This study aimed to explore the potential genes and pathways associated with bupivacaine-induced apoptosis. Human neuroblastoma cell line SH-SY5Y was used in this study. The effect of bupivacaine on cell viability of SH-SY5Y was detected by Cell Counting Kit-8. Transcriptome sequencing was performed for SH-SY5Y cells that were treated and untreated with bupivacaine based on the HiSeq 4000 sequencing platform. The sequencing results were analyzed using bioinformatics methods, including differentially expressed genes (DEGs) identification, functional enrichment analysis, protein–protein interaction (PPI) network analysis and module analysis. The cell viability of SH-SY5Y cells decreased significantly after bupivacaine treatment (p < .01). Based on the HiSeq 4000 sequencing platform, we obtained a global overview of the transcriptome of SH-SY5Y treated with/ without bupivacaine. Bioinformatics analysis identified 335 up-regulated and 294 down-regulated DEGs in bupivacaine group. They were significantly enriched in cell cycle-associated functions and pathways and cAMP signaling pathway. In the PPI network, proliferating cell nuclear antigen (PCNA), v-Akt murine thymoma viral oncogene homolog 3 (AKT3), cyclin-dependent kinase inhibitor 1A (CDKN1A) and cell division cycle 6 (CDC6) had high topology scores. Module analysis obtained two sub-network modules (cluster 1 and cluster 2). PCNA, CDC6, CDKN1A and AKT3 may play important roles in bupivacaine-induced apoptosis. Additionally, bupivacaine may also induce apoptosis via pathways of cell cycle and cAMP signaling pathway.

To minimize leakage and maintain the efficiency of turbomachinery, brush seal can be installed with zero clearance or interference as the flexibility. This leads to contact between the rotor and bristle pack, and may cause self-excited vibration, even instability. In this study, to establishment a mathematical model of the rotor system, a seal force model with bristle interference is proposed and the nonlinear oilfilm force is applied in view of short bearing assumption. The influences of main factors containing rotor rotational speed, installing interval, disc mass, and disk eccentricity on the nonlinear characteristics of the rotor-bearing-brush seal system are conducted by adopting bifurcation diagram, spectrum cascade, axis orbit, and Poincaré map. The results indicate that the vibration amplitude of the system with bristle interference is a bit larger than that without interference. The system stability is enhanced with smaller disk mass at a higher rotational speed. The proposed model could provide valuable reference for the design of a rotor-bearing-seal system.

Background and objective: The area of mangroves is gradually decreasing globally, and mangroves are already one of the most threatened ecosystems. Despite net growth in the mangrove areas in China, the restoration potential of mangroves is still insufficient. This study proposed the Random forest model as an excellent data mining method to map the restoration potential based on the predicted probability of mangrove habitat suitability. Methods: We demonstrated the vital environmental variables influencing habitat suitability. The de-cisive advantages of RFM were parsimonious (variables selection), cost-effective (us-ing existing open-source data), accurate (training AUC was 0.89, testing AUC was 0.91), highly efficient (fast-training speed); and its results had high explanatory power. Here, we first mapped the conservation gaps using the RFM. Results: The results showed that temperature was the most important environmental factor influencing the habitat suit-ability of mangroves. The northern limit of suitable areas was around 24°44' N. The theoretical suitable habitat area for mangrove was 196,566.6 ha (the highly suitable area was 32,551.4 ha, the medium suitable area was 164,015.2 ha). The potential area for mangrove restoration was 176,264 ha (Guangdong with 104215.4 ha, Guangxi with 65957.5 ha). Conclusion: We proposed 24 sites with conservation gaps for mangrove forests restoration and nine potential sites as examples for the further restoration plan. We took one example site with high restoration potential for further explanation: how the key environmental factors influence the habitat suitability and how to use the infor-mation to guide the restoration strategies. RFM can be used as a data mining algo-rithm for the utmost use of the presence-only ecological data, objectively evaluating the suitability of species distribution, and providing scientifically technical data for species restoration planning.
Three degrees of freedom chatter stability prediction in the milling process

Chatter in the cutting process has a great influence on workpiece surface quality, machining efficiency, and service life of the machine tool. This work presents three degrees of freedom (DOF) dynamic model applied to tool chatter for thin-walled structures in milling. Combined with dynamic cutting force modelling in the radial, axial and tangential directions, the stability chart was solved by adopting an improved semi-discretization method in time domain. And the stability prediction model considering regenerative effect was validated by cutting tests with specific axial depth of cut and spindle speed. The cutting experimental results performed at different axial depth of cuts were in accordance with predicted stability lobe diagram (SLD). Moreover, the effects of cutting contact angle and radial immersion ratio on SLD were also analyzed. The study results show that the dynamic model could make a good prediction of chatter stability, and the established approach is high efficient in predicting the chatter stability lobe considering the actual situation.

To date, six viruses belonging to genera Alphabaculovirus, Cypovirus, Iflavirus, Iteravirus, Bidensovirus and Maculavirus were harbored in the silkworm (Bombyx mori) and in the cultured BmN cells. Diseased silkworms with unknown etiology often create economic losses to the farmers. In the present investigation, the abundance and diversity of viruses in the midgut of diseased silkworms with soft rot symptoms were assessed by meta transcriptomic sequencing in order to screen the potential pathogenic virus of the silkworm, B. mori. After removal of undefined reads, clean reads were assembled, and the obtained 76 viral unigenes were annotated to 43 viruses. Among these viruses, the B. mori cypovirus, Pieris rapae virus, Nodaviridae sp., B. mori bidensovirus, Pseudomonas virus pf16, Pseudomonas phage Skulduggery, Klebsiella phage KPN U2874, Equid gamma herpesvirus 2, fowlpox virus and Xestia c-nigrum granulovirus were dominant virus species. These metatran scriptomic sequencing results provided new clues for screening and identifying novel pathogenic virus of the silkworm.
