        Production and release of pigments by plant hairy root cultures

        Recently, plant hairy roots have become of interest because of their active proliferation in a phytohormonefree medium and their capacity to produce valuable metabolites accumulated at comparable levels of the original plant roots. The purpose of our work is to extend the biotechnological application of hairy root cultures through studies on the following subjects. 1) Induction and characterization of hairy roots: Some hairy root clones were induced from red beet (Beta vulgaris), madder (Rubia tinctorum) and horseradish (Armoraeia lapathifolia) by Agrobacterium rhizogenes infection. These hairy roots had high contents of pigments or enzymes. 2) Bioreactor configuration: A turbine-blade fermentor was developed as a bioreactor suitable for the submerged culture of hiary root with a highly branched morphology. In this fermentor, a stainless steel mesh separated the culture space from the agitation space so that the hairy root cells were not in direct contact with the impellers. 3) Extracellular production of pigments: In the cultures of red beet and madder hairy roots, significant amounts of pigments were released from the cells into medium when the cells were subjected to the treatment of O₂ starvation. Both hairy roots were capable of growing even after the treatment. 4) Morphological change of hairy root: By cultivating the horseradish hairy root in the presence of phytohormones, the cell morphology could be changed reversibly between dedifferentiated-cell aggregates and organized hairy root.

      • Cleistocalyx nervosum Extract Ameliorates Chemical-Induced Oxidative Stress in Early Stages of Rat Hepatocarcinogenesis

        Purpose: To study the effect of Cleistocalyx nervosum extract (CE) on diethylnitrosamine (DEN) and phenobarbital (PB) induced oxidative stress in early stages of rat hepatocarcinogenesis. Materials and Methods: Male Wistar rats were divided into 4 groups, with Group 1 as a negative control and Group 2 was a positive control receiving DEN injections once a week and PB in drinking water for 6 weeks. Two weeks before DEN initiation and PB treatment, Groups 3 and 4, were fed with 500 and 1000 mg/kg of CEs, respectively, for 8 weeks. Results: A number of GST-P-positive foci, preneoplastic lesions, in the liver were markedly increased in carcinogen administered rats, but was comparatively decreased in rats treated with 1000 mg/kg of CE. The CE reduced malondialdehyde in serum and in the livers of rats treated with DEN and PB. Moreover, CE significantly increased the activities of glutathione peroxidase and catalase in rat liver. Conclusions: CE appeared to exert its chemopreventive effects by modulating antioxidant status during DEN and PB induced early stages of hepatocarcinogenesis in rats.

      • Preventive Effects of Spirogyra neglecta and a Polysaccharide Extract against Dextran Sodium Sulfate Induced Colitis in Mice

        Ulcerative colitis (UC) results from colonic epithelial barrier defects and impaired mucosal immune responses. In this study, we aimed to investigate the modifying effects of a Spirogyra neglecta extract (SNE), a polysaccharide extract (PE) and a chloroform fraction (CF) on dextran sodium sulfate (DSS)-induced colitis in mice and to determine the mechanisms. To induce colitis, ICR mice received 3% DSS in their drinking water for 7 days. Seven days preceding the DSS treatment, oral administration of SNE, PE and CF at doses of 50, 25 and 0.25 mg/kg body weight (low dose), 200, 100 and 1 mg/kg body weight (high dose) and vehicle was started and continued for 14 days. Histologic findings showed that DSS-induced damage of colonic epithelial structure and inflammation was attenuated in mice pre-treated with SNE, PE and CF. Furthermore, SNE and PE significantly protected colonic epithelial cells from DSS-induced cell cycle arrest, while SNE, PE and CF significantly diminished apoptosis. Proteome analysis demonstrated that SNE and PE might ameliorate DSS-induced colitis by inducing antioxidant enzymes, restoring impaired mitochondria function, and regulating inflammatory cytokines, proliferation and apoptosis. These results suggest that SNE and PE could prevent DSS-induced colitis in ICR mice by protection against and/or aiding recovery from damage to the colonic epithelium, reducing ROS and maintaining normal mitochondrial function and apoptosis.

        Epic is one of the worldwide spread oral traditions. Current studies of epics mainly concern the origin and formation of epics influenced by studies of Homer Issue. That is to study the birth and development of epics based on its written text, taking Greek epic as a model. As to the decline and digression of epics, scholars insist Marx’s viewpoints, specifically, ‘some arts, for instance epics, are accepted by everyone, in this regard, when art production appears as art production, it would not be created same as the ancient classical format which has significant of time in world history’, and consider epics were created in the age of barbarism, and would decline naturally in civilized society. ‘Marx and Engels said Iliad, Odyssey were created in later stage of barbarous society or earlier stage of civilized society, they never said worldwide epics were created in that stage’. For example, Mongolian epics were still prevailing until early 20th century, and some new contents were added then. Some scholars consider epics as the product of ‘a people’s uncivilized stage’ or ‘undeveloped era of art’. In fact, ‘it does not exceed of hundred years since Mongol epics was recorded in scripts the first time. And some are still alive among the people, and some new epics are found till recent days. Mongols have changed and updated the epics with new awareness in various historical stages, instead of remembered the ancient version without any changes.’ While observing the research history of epics, we would find researchers have paid close attention to the origin, development and changes of epics, and paid little attention to the decline of epics. In the article, the author argues the study of decline of Jangar has theoretical and practical importance to understand the vanishing history of dead epics, to understand the necessity of cherishing and spreading existing oral tradition.

        Metabolic Flux Analysis of a Poly-b-hydroxybutyrate Producing Cyanobacterium, Synechococcus sp. MA19, Grown under Photoautotrophic Conditions

        To understand the utilization property of light energy, Synechococcus sp. MA19, a poly-b-hydroxybutyrate (PHB) producer, was cultivated at the different incident light intensities of 15.3, 50.0 and 78.2 W/m2 using media with and without phosphate. From the results of metabolic flux analysis, it was found that the cell yield based on ATP synthesis was estimated as 3.5×10-3 kg-biomass/mol-ATP in these cultures. Under the examined conditions, there were no significant differences in the efficiency of light energy conversion to chemical energies estimated as ATP synthesis and reducing potential (NADH + NADPH) formation whether the PHB synthesis took place or not. The energy converted from light to ATP was kept relatively high around the energy absorbed by the cells of 2.5-3.0 x 106 J h-1 kg-1, whereas the energy of reducing potential was hardly changed in the examined range of the energy absorbed by the cells.

        일본의 민간 고령자를 위한 주거

        Japan has been developing a system of private housing for the elderly since early on, because public buildings alone will not be able to cope with the explosive growth of the elderly population in the future. The core of these systems are fee-based homes for the elderly and serviced senior housing for the elderly. This study aims to summarize the changes in laws and regulations concerning fee-based homes for the elderly and serviced senior housing for the elderly over the last 20 years or so. In addition, the study focuses on the issues of lump-sum payments upon admission to fee-based homes for the elderly and the right to live in the house for a lifetime as important legal issues common to both systems, and attempted to sort out the background and current status of these issues.
