        Genome-wide Association Studies for Osteoporosis: A 2013 Update

        In the past few years, the bone field has witnessed great advances in genome-wide as-sociation studies (GWASs) of osteoporosis, with a number of promising genes identified. In particular, meta-analysis of GWASs, aimed at increasing the power of studies by com-bining the results from different study populations, have led to the identification of nov-el associations that would not otherwise have been identified in individual GWASs. Re-cently, the first whole genome sequencing study for osteoporosis and fractures waspublished, reporting a novel rare nonsense mutation. This review summarizes the im-portant and representative findings published by December 2013. Comments are madeon the notable findings and representative studies for their potential influence and im-plications on our present understanding of the genetics of osteoporosis. Potential limi-tations of GWASs and their meta-analyses are evaluated, with an emphasis on under-standing the reasons for inconsistent results between different studies and clarificationof misinterpretation of GWAS meta-analysis results. Implications and challenges ofGWAS are also discussed, including the need for multi- and inter-disciplinary studies.

      • Roles of E-Cadherin (CDH1) Genetic Variations in Cancer Risk: a Meta-analysis

        E-Cadherin (CDH1) genetic variations may be involved in invasion and metastasis of various cancers by altering gene transcriptional activity of epithelial cells. However, published studies on the association of CDH1 gene polymorphisms and cancer risk remain contradictory, owing to differences in living habits and genetic backgrounds. To derive a more better and comprehensive conclusion, the present meta-analysis was performed including 57 eligible studies of the association between polymorphisms of CDH1 gene promoter -160 C>A, -347 G>GA and 3'-UTR +54 C>T and cancer risk. Results showed that these three polymorphisms of CDH1 were significantly associated with cancer risk. For -160 C>A polymorphism, -160A allele carriers (CA and CA+AA) had an increased risk of cancer compared with the homozygotes (CC), and the similar result was discovered for the -160A allele in the overall analyses. In the subgroup analyses, obvious elevated risk was found with -160A allele carriers (AA, CA, CA+AA and A allele) for prostate cancer, while a decreased colorectal cancer risk was shown with the AA genotype. For the -347 G>GA polymorphism, the GAGA genotype was associated with increased cancer risk in the overall analysis with homozygous and recessive models. In addition, results of subgroup analysis indicated that the elevated risks were observed in colorectal cancer and Asian descendants. For +54 C>T polymorphism, a decreased risk of cancer was found in heterozygous, dominant and allele models. Moreover, +54T allele carriers (CT, CT+TT genotype and T allele) showed a potential protective factor in gastric cancer and Asian descendants.

      • Homeownership Determinants of Young People in Post-socialist China and the Implications for Social Stratification

        This article aims to study the determinants of young Chinese people’s homeownership. More specifically, it aims to compare the ups and downs of ‘redistributive power’ and ‘market ability’, as well as the influence of ‘ascribed’ and ‘achieved’ social status, in the Chinese social stratification after more than three decades of market transition. Firstly, the paper discusses the issues which link the Chinese housing system and young people. Subsequently, logistic regression analysis was applied to investigate the relationship between explanatory variables and homeownership outcomes among young people. In relation to this, the paper distinguishes between independent homeowners (live independently), dependent homeowners (live with home owning parents), and non-home owners, in order to further explore the impact of home-owning parents. Results show that a redistributive power still persists in the Chinese housing allocation, mainly in the form of territorial affiliation (hukou registration). Ascribed social status, such as original hukou site and social status of parents, is still of influence on homeownership among young people. The paper ends with a discussion of the implications of the research findings for the social stratification.

      • Clinical Observation and Therapeutic Evaluation of Rh-endostatin Combined with DP Regimen in Treating Patients with Advanced Esophageal Cancer

        Objective: To observe the curative effects of rh-endostatin combined with DP regimen in treating patients with advanced esophageal cancer and analyze the correlation of CT perfusion (CTP) parameters and the expression of vascular endothelial growth factor (VEGF). Methods: Twenty patients with esophageal cancer confirmed pathologically were randomly divided into combined treatment (rh-endostatin+DP regimen) group and single chemotherapy group, 10 patients in each group, respectively. All patients were given conventional CT examination and CTP imaging for primary tumor. The level of VEGF, the size of tumor and CTP parameters (BF, BV, PS and MTT) before treatment and after 2 cycles of treatment were determined for the comparison and the correlation between CTP parameters and VEGF expression was analyzed. Results: the therapeutic effect of rh-endostatin+DP regimen group was superior to single chemotherapy group. VEGF level after treatment in rh-endostatin+DP regimen group was obviously lower than single chemotherapy group (P<0.01). The expression of VEGF had positive correlation with BF and BV but negative correlation with MTT. Compared with treatment before for rh-endostatin+DP regimen group, BF, BV and PS decreased while MTT increased after treatment (P<0.05). However, there were no significant differences between treatment before and after treatment in single chemotherapy (P>0.05). Conclusions: Rh-endostatin can down-regulate the expression of VEGF in esophageal cancer, change the state of hypertransfusion and high permeability of tumor vessels and had the better curative effect and slighter adverse reactions when combined with chemotherapy.

        Lateral-torsional Buckling Analysis of Cantilever Beam with Tip Lateral Elastic Brace under Uniform and Concentrated Load

        The studies on the lateral-torsional buckling of cantilever steel beam with tip lateral elastic brace are rarely reported. The total potential energy is first established for the lateral-torsional buckling of cantilever steel beam with tip lateral elastic brace under uniform and concentrated load. The modal trial function of the lateral displacement and torsional angle are expressed as trigonometric function combination with six terms. By introducing new dimensionless parameters, the analytical solution of the dimensionless buckling equation for the lateral-torsional buckling of cantilever steel beam with tip lateral elastic brace is obtained. With the help of 1stOpt software which is mathematical optimization analysis software, the non-dimensional critical moment formula of the lateral-torsional buckling of cantilever beam with tip lateral elastic brace is obtained. Then, the accuracy of the formula is verified by ADINA finite element software. The results show that the given critical moment formula is of high accuracy. It provides convenient and simple design method for practical engineering.

