        Transcriptome analysis of Dioscorea zingiberensis identifies genes involved in diosgenin biosynthesis

        Dioscorea zingiberensis is the main plant source of diosgenin, a precursor for the production of steroid hormones used in the pharmaceutical industry. The extraction process of diosgenin from D. zingiberensis can generate high-acid and high-strength wastewater on a large scale and can threaten the environment. Bioengineering microorganisms to produce diosgenin is an effective way to avoid pollution. However, little is known about the genes that are involved in the biosynthesis of diosgenin. We obtained 85,010 unigenes (average length of 1142 bases) from the D. zingiberensis transcriptome through RNAseq. A large number of unigenes (59,368; 69.83%) were annotated, and 2488 unigenes were assigned to 27 secondary- metabolite pathways. In our database, 66 unigenes encoding up to 40 key enzymes were found to be present in diosgenin biosynthesis pathways. In addition, we found 203 unigenes encoding CYP450 proteins and 47 unigenes encoding UGT proteins that may be involved in modifications of a downstream pathway. The expression patterns of key diosgenin biosynthesis genes were studied to identify the most important members of the enzyme family. These results add to the available genetic data of D. zingiberensis and lay the foundation for the further production of diosgenin using genetic engineering.

        Recently, all-solid-state batteries (ASSBs) have attracted increasing interest owing to their higher energy density and safety. As the core material of ASSBs, the characteristics of the solid electrolyte largely determine the performance of the battery. Thus far, a variety of inorganic solid electrolytes have been studied, including the NASICON-type, LISICON-type, perovskite-type, garnet-type, glassy solid electrolyte, and so on. The garnet Li<sub>7</sub>La<sub>3</sub>Zr<sub>2</sub>O<sub>12</sub> (LLZO) solid electrolyte is one of the most promising candidates because of its excellent comprehensively electrochemical performance. Both, experiments and theoretical calculations, show that cubic LLZO has high room-temperature ionic conductivity and good chemical stability while contacting with the lithium anode and most of the cathode materials. In this paper, the crystal structure, Li-ion transport mechanism, preparation method, and element doping of LLZO are introduced in detail based on the research progress in recent years. Then, the development prospects and challenges of LLZO as applied to ASSBs are discussed.

      • Numerical Investigation of a Liquid-Gas Ejector Used for Shipping Ballast Water Treatment

        <P>Shipping ballast water can have significant ecological and economic impacts on aquatic ecosystems. Currently, water ejectors are widely used in marine applications for ballast water treatment owing to their high suction capability and reliability. In this communication, an improved ballast treatment system employing a liquid-gas ejector is introduced to clear the ballast water to reduce environmental risks. Commonly, the liquid-gas ejector uses ballast water as the primary fluid and chemical ozone as the secondary fluid. In this study, high-pressure water and air, instead of ballast water and ozone, are considered through extensive numerical and experimental research. The ejector is particularly studied by a steady three-dimensional multiphase computational fluid dynamics (CFD) analysis with commercial software ANSYS-CFX 14.5. Different turbulence models (including standardk-ε, RNGk-ε, SST, andk-ω) with different grid size and bubble size are compared extensively and the experiments are carried out to validate the numerical design and optimization. This study concludes that the RNGk-εturbulence model is the most efficient and effective for the ballast water treatment system under consideration and simple change of nozzle shape can greatly improve the ejector performance under high back pressure conditions.</P>

        New Structure Design for 126kV HV Vacuum Circuit Breaker

        This paper analyzes the advantages of double breaks under series connection for 126kV HV vacuum circuit breaker. It makes efforts in the break designing to avoid hidden danger brought by voltage-sharing capacitor. Voltage is evenly distributed in the shape of ▽ at the basic break that asks for no voltage-sharing capacitors. Permanent magnetic actuator is used to ensure the synchronization of opening and closing of double breaks. Based on this layout, this paper designs a new actuator and motion system for vacuum circuit breaker optimized by virtual prototype software ADAMS and genetic algorithm. Laboratory tests have proved that the opening and closing speed of the breaker meets the speed standard and that the 126kV HV vacuum circuit breaker is feasible.

        Mathematical Model of Two-Degree-of-Freedom Direct Drive Induction Motor Considering Coupling Effect

        The two-degree-of-freedom direct drive induction motor, which is capable of linear, rotary and helical motion, has a wide application in special industry such as industrial robot arms. It is inevitable that the linear motion and rotary motion generate coupling effect on each other on account of the high integration. The analysis of this effect has great significance in the research of two-degreeof- freedom motors, which is also crucial to realize precision control of them. The coupling factor considering the coupling effect is proposed and addressed by 3D finite element method. Then the corrected mathematical model is presented by importing the coupling factor. The results from it are verified by 3D finite element model and prototype test, which validates the corrected mathematical model.
