Transcriptome analysis of the short-term photosynthetic sea slug Placida dendritica

The intimate physical interaction between food algae and sacoglossan sea slug is a pertinent system to test the theory that “you are what you eat.” Some sacoglossan mollusks ingest and maintain chloroplasts that they acquire from the algae for photosynthesis. The basis of photosynthesis maintenance in these sea slugs was often explained by extensive horizontal gene transfer (HGT) from the food algae to the animal nucleus. Two large-scale expressed sequence tags databases of the green alga Bryopsis plumosa and sea slug Placida dendritica were established using 454 pyrosequencing. Comparison of the transcriptomes showed no possible case of putative HGT, except an actin gene from P. dendritica, designated as PdActin04, which showed 98.9% identity in DNA sequence with the complementary gene from B. plumosa, BpActin03. Highly conserved homologues of this actin gene were found from related green algae, but not in other photosynthetic sea slugs. Phylogenetic analysis showed incongruence between the gene and known organismal phylogenies of the two species. Our data suggest that HGT is not the primary reason underlying the maintenance of short-term kleptoplastidy in Placida dendritica.

Biological response of cells to variable conditions should affect the expression level of certain genes. Quantification of these changes in target genes needs stable internal controls. Real-time quantitative polymerase chain reaction (PCR) has traditionally used reference or ‘housekeeping’ genes, that are considered to maintain equal expression in different conditions, to evaluate changes in target genes between samples and experimental conditions. Recent studies showed that some housekeeping genes may vary considerably in certain biological samples. This has not been evaluated in red algae. In order to identify the optimal internal controls for real-time PCR, we studied the expression of eleven commonly used housekeeping genes; elongation factor 1-alpha, glyceraldehyde-3-phosphate dehydrogenase, β-actin, polyubiquitin, 30S ribosomal gene, 60S ribosomal gene, beta-tubulin, alpha-tubulin, translation initiation factor, ubiquitin-conjugating enzyme, and isocitrate dehydrogenase in different life-history stages of Bostrychia moritziana. Our results suggest that glyceraldehyde-3-phosphate dehydrogenase (GAPDH) and 30S ribosomal gene, have the most stable gene expression levels between the different life history stages (male, female, carposporophyte, and tetrasporophyte), while the other genes are not satisfactory as internal controls. These results suggest that the combinations of GAPDH and 30S would be useful as internal controls to assess expression level changes in genes that may control different physiological processes in this organism or that may change in different life history stages. These results may also be useful in other red algal systems.

A various algorism has been studied to extract possibly every energy from a wind turbine in conjunction with the increase of concern about wind power system. In order to verify these control algorism, it is essential to make the most similar conditions to the real wind turbine's environment. Therefore, using separately excited DC motor a wind turbine the most similar to the real turbine is simulated. Tower shadow effect and Wind shear effect are considered as well as inertia emulation. For the control of Back-to-Back Converter Vector current control methods and space vector pulse width modulation are used and for reducing THD of grid current LCL filter is considered. This simulation results verified the energy produced by wind all flows into the utility under the consideration of the characteristics of a wind turbine. The result of this paper is expected to be used as a basic material for analyzing the characteristics of the wind turbine generator.

The two-degree-of-freedom direct drive induction motor, which is capable of linear, rotary and helical motion, has a wide application in special industry such as industrial robot arms. It is inevitable that the linear motion and rotary motion generate coupling effect on each other on account of the high integration. The analysis of this effect has great significance in the research of two-degree-of-freedom motors, which is also crucial to realize precision control of them. The coupling factor considering the coupling effect is proposed and addressed by 3D finite element method. Then the corrected mathematical model is presented by importing the coupling factor. The results from it are verified by 3D finite element model and prototype test, which validates the corrected mathematical model.
