        Dynamic analysis of thin-walled open section beam under moving vehicle by transfer matrix method

        Three dimensional coupled bending-torsion dynamic vibrations of thin-walled open section beam subjected to moving vehicle are investigated by transfer matrix method. Through adopting the idea of Newmark-${\beta}$ method, the partial differential equations of structural vibration can be transformed to the differential equations. Then, those differential equations are solved by transfer matrix method. An iterative scheme is proposed to deal with the coupled bending-torsion terms in the governing vibration equations. The accuracy of the presented method is verified through two numerical examples. Finally, with different eccentricities of vehicle, the torsional vibration of thin-walled open section beam and vertical and rolling vibration of truck body are investigated. It can be concluded from the numerical results that the torsional vibration of beam and rolling vibration of vehicle increase with the eccentricity of vehicle. Moreover, it can be observed that the torsional vibration of thin-walled open section beam may have a significant nonlinear influence on vertical vibration of truck body.

        Using low-coverage whole genome sequencing technique to analyze the chromosomal copy number alterations in the exfoliative cells of cervical cancer

        Objectives: We analyzed the chromosomal-arm-level copy number alterations (CNAs) in the cervical exfoliative cell and tissue samples by using the low-coverage whole genomic sequencing technique. Methods: In this study, we retrospectively collected 55 archived exfoliated cervical cell suspension samples and the corresponding formalin-fixed and paraffin-embedded tissue section samples including 27 invasive cervical cancer and 28 control cases. We also collected 19 samples of the cervical exfoliative cells randomly from women to verify the new algorithm model. We analyzed the CNAs in cervical exfoliated cell and tissue samples by using the low-coverage next generation of sequencing. Results: In the model-building study, multiple chromosomal-arm-level CNAs were detected in both cervical exfoliated cell and tissue samples of all cervical cancer cases. By analyzing the consistency of CNAs between exfoliated cells and cervical tissue samples, as well as the heterogeneity in individual patient, we also established a C-score algorithm model according to the chromosomal-arm-level changes of 1q, 2q, 3p, 7q. The C-score model was then validated by the pathological diagnosis of all 74 exfoliated cell samples (including 55 cases in model-building group and 19 cases in verification group). In our result, a cutoff value of C-score >6 showed 100% sensitivity and 100% specificity in the diagnosis of cervical cancer. Conclusion: In this study, we found that CNAs of cervical exfoliated cell samples could robustly distinguish invasive cervical cancer from cancer-free tissues. And we have also developed a C-score algorithm model to process the sequencing data in a more standardized and automated way.

        Clearance Measurement Equipment for Gas Lubricated Dynamic Pressure Bearing of Gyro Motor

        The clearance of gas lubricated dynamic pressure bearing of gyro motor refers specifically to small internal air gap between the rotor and stator. The clearance size is an important indicator of running performance of a motor. In this work, a dedicated clearance measurement equipment is developed. The clearance is converted to external micro displacement and measured by dual inductive probes with relative measurement principle. For this purpose, three main functional modules are designed together with corresponding electronic and pneumatic control systems. The clamp fixation module helps to support the measured bearing flexibly at the shaft end, which is conducive to protect the bearing and ensure the smoothness of force application. The force application module consists of a 3-D precision motion platform and a triaxial force sensor. It converts internal air gap to external micro displacement in cooperation with the clamp fixation module. The displacement measurement module is a 2-D precision motion platform carrying dual inductive probes. Based on theoretical analysis and practical experiments, the measurement accuracy is superior to 0.3 μm. Moreover, it realizes controllable and continuous force application, which is suitable for batch measurement.

        Ginsenoside Rh2 attenuates microglial activation against toxoplasmic encephalitis via TLR4/NF-kB signaling pathway

        Background: Ginsenoside Rh2 (GRh2) is a characterized component in red ginseng widely used in Korea and China. GRh2 exhibits a wide range of pharmacological activities, such as anti-inflammatory, antioxidant, and anticancer properties. However, its effects on Toxoplasma gondii (T. gondii) infection have not been clarified yet. Methods: The effect of GRh2 against T. gondii was assessed under in vitro and in vivo experiments. The BV2 cells were infected with tachyzoites of T. gondii RH strain, and the effects of GRh2 were evaluated by MTT assay, morphological observations, immunofluorescence staining, a trypan blue exclusion assay, reverse transcription PCR, and Western blot analyses. The in vivo experiment was conducted with BALB/c mice inoculated with lethal amounts of tachyzoites with or without GRh2 treatment. Results and conclusion: The GRh2 treatment significantly inhibited the proliferation of T. gondii under in vitro and in vivo studies. Furthermore, GRh2 blocked the activation of microglia and specifically decreased the release of inflammatory mediators in response to T. gondii infection through TLR4/NF-kB signaling pathway. In mice, GRh2 conferred modest protection from a lethal dose of T. gondii. After the treatment, the proliferation of tachyzoites in the peritoneal cavity of infected mice markedly decreased. Moreover, GRh2 also significantly decreased the T. gondii burden in mouse brain tissues. These findings indicate that GRh2 exhibits an antieT. gondii effect and inhibits the microglial activation through TLR4/NF-kB signaling pathway, providing the basic pharmacological basis for the development of new drugs to treat toxoplasmic encephalitis.

