of these two AFLP markers were sequenced and successfully converted into sequence characterized amplified region (SCAR) markers. This suite of markers could be valuable in developing new elite restorers in rapeseed.

        Experimental study on characteristic of sloshing impact load in elastic tank with low and partial filling under rolling coupled pitching

        A series of experiments covering lowest three natural frequencies of rolling coupled pitching were conducted to investigate liquid sloshing with low liquid depth. The test results show that the most violent liquid sloshing in rolling and pitching is located in the vicinity of the first order natural frequency (f1). When the excitation frequency of rolling and pitching is located between 0.98f1 and 1.113f1, roof-bursting phenomenon of liquid appeared, and the maximum impact pressure is at 1.09f1. When the external excitation frequency is at 1.113f1, the number of sloshing shocks decreases sharply. Furthermore, the space distribution of the impact pressure on the left bulkhead and the top bulkhead was analyzed. It is concluded that with low liquid filling, the impact load is greater near the free surface and the top of tank, and the impact position of the side bulkhead increases with the increasing of the frequency near the resonant frequency.

        The Effect of miR-361-3p Targeting TRAF6 on Apoptosis of Multiple Myeloma Cells

        microRNA-361-3p (miR-361-3p) is involved in the carcinogenesis of oral cancer and pancreatic catheter adenocarcinoma, and has anti-carcinogenic effects on non-small cell lung cancer (NSCLC). However, its effect on multiple myeloma (MM) is less reported. Here, we found that upregulating the expression of miR-361-3p inhibited MM cell viability and promoted MM apoptosis. We measured expressions of tumor necrosis factor receptor-associated factor 6 (TRAF6) and miR-361-3p in MM cells and detected the viability, colony formation rate, and apoptosis of MM cells. In addition, we measured expressions of apoptosis-related genes Bcl-2, Bax, and Cleaved caspase-3 (C caspase-3). The binding site between miR-361-3p and TRAF6 was predicted by TargetScan. Our results showed that miR-361-3p was low expressed in the plasma of MM patients and cell lines, while its overexpression inhibited viability and colony formation of MM cells and increased the cell apoptosis. Furthermore, TRAF6, which was predicted to be a target gene of miR-361-3p, was highexpressed in the plasma of patients and cell lines with MM. Rescue experiments demonstrated that the effect of TRAF6 on MM cells was opposite to that of miR-361-3p. Upregulation of miR-361-3p induced apoptosis and inhibited the proliferation of MM cells through targeting TRAF6, suggesting that miR-361-3p might be a potential target for MM therapy.

        Study on the Solid Phase Extraction of Hg(II)-SBDTR Chelate with C<sub>18</sub> Disks and Its Application to the Determination of Mercury in Tobacco and Tobacco Additive

        A sensitive, selective and rapid method has been developed for the determination of mercury based on the rapid reaction of mercury(II) with p-sulfobenzylidenethiorhodanine (SBDTR) and the solid phase extraction of the colored chelate with $C_{18}$ disks. In the presence of pH 3.5 sodium acetate-acetic acid buffer solution and Emulsifier-OP medium, SBDTR reacts with mercury(II) to form a red chelate of a molar ratio 1 : 2 (mercury to SBDTR). This chelate was prconcentrated by solid phase extraction with $C_{18}$ disks. An enrichment factor of 50 was achieved. The molar absorptivity of the chelate is $1.28{\times}10^5 L{\cdot}mol^{-1}{\cdot}cm^{-1}$ at 545 nm in measured solution. Beer's law is obeyed in the range of 0.01-3 ${\mu}$g/mL. The relative standard deviation for eleven replicates sample of 0.01 ${\mu}$g/mL is 1.65%. This method was applied to the determination of mercury in tobacco and tobacco additive with good results.

        Preparation and characterization of flaky FeSiAl composite magnetic powder core coated with MnZn ferrite

        The flattening of FeSiAl soft magnetic powder was achieved by ball milling process, and MnZn/FeSiAl composite magnetic powder core was prepared by press molding. The effect of different coating amount of MnZn ferrite on the soft magnetic properties of FeSiAl was studied. At the same time, the optimal stress-relieving annealing temperature of the composite magnetic powder core is revealed. The results showed that the addition of MnZn ferrite affected the magnetic properties such as saturation magnetization (Ms), initial permeability (μi) and power loss (Pcm) of FeSiAl soft magnetic. With the increase of MnZn ferrite addition content, the saturation magnetization of composites decreased gradually, and the magnetic permeability increased first and then decreased, and the loss decreased first and then increased. When the addition content of MnZn ferrite was 5%, the permeability reached the maximum, which was 28.1% higher than that of the pure FeSiAl magnetic powder core under the same conditions. At the same time, the loss was the lowest, which was 13.3% lower than the pure FeSiAl powder core under the same conditions. When the annealing temperature is around 650 °C, the magnetic powder core has the largest magnetic permeability and the lowest loss.

