        쥐 해마 HT22 세포에서 글루타메이트 유도 산화 스트레스에 대한 Salacca wallichiana 추출물의 신경 보호 효과

        Glutamate is an excitatory neurotransmitter distributed in the central nervous system of mammals. However, high concentrations of glutamate are known to cause neurodegenerative diseases such as Alzheimer’s disease, Parkinson’s disease, and stroke by causing nerve cell death. In this study, the antioxidant activity and neuroprotective effect of subtropical natural products were analyzed. Among 11 subtropical plant extracts mainly tested, Sallacca wallichiana extract (SE) showed the greatest free radical scavenging activity. Then, we confirmed through WST-1 assay that SE protected HT22 cells against glutamate-induced cell death in a concentration-dependent manner. The protective effects of SE against glutamate-induced apoptosis in HT22 cells were also confirmed by flow cytometry analysis using Annexin V/PI double staining. We also confirmed using H2DCF-DA single staining that SE inhibits glutamate-induced intracellular reactive oxygen species. And we were confirmed through that SE inhibited glutamateinduced phosphorylation of Mitogen-activated Protein kinases. Consequently, our results propose that SE may contribute to the development of therapeutics to prevent neurodegenerative diseases.

      • Ethnomedicinal plants and traditional knowledge among three Chin indigenous groups in Natma Taung National Park (Myanmar)

        <P><B>Abstract</B></P> <P><B>Ethnopharmacological relevance</B></P> <P>This research describes the ethnomedicinal plants as used by three Chin indigenous groups inhabiting areas at Natma Taung National Park (NTNP) in bio-culturally diverse Myanmar. The aim of this study was (1) to identify wild medicinal species and evaluate their local importance in local peoples’ healthcare as well as in protected area conservation; and (2) to compare traditional medicinal plant knowledge among the informants.</P> <P><B>Materials and methods</B></P> <P>A total of 206 Müün, Ng’gah and Daai informants from 20 villages were interviewed using semi-structured questionnaires. Species use-reports were computed to determine plant local importance and the informant consensus factor. Descriptive and the inferential statistics Mann–Whitney <I>U</I> and Kruskal–Wallis tests were employed to evaluate and compare the informants' traditional medicinal knowledge.</P> <P><B>Results</B></P> <P>A total of 75 wild ethnomedicinal taxa in 40 plant families across 16 ICPC-based disease categories were recorded. Species which recorded the highest number of use-reports appeared to play an important role not only in informants’ primary healthcare and in park conservation, but also in local livelihood.</P> <P><B>Conclusion</B></P> <P>This study presents the diversity of ethnomedicinal plants and their local importance in Chin indigenous peoples’ healthcare.         Syzygium claviflorum 추출물의 항산화 활성 및 각질형성세포 분화유도 효과

        We validated the physiological activity of Syzygium claviflorum (Roxb.) Wall. ex A.M. Cowan & Cowan (S. claviflorum) extracts (leaves, stems, fruits, and flowers) as a cosmetic ingredient. Firstly, S. claviflorum extracts removed over 80% of free radicals at various concentrations in antioxidant experiments using the DPPH and ABTS assay. In cytotoxicity experiments using human epidermal keratinocytes, S. claviflorum extracts showed low cytotoxicity. In addition, S. claviflorum extracts significantly increased the expression of keratin (KRT)1, KRT2, KRT9, KRT10, which are differentiation markers of keratinocytes, as well as genes involved in the maintenance of skin barrier function, including involucrin (IVL), loricrin (LOR), filaggrin (FLG), and claudin1 (CLDN1). In particular, the expression of FLG protein, inhibited by interleukin (IL)-4/IL-13 in atopic dermatitis, was restored by S. claviflorum extracts in in vitro experiments. Therefore, S. claviflorum extracts with excellent antioxidant efficacy and skin barrier improvement function will be useful materials for the development of future atopic dermatitis treatments and cosmetics. 사이자이지움 클래비플로룸(Syzygium claviflorum (Roxb.) Wall. ex A.M. Cowan & Cowan, S. claviflorum)의 추출물 (잎, 줄기, 열매, 꽃)의 화장품 소재로써 활용되기 위한 생리활성 능력을 검증하였다. 첫번째로, S. claviflorum 추출물은 DPPH와 ABTS assay법을 이용한 항산화 실험에서 다양한 농도로 처리한결과, 약 80% 이상의 자유 라디칼을 제거하였다. 사람 피부 표피 각질형성세포(human epidermal keratinocytes) 를 이용한 세포독성 실험에서는 S. claviflorum 추출물은 낮은 세포독성을 보였다. 또한, S. claviflorum 추출물은 각질형성세포의 분화인자 (keratin (KRT)1, KRT2, KRT9, KRT10)와 피부장벽의 기능 유지에 중요한involucrin (IVL), loricrin (LOR), filaggrin (FLG)과 claudin1 (CLDN1) 유전자의 발현을 현저히 증가시켰다. 특히, in vitro 아토피 피부염 실험에서 interleukin (IL)-4/IL-13에 의해 억제된 FLG 단백질 발현이 S. claviflorum 추출물에 의해 회복되었다. 따라서, 뛰어난 항산화 효능과 피부장벽 개선 기능을 보유한 S. claviflorum 추출물은 향후 아토피 피부염 치료제 및 화장품 개발에 유용한 소재가 될 것이다.
