        A Fusion Protein of Derp2 Allergen and Flagellin Suppresses Experimental Allergic Asthma

        Purpose: The house dust mite (HDM) is one of the most important sources of indoor allergens and a significant cause of allergic rhinitis and allergic asthma. Our previous studies demonstrated that Vibrio vulnificus flagellin B (FlaB) plus allergen as a co-treatment mixture improved lung function and inhibited eosinophilic airway inflammation through the Toll-like receptor 5 signaling pathway in an ovalbumin (OVA)- or HDM-induced mouse asthma model. In the present study, we fused the major mite allergen Derp2 to FlaB and compared the therapeutic effects of the Derp2-FlaB fusion protein with those of a mixture of Derp2 and FlaB in a Derp2-induced mouse asthma model. Methods: BALB/c mice sensitized with Derp2 + HDM were treated with Derp2, a Derp2 plus FlaB (Derp2 + FlaB) mixture, or the Derp2-FlaB fusion protein 3 times at 1-week intervals. Seven days after the final treatment, the mice were challenged intranasally with Derp2, and airway responses and Derp2-specific immune responses were evaluated. Results: The Derp2-FlaB fusion protein was significantly more efficacious in reducing airway hyperresponsiveness, lung eosinophil infiltration, and Derp2-specific IgE than the Derp2 + FlaB mixture. Conclusions: The Derp2-FlaB fusion protein showed a strong anti-asthma immunomodulatory capacity, leading to the prevention of airway inflammatory responses in a murine disease model through the inhibition of Th2 responses. These findings suggest that the Derp2-FlaB fusion protein would be a promising vaccine candidate for HDM-mediated allergic asthma therapy.

        A Novel Method for Pavement Transverse Crack Detection Based on 2D Reconstruction of Vehicle Vibration Signal

        Pavement transverse cracks are associated with pavement performance. However, due to lowrecognition accuracy, the transverse cracking detection through vehicle’s vibration responsehas hardly been considered. This paper proposes a novel method for pavement transversecracking detection using reconstructed vehicle vibration signal. An accelerometer installed inthe vehicle collects the vibration response between the vehicle and the pavement transversecracks. A brand-new approach to data processing is suggested that transforms a onedimensionalvibration signal into a two-dimensional vibration image. The two-dimensionalvibration image generated can obtain different features effectively from the time domain. Based on Gray Level Concurrence Matrix (GLCM) algorithm, a characteristic index establishedfrom the vibration image’s contrast can distinguish transverse cracks from uncracked sections. Then its feasibility was investigated by 410 pavement sections, the accuracy of the index is93.84%. The transverse crack detection method proposed in this paper seems promising forprecisely identifying pavement transverse cracks.

      • Personalized Course Evolutionary Based on Genetic Algorithm

        The paper presents an evolution of personalized courses based on genetic algorithms (PCEGA). The genetic algorithm are successfully applied in the dynamic update process of the course during the whole learning process. Under this framework of this algorithm, the target user model updates dynamically, and the courses evolve during the process. It provides a good general purpose and scalable framework that addresses the personalized course generation in an online learning environment.

        Fluorination of dichlorodifluoromethane to synthesize tetrafluoromethane over Cr_2O_3–AlF_3 catalyst

        A series of Cr_2O_3–AlF_3 catalysts with different Cr_2O_3 contents were prepared by a deposition–precipitation method for the fluorination of dichlorodifluoromethane (CCl_2F_2) to synthesize tetrafluoromethane (CF4). The highest activity was obtained on the Cr_2O_3–AlF_3 catalyst containing 61.2 wt.% of Cr_2O_3, with the CF_4 yield of 94% at 400℃. X-ray diffraction (XRD) and Raman spectroscopy results indicated the presence of bulk Cr_2O_3 and a-AlF_3 phases in the catalysts. Ammonia temperature programmed desorption (NH3-TPD) and Fourier transformation infrared spectroscopy (FTIR) of pyridine chemisorption revealed strong Lewis acidity of the catalysts. The Cr species and Lewis acid in the Cr_2O_3–AlF_3 catalysts were the active sites of fluorination and dismutation reaction of CCl_2F_2, respectively. This indicated bi-function catalytic performance of Cr_2O_3–AlF_3 catalysts for fluorination and dismutation reaction.

      • Elimination of Overshoot and Oscillation in the Auxiliary Branch of a SiC Auxiliary Resonant Commutated Pole Inverter (ARCPI)

        This paper investigates the mechanism of the overshoot and oscillation in the auxiliary branch and its elimination in a silicon carbide (SiC) auxiliary resonant commutated pole inverter (ARCPI). A mathematical model of the auxiliary branch including the parasitic capacitance of SiC MOSFETs has been derived, which reveals the oscillation mechanism in the auxiliary branch. The model shows that this oscillation is an inherent characteristic of the SiC ARCPI. To eliminate the overshoot and oscillation, a SiC ARCPI with two clamping diodes inserted between the DC bus and the auxiliary branch has been presented. The proposed auxiliary branch oscillation model and the performance of the clamped ARCPI are validated on a 3-kW, 3-phase SiC ARCPI prototype. Even though the overshoot and oscillation in the auxiliary branch has an insignificant impact on the output voltage, experimental results show that without clamping, the overshoot is more than 150% and the oscillation frequency is higher than 4 MHz. This significantly increases the required voltage ratings of the devices in the auxiliary branch. Furthermore, the overshoot and oscillation frequency in the auxiliary branch increases with the DC bus voltage because the output capacitance of the SiC MOSFETs reduces with the voltage. This degrade the electromagnetic interference (EMI) performance of the SiC ARCPI. In comparison, the clamped SiC ARCPI can mitigate the overshoot and oscillation and improve the EMI performance of the auxiliary branch. Specifically, around 6 MHz the spectral amplitude can be reduced by about 17% by the clamped ARCPI.