        Performance validation and application of a mixed force-displacement loading strategy for bi-directional hybrid simulation

        Hybrid simulation (HS) is a versatile tool for structural performance evaluation under dynamic loads. Although real structural responses are often multiple-directional owing to an eccentric mass/stiffness of the structure and/or excitations not along structural major axes, few HS in this field takes into account structural responses in multiple directions. Multi-directional loading is more challenging than uni-directional loading as there is a nonlinear transformation between actuator and specimen coordinate systems, increasing the difficulty of suppressing loading error. Moreover, redundant actuators may exist in multi-directional hybrid simulations of large-scale structures, which requires the loading strategy to contain ineffective loading of multiple actuators. To address these issues, lately a new strategy was conceived for accurate reproduction of desired displacements in bi-directional hybrid simulations (BHS), which is characterized in two features, i.e., iterative displacement command updating based on the Jacobian matrix considering nonlinear geometric relationships, and force-based control for compensating ineffective forces of redundant actuators. This paper performs performance validation and application of this new mixed loading strategy. In particular, virtual BHS considering linear and nonlinear specimen models, and the diversity of actuator properties were carried out. A validation test was implemented with a steel frame specimen. A real application of this strategy to BHS on a full-scale 2-story frame specimen was performed. Studies showed that this strategy exhibited excellent tracking performance for the measured displacements of the control point and remarkable compensation for ineffective forces of the redundant actuator. This strategy was demonstrated to be capable of accurately and effectively reproducing the desired displacements in large-scale BHS.

        Effect of Two-step Time-restricted Feeding on the Fattening Traits in Geese

        The present study was conducted to determine whether the two-step time-restricted feeding improves the fattening traits of one-step time-restricted feeding in geese. Thirty-six 8-wk-old geese were allotted into one of three groups. Group R1 (the 1-step restricted feeding group) was allowed access to feed for 2 h in the morning from 8 wk to 14 wk of age. Group R2 (the 2-step restricted feeding group) was treated as Group R1, but was additionally fed for 2 h in the afternoon from 12 wk to 14 wk of age. Group C (the control group) was fed ad libitum from 8 wk to 14 wk of age. Feed intake and body weight (BW) were recorded daily and weekly, respectively. At 14 wk of age, the blood samples were collected to determine the fasting plasma levels of glucose, triacylglycerols and uric acid before sacrifice. The results showed that daily feed intake (DFI) was lower, feed efficiency (FE) was higher in both Groups R1 and R2 than in Group C, and daily gain (DG) in Group R2 was higher than in Group R1 during the whole experimental period (p<0.05). Group R1 exhibited lower abdominal and visceral fat weights in carcass than did Group C (p<0.05), and Group R2 was in intermediate. The fasting plasma glucose levels in Group C were higher, and triacylglycerol levels in Group R1 were higher, compared with the other groups (p<0.05). It is concluded that time-restricted feeding in the fattening period not only increases FE but reduces DFI, and the additional meal during the late fattening period improves the DG without the expense of FE in geese.

      • Mortality Characteristic and Prediction of Nasopharyngeal Carcinoma in China from 1991 to 2013

        Background: To analyze the mortality distribution of nasopharyngeal carcinoma in China from 1991 to 2013, to predict the mortality in the ensuing five years, and to provide evidence for prevention and treatment of nasopharyngeal carcinoma. Materials and Methods: Mortality data for Nasopharyngeal Carcinoma in China from 1991 to 2013 were used to describe its epidemiological characteristics, such as the change of the standardized mortality rate, sex and age differences, urban-rural differences. Trend-surface analysis was used to study the geographical distribution of the mortality. Curve estimation, time series, gray modeling, and joinpoint regression were used to predict the mortality for the ensuing five years in the future. Results: In China, the standardized mortality rate of Nasopharyngeal Carcinoma increased with time from 1996, reaching the peak values of $1.45/10^5$ at the year of 2002, and decreased gradually afterwards. With males being 1.51 times higher than females, and the city had a higher rate than the rural during the past two decades. The mortality rate increased from age 40. Geographical analysis showed the mortality rate increased from middle to southern China. Conclusions: The standardized mortality rate of Nasopharyngeal Carcinoma is falling. The regional disease control for Nasopharyngeal Carcinoma should be focused on Guangdong province of China, and the key targets for prevention and treatment are rural men, especially after the age of 40. The mortality of Nasopharyngeal Carcinoma will decrease in the next five years.

      • Meta-analysis of the Association Between GSTM1 and GSTT1 Gene Polymorphisms and Cervical Cancer

        Aim: We conducted a meta-analysis to analyze the influence of GSTM1 and GSTT1 gene polymorphisms on cervical cancer risk, and explore gene-environment interactions. Methods: Identification of relevant studies was carried out through a search of Medline and the EMbase up to Oct. 2011. All case-control studies that investigated the association between GSTM1 and GSTT1 gene polymorphisms and risk of cervical cancer were included. The pooled odds ratio (OR) was used for analyses of results and the corresponding 95% confidence intervals (CI) were estimated. Results: A total of 21 case-control studies were included in the meta-analysis of GSTM1 (2,378 cases and 2,639 controls) and GSTT1 (1,229 cases and 1,223 controls) genotypes. The overall results showed that the GSTM1 null was related to an increased risk of cervical cancer (OR=1.50, 95% CI=1.21-1.85). Subgroup analysis were performed based on smoking and ethnicity. Our results showed that smokers with null GSTM1 genotype had a moderate increased risk of cervical cancer (OR=1.85, 95% CI=1.07-3.20). For the ethnicity stratification, moderate significantly increased risk of null GSTM1 genotype was found in Chinese (OR=2.12, 95% CI=1.43-3.15) and Indian populations (OR=2.07, 95% CI=1.49-2.88), but no increased risk was noted in others. Conclusion: This meta-analysis provided strong evidence that the GSTM1 genotype is associated with the development of cervical cancer, especially in smokers, and Chinese and Indian populations. However, no association was found for GSTT1 null genotype carriers.