        Experimental and analytical study on continuous GFRP concrete decks with steel bars

        A hybrid bridge deck is proposed, which includes steel bars, concrete and glass-fiber-reinforced-polymer (GFRP) plates with channel sections. The steel bar in the negative moment region can increase the flexural stiffness, improve the ductility, and reduce the GFRP ratio. Three continuous decks with different steel bar ratios and a simply supported deck were fabricated and tested to study the mechanical performance. The failure mode, deflection, strain distribution, cracks and support reaction were tested and discussed. The steel bar improves the mechanical performance of continuous decks, and a theoretical method is proposed to predict the deformation and the shear capacity. The experimental results show that all specimens failed with shear failure in the positive moment region. The increase of steel bar ratio in the negative moment region can achieve an enhancement in the flexural stiffness and reduce the deflection without increasing GFRP. Moreover, the continuous deck can achieve a yield load, and the negative moment can be carried by GFRP plates after the steel bar yields. Finally, a nonlinear analytical method for the deflection calculation was proposed and verified, with considering the moment redistribution, non-cracked sections and nonlinearity of material. In addition, a simplified calculation method was proposed to predict the shear capacity of GFRP-concrete decks.

      • Plumbagin from Plumbago Zeylanica L Induces Apoptosis in Human Non-small Cell Lung Cancer Cell Lines through NF-κB Inactivation

        Objective: To detect effects of plumbagin on proliferation and apoptosis in non-small cell lung cancer cell lines, and investigate the underlying mechanisms. Materials and Methods: Human non-small cell lung cancer cell lines A549, H292 and H460 were treated with various concentrations of plumbagin. Cell proliferation rates was determined using both cell counting kit-8 (CCK-8) and clonogenic assays. Apoptosis was detected by annexin V/propidium iodide double-labeled flow cytometry and TUNEL assay. The levels of reactive oxygen species (ROS) were detected by flow cytometry. Activity of NF-${\kappa}B$ was examined by electrophoretic mobility shift assay (EMSA) and luciferase reporter assay. Western blotting was used to assess the expression of both NF-${\kappa}B$ regulated apoptotic-related gene and activation of p65 and $I{\kappa}B{\kappa}$. Results: Plumbagin dose-dependently inhibited proliferation of the lung cancer cells. The IC50 values of plumbagin in A549, H292, and H460 cells were 10.3 ${\mu}mol/L$, 7.3 ${\mu}mol/L$, and 6.1 ${\mu}mol/L$ for 12 hours, respectively. The compound concentration-dependently induced apoptosis of the three cell lines. Treatment with plumbagin increased the intracellular level of ROS, and inhibited the activation of NK-${\kappa}B$. In addition to inhibition of NF-${\kappa}B$/p65 nuclear translocation, the compound also suppressed the degradation of $I{\kappa}B{\kappa}$. ROS scavenger NAC highly reversed the effect of plumbagin on apoptosis and inactivation of NK-${\kappa}B$ in H460 cell line. Treatment with plumbagin also increased the activity of caspase-9 and caspase-3, downregulated the expression of Bcl-2, upregulated the expression of Bax, Bak, and CytC. Conclusions: Plumbagin inhibits cell growth and induces apoptosis in human lung cancer cells through an NF-${\kappa}B$-regulated mitochondrial-mediated pathway, involving activation of ROS.

        Effects of dietary valine:lysine ratio on the performance, amino acid composition of tissues and mRNA expression of genes involved in branched-chain amino acid metabolism of weaned piglets

        Objective: The goal of this study was to investigate the effects of dietary standard ileal digestible (SID) valine:lysine ratios on performance, intestinal morphology, amino acids of liver and muscle, plasma indices and mRNA expression of branched-chain amino acid (BCAA) metabolism enzymes. Methods: A total of 144 crossbred pigs (Duroc×Landrace×Large White) weaned at 28±4 days of age (8.79±0.02 kg body weight) were randomly allotted to 1 of 4 diets formulated to provide SID valine:lysine ratios of 50%, 60%, 70%, or 80%. Each diet was fed to 6 pens of pigs with 6 pigs per pen (3 gilts and 3 barrows) for 28 days. Results: Average daily gain increased quadratically (p<0.05), the villous height of the duodenum, jejunum and ileum increased linearly (p<0.05) as the SID valine:lysine ratio increased. The concentrations of plasma α-keto isovaleric and valine increased linearly (p<0.05), plasma aspartate, asparagine and cysteine decreased (p<0.05) as the SID valine:lysine ratio increased. An increase in SID lysine:valine levels increased mRNA expression levels of mitochondrial BCAA transaminase and branched-chain α-keto acid dehydrogenase in the longissimus dorsi muscle (p<0.05). Conclusion: Using a quadratic model, a SID valine:lysine ratio of 68% was shown to maximize the growth of weaned pigs which is slightly higher than the level recommended by the National Research Council [6].
