        Cloning and Expression of the Duck Leptin Gene and the Effect of Leptin on Food Intake and Fatty Deposition in Mice

        Leptin is the adipocyte-specific product of the obese gene and plays a major role in food intake and energy metabolism. Leptin research was mainly focused on mammalian species, but understanding of leptin and its function in poultry is very poor. In this study, the duck leptin gene was amplified using the reverse transcription-polymerase chain reaction (RT-PCR) from duck liver RNA. The cDNA fragment was inserted into the pET-28a expression vector, and the resulting plasmid was expressed in Escherichia coli BL21 (DE3). Experimental mice were given an intraperitoneal injection of 10 mg/kg leptin dissolved in phosphate buffered saline (PBS), while the control mice were injected with PBS. The effect of leptin on food intake, body weight and fatty deposition in mice was detected. Sequence analysis revealed that duck leptin had a length of 438 nucleotides which encoded a peptide with 146 amino acid residues. The sequence shares highly homology to other animals. The coding sequence of duck leptin was 84 and 86% identical to human and pig leptin nucleotides sequence. Highest identity was with the rat coding sequence (95%). The identity of the amino acid sequence was 84, 82 and 96% respectively compared to that of the human, pig and rat. Results of SDS-PAGE analysis indicated that a fusion protein was specifically expressed in E. coli BL21 (DE3). The purified product was found to be biologically active during tests. Continuous administration of recombinant duck leptin inhibited food intake. Despite the decrease of food intake, leptin significantly induced body weight and fatty deposition. These changes were accompanied by a significant down-secretion of plasma glucose, cholesterol, triglyceride and insulin levels in mice. The observations provide evidence for an inhibitory effect of leptin in the regulation of food intake and for a potential role of duck leptin in the regulation of lipogenesis.

        Pointing to the demand of large scale internet websites want to improve users’ loyalty and flows maintaining. A novel conception of analysis engine for massive user behavior is proposed in this paper, which combines static analysis with dynamic monitoring for user behavior. It apply some improved data mining model based on cloud computing to analysis Web log data and contextual information of the page acquired real-time. Meanwhile, using cloud database for efficient processing and storaging. The results show that this system could significantly improve the effect and efficiency of user behavior analysis.

        circTUBGCP5 promotes liver cancer progression and glycolysis by up‑regulating the expression of ACSL4

        A large number of researches have shown that circular RNA (circRNA) is new hope for the diagnosis or treatment of tumors, including liver cancer (LCa). However, it remains largely unclear the role of circRNA in the progression of LCa and its molecular mechanism. GSE164803 microarray dataset was applied to identify dysregulated circRNAs in LCa and noncancerous tissues. CircTUBGCP5 (hsa_circ_0034049) was selected for further research. Biological functions of circTUBGCP5 were investigated by EdU, colony formation, flow cytometry, glucose consumption and lactate production assay, and in vivo tumorigenesis. RNA pull-down assay and dual-luciferase reporter assay were used to investigate the interaction between circTUBGCP5, miR-144-3p, and ACSL4. We demonstrated that circTUBGCP5 was significantly up-regulated in LCa tissues and cells. CircTUBGCP5 promoted LCa cell proliferation, anti-apoptotic ability, glycolysis, and tumorigenesis at least partially by sponging miR-144-3p to regulate ACSL4 protein level. In conclusion, circTUBGCP5 is a forceful contributor to malignant behaviors and glycolysis of LCa via modulating the circTUBGCP5/miR-144-3p/ACSL4 axis, which has provided a target for the diagnosis and treatment of LCa patients.

      • Bayesian inference of clustering and multiple Gaussian graphical models selection

        We consider a Bayesian framework for clustering the high-dimensional data and learning sparse multiple graphical models simultaneously. Diferent from most pre-vious multiple graphs learning methods which assume that the cluster information is known in advance, we impose a multi-distribution prior for the cluster labels. Then a joint spike-and-slab graphical lasso prior is imposed for the precision matrices, which can induce a sparsity and homogeneity of the heterogeneous graphical models across all clusters adaptively. Additionally, by imposing a structural Markov random feld (MRF) prior, the proposed method can also cluster the network-linked data without the independence assumption of the samples. Then a fast Expectation Maxi-mization (EM) algorithm is utilized for the posterior inference. The proposed model can get a signifcant improvement both in clustering error and graphical selection precision. The simulations and real data analysis are shown to demonstrate the per-formance of our method.

        An Improved Hybrid Canopy-Fuzzy C-Means Clustering Algorithm Based on MapReduce Model

        The fuzzy c-means (FCM) is a frequently utilized algorithm at present. Yet, the clustering quality and convergence rate of FCM are determined by the initial cluster centers, and so an improved FCM algorithm based on canopy cluster concept to quickly analyze the dataset has been proposed. Taking advantage of the canopy algorithm for its rapid acquisition of cluster centers, this algorithm regards the cluster results of canopy as the input. In this way, the convergence rate of the FCM algorithm is accelerated. Meanwhile, the MapReduce scheme of the proposed FCM algorithm is designed in a cloud environment. Experimental results demonstrate the hybrid canopy-FCM clustering algorithm processed by MapReduce be endowed with better clustering quality and higher operation speed.

        Neutrality between a Vertically Integrated Cable Provider and an Over-the-Top Video Provider

        We are concerned with whether a vertically integratedbroadband and content provider can unreasonably advantage itselfover competing content providers, either by selling quality-ofservice(QoS) to content providers at unreasonably high prices, orby refusing to provide access to QoS to competing content. We addressthis question by modeling the competition between one suchvertically integrated provider and one over-the-top (OTT) contentprovider. The broadband provider decides whether to deploy QoS,and if so it also determines the QoS price if sold to either the OTTcontent provider or directly to users. We analytically determinewhen the broadband provider will sell QoS and when the OTT contentprovider or users will purchase QoS. We characterize the optimalQoS and video service prices. The Internet service provider(ISP)’s market share increases with the difference in the value ofthe two video services and decreases with the difference in the correspondingcosts. Numerical results illustrate the effect of QoSprice on content price, the variation of prices and profit with QoSprice, and the variation of prices and market shares with the benefitof QoS. The ISP may sell QoS to users at a lower price thanwhenQoS is sold to theOTT provider.