        In terms of an implication of this study, it was confirmed that hallyu celebrity-oriented imitative psychology, South Korean cosmetics brand image, PPL attitude and others have effects on purchase intention on South Korean cosmetic products and in particular, of the path coefficients, this PPL attitude variable was proved to be the most influential factor and after all, PPL strategies of South Korea’s dramas were verified as a marketing strategy that would work effectively on Chinese consumers. South Korea needs to keep creating cultures and dramas of good quality, and they also must come up with strategies to promote their creations to China.

        오프라인 매장에서의 온라인 채널 통합 운영방안에 관한 연구

        Due to recent proliferation of the mobile shopping channels, customers increasingly tend to purchase using online channel while experiencing physical products in offline shops. This phenomenon requires traditional offline retailers to consider integrating online channels. In this study, we propose strategic options for the traditional offline retailers regarding the online channel integration, and provide corresponding decision models to maximize the expected profits. We also investigate how the strategic options vary with the product characteristics, by categorizing the products based on inventory cost, demand uncertainty, and fitness to the online channels. By analyzing numerical examples we illustrate how the best online channel integration strategy should be differentiated depending on the product categories.

        Diverse nectar robbers on Alpinia roxburghii Sweet (Zingiberaceae)

        This study records for the first time three mammal species as nectar robbers on the ginger Alpinia roxburghii Sweet. We examined the behavior of nectar robbers and compared with earlier studies on a single plant species. We recorded seven species of nectar robbers: three squirrels, one bird, and three bees. Timing of robbing nectars were similar; however, robbing behavior differed among robbers. In particular, squirrels damaged the flower parts while robbing the nectar.

        Hsa-miRNA-143-3p Reverses Multidrug Resistance of Triple-Negative Breast Cancer by Inhibiting the Expression of Its Target Protein Cytokine-Induced Apoptosis Inhibitor 1 In Vivo

        Purpose: Multidrug resistance (MDR) remains a major obstacle in the treatment of triple-negative breast cancer (TNBC) with conventional chemotherapeutic agents. A previous study demonstrated that hsa-miRNA-143-3p plays a vital role in drug resistance of TNBC. Downregulation of hsa-miRNA-143-3p upregulated the expression of its target protein cytokine-induced apoptosis inhibitor 1 (CIAPIN1) in order to activate MDR, while upregulation of hsa-miRNA-143-3p effectively enhances the sensitivity of drug-resistant TNBC cells to chemotherapeutics. The present study aimed to further verify these findings in vivo. Methods: We established a hypodermic tumor nude mice model using paclitaxel- resistant TNBC cells. We expressed ectopic hsa-miRNA- 143-3p under the control of a breast cancer-specific human mammaglobin promoter that guided the efficient expression of exogenous hsa-miRNA-143-3p only in breast cancer cells. Thereafter, we overexpressed hsa-miRNA-143-3p in xenografts using a recombinant virus system and quantified the expression of hsa-miRNA-143-3p, CIAPIN1 protein, and proteins encoded by related functional genes by western blot. Results: We successfully completed the prospective exploration of the intravenous virus injection pattern from extensive expression to targeted expression. The overexpression of hsa-miRNA-143-3p significantly alleviated chemoresistance of TNBC by inhibiting viability. In addition, we observed that the expression of CIAPIN1 as a hsa-miRNA-143-3p target protein was remarkably decreased. Conclusion: We partly illustrated the mechanism underlying the hsa-miRNA-143-3p/CIAPIN1 drug resistance pathway. HsamiRNA- 143-3p as a tumor suppressive microRNA may be a novel target to effectively reverse MDR of TNBC in vivo.

        Identification of novel rheumatoid arthritis-associated MiRNA-204-5p from plasma exosomes

        Rheumatoid arthritis (RA) is an autoimmune disease characterized by infiltration of immune cells in the synovium. However, the crosstalk of immune cells and synovial fibroblasts is still largely unknown. Here, global miRNA screening in plasma exosomes was carried out with a custom microarray (RA patients vs. healthy controls = 9:9). A total of 14 exosomal miRNAs were abnormally expressed in the RA patients. Then, downregulated expression of exosomal miR-204-5p was confirmed in both the replication (RA patients vs. healthy controls = 30:30) and validation groups (RA patients vs. healthy controls = 56:60). Similar to the findings obtained in humans, a decreased abundance of exosomal miR-204-5p was observed in mice with collagen-induced arthritis (CIA). Furthermore, Spearman correlation analysis indicated that plasma exosomal miR-204-5p expression was inversely correlated with disease parameters of RA patients, such as rheumatoid factor, erythrocyte sedimentation rate, and C-reactive protein. In vitro, our data showed that human T lymphocytes released exosomes containing large amounts of miR-204-5p, which can be transferred into synovial fibroblasts, inhibiting cell proliferation. Overexpression of miR-204-5p in synovial fibroblasts suppressed synovial fibroblast activation by targeting genes related to cell proliferation and invasion. In vivo assays found that administration of lentiviruses expressing miR-204-5p markedly alleviated the disease progression of the mice with CIA. Collectively, this study identified a novel RA-associated plasma exosomal miRNA-204-5p that mediates the communication between immune cells and synovial fibroblasts and can be used as a potential biomarker for RA diagnosis and treatment.