        Sputum Transcriptomics Reveals FCN1+ Macrophage Activation in Mild Eosinophilic Asthma Compared to Non-Asthmatic Eosinophilic Bronchitis

        Purpose: Eosinophilic asthma (EA) and non-asthmatic eosinophilic bronchitis (EB) share similar eosinophilic airway inflammation. Unlike EA, EB did not present airway hyperresponsiveness or airflow obstruction. We aimed to compare the mechanism underlying the different manifestations between EA and EB via sputum transcriptomics analysis. Methods: Induced-sputum cells from newly physician-diagnosed EA, EB patients, and healthy controls (HCs) were collected for RNA sequencing. Results: Bulk RNA sequencing was performed using sputum cells from patients with EA (n = 18), EB (n = 15) and HCs (n = 28). Principal component analysis revealed similar gene expression patterns in EA and EB. The most differentially expressed genes in EB compared with HC were also shared by EA, including IL4, IL5 IL13, CLC, CPA3, and DNASE1L3. However, gene set enrichment analysis showed that the signatures regulating macrophage activation were enriched in EA compared to EB. Sputum cells were profiled using single-cell RNA sequencing. FABP4+ macrophages, SPP1+ macrophages, FCN1+ macrophages, dendritic cells, T cells, B cells, mast cells, and epithelial cells were identified based on gene expression profiling. Analysis of cell-cell communication revealed that interactions between FCN1+ macrophages and other cells were higher in EA than in EB. A wealth of transforming growth factor beta (TGF-β) and vascular endothelial growth factor (VEGF) interactions between FCN1+ macrophages and other cells have been shown in EA. The gene expression levels of EREG, TGFBI, and VEGFA in FCN1+ macrophages of EA were significantly higher than those of EB. Furthermore, signatures associated with the response to TGF-β, cellular response to VEGF stimulus and developmental cell growth were enriched in FCN1+ macrophages of EA compared to those of EB. Conclusions: FCN1+ macrophage activation associated with airway remodeling processes was upregulated in EA compared to that in EB, which may contribute to airway hyperresponsiveness and airflow obstruction.

        东北亚地区双重均衡格局的形成及出路

        本文从现实主义的角度出发,认为东北亚存在的双重均衡体系是构成东 北亚当前地区安全格局的基础。一是中美在东北亚地区形成了以实力分布为基础的 威胁均衡。二是在次区域朝鲜半岛,美韩同盟与朝鲜之间也形成一种威胁均衡。但 是,东北亚的这种平衡秩序是不稳定的,地区的任何不稳定因素或不理性行为都可 能诱发难以掌控的安全后果。本文认为,要避免这种危险,就必须要改变东北亚目 前以威慑为主的互动方式。通过和解,扭转敌我刻板印象,构建东北亚各国新的身 份及关系定位。

        Who Restrains Who? Sino-DPRK Strategic Interaction during the Second Nuclear Crisis

        This paper seeks to find an answer to the question of why China’s effort to curb North Korea’s nuclear development has failed to produce expected outcomes. Since the second North Korean nuclear crisis in 2002, China, as the most, if not sole, influential country to North Korea, has been attempting to restrain the North’s nuclear armament. Nevertheless, Beijing has always failed to sustain its pressure on Pyongyang long enough to restrain Pyongyang’s nuclear ambition; instead, it has often ended up with China’s appeasement policy. Witnessing such failure, some scholars assert that Beijing is able, but not willing, to curb Pyongyang’s nuclear armament, while others claim that Beijing’s leverage over Pyongyang is a lot more limited than widely thought. Yet, both schools of thought, focusing narrowly on China’s capability or willingness to restrain North Korea, has failed to depict a full picture. By contrast, this study, employing a game theory approach, views Sino-DPRK relations as a strategic interaction between restrainer and restrainee. It argues that North Korea’s tactical deception and military adventurism shuffle China’s political priorities on the Korean peninsula, thereby preventing China from pressuring North Korea.

      • Data-driven estimation of change-points with mean shift

        Asymptotically linear negative quadrant dependent (ALNQD) sequence is more general than negatively associated (NA) sequence and \rho∗-mixing sequence. Based on ALNQD errors, the CUSUM estimator of mean change-point is studied with common conditions. We obtain a limiting distribution of CUSUM estimator, which can be used to judge the existence of mean change-point. Meanwhile, some weak and strong convergence rates are established for the CUSUM estimator of change-point. In addition, a data-driven algorithm of multiple change-point detection is given, which performs better than Lasso algorithm. Last, multiple change-point detection simulations and real data analysis illustrate the accuracy of our results.
